Different types of fetal nucleated cells can be found in maternal blood, providing the possibility of non-invasive prenatal diagnosis. For this purpose, we have studied fetal erythroblasts. We discovered that haemoglobin-containing cells treated with 2,3-bisphosphoglycerate (BPG) can be visualized by a peroxidase reaction, which at the same time visualizes an in situ hybridization (ISH) signal, specific for the X, Y or 21 chromosome. In order to prove that the BPG-positive cells were erythroid, an anti-glycophorin A (GPA) antiserum combined with a staphylococcal rosette technique was used. To enrich for erythroblasts, leukocytes were depleted from maternal blood by treatment with anti-CD45 monoclonal antibody and passage over an anti-mouse IgG-coated glass bead column. To evaluate the potential of the method for clinical use, we studied maternal blood samples from 18 women referred to us for prenatal diagnosis between 6 and 19 weeks of gestation. Erythroblasts were found in 13 out of 14 normal pregnancies. Erythroblasts with a Y-signal were found as early as 9 weeks of gestation, but at 6 weeks the Y-signal was seen in BPG-negative cells only. These cells showed an epithelioid morphology indicating that they were cytotrophoblasts. The BPG-ISH method provides a simple technique for identifying erythroblasts and simultaneously visualizing a desired probe.  相似文献

The organic carbon balance of a lake with high input of allochthonous organic carbon is modelled integrating physical, chemical and biological processes. The physical model captures the behaviour of real thermal stratification in the lake for different flow situations during the period 1993–1997. The dissolved organic carbon model is based on simulated trajectories of water parcels. By tracking parcels, account is kept of environmental factors such as temperature and radiation as well as DOC quality for each parcel. The DOC concentration shows seasonal variations primarily dependent on inflow. The organic matter degradation (bacterial- and photodegradation) in the lake amounts to 1.5–2.5 mg C l–1 yr–1, where photooxidation is responsible for approximately 10%. The estimated DIC production in the lake is large compared to sediment mineralisation and primary production. The main conclusion is that the model with the selected parameterisations of the degradation processes reasonably well describes the DOC dynamics in a forest lake.  相似文献

Fetal alcohol spectrum disorders (FASD) are a consequence of prenatal alcohol exposure (PAE). The etiology of the complex FASD phenotype with growth deficit, birth defects, and neurodevelopmental impairments is under extensive research. Both genetic and environmental factors contribute to the wide phenotype: chromosomal rearrangements, risk and protective alleles, environmental-induced epigenetic alterations as well as gene-environment interactions are all involved. Understanding the molecular mechanisms of PAE can provide tools for prevention or intervention of the alcohol-induced developmental disorders in the future. By revealing the alcohol-induced genetic and epigenetic alterations which associate with the variable FASD phenotypes, it is possible to identify biomarkers for the disorder. This would enable early diagnoses and personalized support for development of the affected child.  相似文献

Similarity among group members may serve as a defence against visually hunting predators that preferentially attack individuals who are phenotypically different from the group majority. The presence of such odd individuals in an otherwise homogeneous group may, however, increase the vulnerability of the other group members as well. Individuals might thus be expected to form uniform groups in order to decrease predation risk, not only in trying to avoid being odd in a group, but also when attempting to avoid being accompanied by odd individuals. This hypothesis was tested with small and large three-spined sticklebacks, Gasterosteus aculeatus. Focal fish were offered the choice between a shoal consisting of conspecifics that were all similar in body length to the focal fish (matching shoal) and a shoal in which one or a few individuals differed in body length from the focal fish (non-matching shoal). In the control experiment, all individuals in the non-matching shoal differed in size from the focal fish. The control confirmed that individuals preferably joined the matching shoal when the alternative option was to be odd in another one. However, when the alternative for size-assortative shoaling was to belong to the majority in a mixed shoal, the shoal choice of individuals appeared on average to be random. Visual contact with a live pike, Esox lucius, did not affect the shoal choice pattern. Furthermore, despite the frequency-dependent nature of the oddity effect, varying the number of odd individuals in the non-matching shoal did not have a significant effect on individual's shoaling decisions. These results suggest that size-assortativeness in fish shoals is not a result of individuals avoiding being among the majority in a mixed group. Climate change vulnerability is disproportionately distributed between different population segments in society. This study qualitatively explores how key stakeholders in municipalities (i.e. planning and operational staff in municipalities and the vulnerable themselves) construct social vulnerability in relation to climate change with a specific focus on thermal stress (i.e. heat waves) and which adaptive responses they identify at different levels. The empirical material consists of five focus groups with actors in a large Swedish municipality where the “Vulnerability Factor Card Game” was used as stimulus material to create 10 fictional individuals. The results show that there is a substantial amount of local knowledge about vulnerability drivers and inter-relations between social factors and vulnerability. Local decision-makers also defined a wide range of possible adaptation measures at different municipal levels. Our study clearly indicates that contextualised knowledge, which could complement the quantitative approaches in research, is abundant among municipal planners, staff employed at municipal operations such as health care, and among the vulnerable themselves. This knowledge remains untapped by research to a great extent and only seems to have an insignificant influence on policy-making. In particular, how impacts vary between different social and demographic groups and how adaptation strategies that target the most vulnerable could be defined are of great interest. The present study clearly indicates that social hierarchy may produce increased inequality in the specific context of climate change, vulnerability and adaptive responses at different levels.  相似文献

This paper examines the science-practice interface in the process of adapting to climate change in society. This paper analyses science-based stakeholder dialogues with climate scientists, municipal officers and private individual forest owners in Sweden, and examines how local experts both share scientific knowledge and experience and integrate it into their work strategies and practices. The results demonstrate how local experts jointly conceptualise climate adaptation, how scientific knowledge is domesticated among local experts in dialogue with scientific experts, the emergence of anchoring devices, and the boundary-spanning functions that are at work in the respective sectors.  相似文献

The relationship between plant diversity and productivity has largely been attributed to niche complementarity, assuming that plant species are complementary in their resource use. In this context, we conducted an 15N field study in three different grasslands, testing complementarity nitrogen (N) uptake patterns in terms of space, time, and chemical form as well as N strategies such as soil N use, symbiotic N fixation, or internal N recycling for different plant species. The relative contribution of different spatial, temporal, and chemical soil N pools to total soil N uptake of plants varied significantly among the investigated plant species, within and across functional groups. This suggests that plants occupy distinct niches with respect to their relative N uptake. However, when the absolute N uptake from the different soil N pools was analyzed, no spatial, temporal, or chemical variability was detected, but plants, and in particular functional groups, differed significantly with respect to their total soil N uptake irrespective of treatment. Consequently, our data suggest that absolute N exploitation on the ecosystem level is determined by species or functional group identity and thus by community composition rather than by complementary biodiversity effects. Across functional groups, total N uptake from the soil was negatively correlated with leaf N concentrations, suggesting that these functional groups follow different N use strategies to meet their N demands. While our findings give no evidence for a biodiversity effect on the quantitative exploitation of different soil N pools, there is evidence for different and complementary N strategies and thus a potentially beneficial effect of functional group diversity on ecosystem functioning.  相似文献

Vertical small-scale variation in phenoxy acid herbicide degradation across a landfill leachate plume fringe was studied using laboratory degradation experiments. Sediment cores (subdivided into 5 cm segments) were collected in the aquifer and the sediment and porewater were used for microcosm experiments (50 experiments) and for determination of solid organic carbon, solid-water partitioning coefficients, specific phenoxy acid degraders and porewater chemistry. Results from a multi-level sampler installed next to the cores provided information on the plume position and oxygen concentration in the groundwater. Oxygen concentration was controlled individually in each microcosm to mimic the conditions at their corresponding depths. A highly increased degradation potential existed at the narrow plume fringe (37.7 to 38.6 masl), governed by the presence of phenoxy acids and oxygen. This resulted in the proliferation of a microbial population of specific phenoxy acid degraders, which further enhanced the degradation potential for phenoxy acids at the fringe. The results illustrate the importance of fringe degradation processes in contaminant plumes. Furthermore, they highlight the relevance of using high-resolution sampling techniques as well as controlled microcosm experiments in the assessment of the natural attenuation capacity of contaminant plumes in groundwater.  相似文献
