The soil nematode Caenorhabditis elegans is frequently used in ecotoxicological studies due to its wide distribution in terrestrial habitats, its easy handling in the laboratory, and its sensitivity against different kinds of stress. Since its genome has been completely sequenced, more and more studies are investigating the functional relation of gene expression and phenotypic response. For these reasons C. elegans seems to be an attractive animal for the development of a new, genome based, ecotoxicological test system. In recent years, the DNA array technique has been established as a powerful tool to obtain distinct gene expression patterns in response to different experimental conditions. Using a C. elegans whole genome DNA microarray in this study, the effects of five different xenobiotics on the gene expression of the nematode were investigated. The exposure time for the following five applied compounds beta-NF (5 mg/l), Fla (0.5 mg/l), atrazine (25 mg/l), clofibrate (10 mg/l) and DES (0.5 mg/l) was 48+/-5 h. The analysis of the data showed a clear induction of 203 genes belonging to different families like the cytochromes P450, UDP-glucoronosyltransferases (UDPGT), glutathione S-transferases (GST), carboxylesterases, collagenes, C-type lectins and others. Under the applied conditions, fluoranthene was able to induce most of the induceable genes, followed by clofibrate, atrazine, beta-naphthoflavone and diethylstilbestrol. A decreased expression could be shown for 153 genes with atrazine having the strongest effect followed by fluoranthene, diethylstilbestrol, beta-naphthoflavone and clofibrate. For upregulated genes a change ranging from approximately 2.1- till 42.3-fold and for downregulated genes from approximately 2.1 till 6.6-fold of gene expression could be affected through the applied xenobiotics. The results confirm the applicability of the gene expression for the development of an ecotoxicological test system. Compared to classical tests the main advantages of this new approach will be the increased sensitivity and it's potential for a substance class specific effect determination as well as the large numbers of genes that can be screened rapidly at the same time and the selection of well regulated marker genes to study more in detail.  相似文献

Coral reefs are highly dynamic and productive marine ecosystems, providing habitat and refuge for an enormous number of species including fish, invertebrates and algae. With increased anthropogenic pressures and global climate change, many coral reefs are rapidly declining. Currently, there is limited knowledge on condition and community assemblage composition of shallow fringing coral reefs along the south-eastern coast of Queensland, Australia. With increased demand to determine existence of coastal fringing reefs by National Regional Management groups, a rapid cost effective method to determine reef composition and condition was required. The aim of this study was to determine the benthic structure and extent of two small coastal fringing reefs (Hummock Hill Reef and Stringers Reef) along the Southern Great Barrier Reef. Reef substrate assessments were carried out using a rapid assessment technique and a Point Intercept Method (PIM). The data were analysed and classified using a Geographic Information System (GIS). Percent substrate cover was calculated using a visual basic image analysis program. The Point intercept method showed higher accuracy over the rapid assessment technique (up to 15–40% difference) and was thus deemed a more suitable classification tool for reefs with high structural complexity and heterogeneity. This study focused on piloting a rapid, cost effective Point Intercept Technique using random point count methodology to document coral benthic habitat and extent over a commonly used rapid assessment method as a tool for reef coastal management and conservation. The two techniques were compared and substrate classification success, limitations and errors were discussed.  相似文献

The Kings Cross/St. Pancras area has been a rail transport hub from Central London to the Midlands and North Eastern England since the middle of the nineteenth century and dust from the Kings Cross railway lands was even described by Charles Dickens.1 The Channel Tunnel high speed Rail Link (CTRL) is being constructed between 2001 and 2007 to connect continental Europe to Central London. The CTRL and associated development will help regenerate an area that is surrounded by deprived high density housing estates. London Borough of Camden has set up an extensive particulate monitoring programme to determine if there are any health impacts on local residents from emissions from the demolition and construction work. This article assesses the results from particulate measurements in 2003–2004 and compares this to baseline conditions before the redevelopment work began. The evidence suggests that although 2003 had higher than average pollution levels in the UK as a whole, even higher levels of particulates in the size range PM10–PM2.5 (defined as PMcoarse) were measured in Kings Cross. Due to the size of these particulates, they tend to be released from construction works rather than transport or secondary particulate sources. Concentrations in 2004 were lower than 2003, but this was primarily due to meteorological conditions. The paper also looks at the health of the local population and discusses whether these elevated levels are creating a problem and legally constitute a statutory nuisance. The innovative bioassay described here involves chlorophyll a fluorescence measurements of gametes from the macroalgae, Hormosira banksii, where gametes (eggs) were exposed to Diuron, Irgarol and Bromacil. Response was assessed as percent inhibition from control of effective quantum yield (DeltaF/Fm') of photosystem II, herein referred to as % PSII Inhibition. This was measured with the dual-channelled pulse amplitude modulated (PAM) fluorometer, ToxY-PAM. The fluorescence bioassay was run simultaneously with an established H. banksii germination bioassay to compare sensitivity, precision, and time-to-result. The fluorescence bioassay gave highly sensitive results evidenced by EC(50)s (% PSII Inhibition) for Diuron, Irgarol and Bromacil being three, four and three orders of magnitude (respectively) lower than EC50s generated from the germination bioassays. Precision of the fluorescence bioassay was demonstrated with low coefficient of variations (<30%) for all three toxicants. With regard to time, the fluorescence bioassay gave results within 6h, as opposed to more than 50h for the germination bioassay.  相似文献

This study determined the condition and reproductive output of a common estuarine toadfish, Tetractenos glaber, in two metal contaminated and two reference estuaries near Sydney, Australia. Female toadfish from metal contaminated estuaries were smaller and younger than in reference estuaries; however, it could not be resolved whether these differences were due to direct effects of metal contamination or differences in nutritional value of prey. Lipid content in liver and gonad tissues was inversely related with levels of As, Pb, Cd and Co in sediment. In contrast, protein content in liver, gonad and muscle tissues was positively related to sediment levels of Ni and Co. Increased levels of Pb in gonads were associated with decreased oocyte diameter and density. This suggests a reduction in egg size and fecundity, which ultimately may lead to a decline in female reproductive output. Changes in fish health and reproduction caused by chemical pollutants may alter fish population and community structure.  相似文献

Differences in stress tolerance and reproductive traits may drive the competitive hierarchy between non-indigenous and indigenous species and turn the former ones into successful invaders. In the northern Baltic Sea, the non-indigenous Gammarus tigrinus is a recent invader of littoral ecosystems and now occupies comparable ecological niches as the indigenous G. zaddachi. In laboratory experiments on specimens collected between June and August 2009 around Tvärminne in southern Finland (59°50′N/23°15′E), the tolerances towards heat stress and hypoxia were determined for the two species using lethal time, LT50, as response variable. The brood size of the two species was also studied and some observations were made on maturation of juveniles. Gammarus tigrinus was more resistant to hypoxia and survived at higher temperatures than G. zaddachi. Brood size was also greater in G. tigrinus than in G. zaddachi and G. tigrinus matured at a smaller size and earlier than G. zaddachi. Hence, there are clear competitive advantages for the non-indigenous G. tigrinus compared to the indigenous G. zaddachi, and these may be further strengthened through ongoing environmental changes related to increased eutrophication and a warming climate in the Baltic Sea region.  相似文献

Natural and synthetic steroidal hormones can be carried to agricultural soil through fertilization with municipal biosolids, livestock manure, or poultry manure. The persistence and pathways of dissipation of [4-(14)C]-testosterone and of [4-(14)C]-17beta-estradiol in organic-amended soils were investigated using laboratory microcosms. Testosterone dissipation was investigated over a range of amendment concentrations, temperatures, and soil types. Under all conditions the parent compound and transformation products were dissipated within a few days. Addition of swine manure slurry to soil hastened the transformation of testosterone and 17beta-estradiol to the corresponding less hormonally active ketones, 4-androstene-3,17-dione and estrone. Two other testosterone transformation products, 5alpha-androstan-3,17-dione and 1,4-androstadiene-3,17-dione, were also detected. Experiments with sterilized soil and sterilized swine manure slurry suggested that the transformation of (14)C-labeled hormonal parent compounds was mainly caused by microorganisms in manure slurry, while mineralization of the hormones to (14)CO(2) required viable soil microorganisms. Organic amendments transiently inhibited the mineralization of [4-(14)C]-testosterone, perhaps by inhibiting soil microorganisms, or by enhancing sorption and reducing the bioavailability of testosterone or transformation products. Overall, organic amendments influenced the pathways and kinetics of testosterone and estradiol dissipation, but did not increase their persistence.  相似文献

A new statistical testing approach using a weighted logrank statistic is developed for rodent tumorigenicity assays that have a single terminal sacrifice but not cause-of-death data. Instead of using cause-of-death assignment by pathologists, the number of fatal tumors is estimated by a constrained nonparametric maximum likelihood estimation method. For data lacking cause-of-death information, the Peto test is modified with estimated numbers of fatal tumors and a Fleming–Harrington-type weight, which is based on an estimated tumor survival function. A bootstrap resampling method is used to estimate the weight function. The proposed testing method with the weight adjustment appears to improve the performance in various situations of single-sacrifice animal experiments. A Monte Carlo simulation study for the proposed test is conducted to assess size and power of the test. This testing approach is illustrated using a real data set.  相似文献

Summary Garrett Hardin has written extensively about the biological, ecological and social impacts of various approaches to allocating natural resources. His work has been based largely upon the theory of the tragedy of the commons, population genetics and the application of biological principles to analogous social processes. He points out that policies that are morally appealing, because of their short-term benefits, may lead to disaster in the years ahead. His recommendations have stimulated considerable controversy and have been the object of moral criticism. They should not, however, be dismissed without careful consideration. Ralph H. Lutts is director of a natural history museum and an environmental education center. He is also adjunct assistant professor of environmental studies in the School of Natural Science of Hampshire College, Amherst, Massachusetts.
