In the majority of congenital peroxisomal disorders, β-oxidation of very long chain fatty acids is deficient. We have investigated the appearance and localization of the three peroxisomal β-oxidation enzymes in normal fetal liver (fertilization age between 5 and 18 weeks) with protein A- gold immunocytochemistry and silver enhancement for light microscopic visualization. With specificity-tested polyclonal antibodies, acyl-CoA-oxidase, bifunctiooal enzyme, and 3-oxoacyl-CoA thiolase were localized in the peroxisomes of the parenchymal cells, which appear as brown or black granules. In the youngest specimen, no immunopositive reaction was obtained. A weak reaction with anti-thiolase was obtained at the age of 6–7 weeks. At a fertilization age of 8 weeks, peroxisomes could be distinctly visualized after immunostaining for all three enzymes. From a staining series with anti-thiolase on simultaneously treated slides, it appears that the amount of antigen per peroxisome and the organelle size increase between the seventh and eighteenth weeks. These data should enable a more specific diagnosis in fetal liver biopsies from pregnancies at risk and after termination of pregnancy.  相似文献

Scavenging activities of melatonin, which is a pineal secretory product and functions in circadian biology, and its related compounds against reactive oxygen species such as superoxide anion radical, hydrogen peroxide, hydroxyl radical and singlet oxygen as well as organic peroxide radical (t-BuOO”) were evaluated chemically by using electron spin resonance-spin trap and chemiluminescence methods. Antioxidative activity of the compounds was estimated by IC50 value (µM), 50% inhibiting concentration of a compound against reactive oxygen species formed in each system, and the second-order rate constants (k2) for the reactions of the compounds and superoxide anion radical or hydroxyl radical. Because melatonin has exhibited the highest scavenging activity against t-BuOO”, the biochemical anti-lipid peroxide radical scavenging activities of melatonin were examined. We found that melatonin exhibits higher anti-lipid peroxidative activity in the rat brain microsomes than in the rat liver microsomal and liposomal systems, suggesting that melatonin may function as a treatment for reactive oxygen species-related diseases of the brain.  相似文献

Concentrations of polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDFs) and coplanar biphenyls (Co-PCBs) were determined in Japanese human adipose tissues. Temporal trends were assessed by comparing data collected during 1970-1971, 1994-1996 and 2000. Mean TEQ levels of PCDD/Fs in human adipose tissue showed a significant decrease from 31.6 +/- 9.2 pg-TEQ/g fat wt in 1970-1971 to 11.9 +/- 7.4 pg-TEQ/g fat wt in 2000, and coplanar PCBs decreased from 35.4 +/- 21.9 pg-TEQ/g fat wt in 1970-1971 to 15.3 +/- 8.2 pg-TEQ/g fat wt in 2000. However, some PCB congener concentrations did not change during this time. A comparison of data using the latest PCB-TEQ showed no significant changes. Therefore, research on source characterization and monitoring of short-term temporal trends on these compounds should be carried out continuously.  相似文献

The diel vertical distribution patterns of Japanese common squid, Todarodes pacificus, paralarvae were examined using a Multiple Opening Closing Net and Environmental Sensing System (MOCNESS) in the southwest Sea of Japan near the Oki Islands (Japan) during five late-autumn surveys in 1998–2002. A total of 1,511 paralarvae ranging in mantle length (ML) from 0.7 to 7.3 mm were collected at 63 of the 68 stations surveyed. Most (84%) were collected above 75 m depth and in the mixed layer. The vertical distribution patterns varied little between day and night. Hatchling-sized (<1.0 mm ML) paralarvae were abundant at 0–25 m depth, and paralarval ML increased with increasing sampling depth. Our results suggest that T. pacificus paralarvae do not exhibit large diel vertical migration patterns, but as they increase in size, paralarvae gradually descend in the water column and the variability in depth increases with ontogeny.  相似文献

The phylogenetic relationships among local populations of undescribed vestimentiferan species that belong to the genera Lamellibrachia and Escarpia and had been collected at six sites around Japan were analyzed on the basis of the partial (1023 bp) nucleotide sequences of the mitochondrial gene for cytochrome oxidase I (COI), using a pogonophoran and a polychaete as outgroups. The identical amino acid sequence was deduced from the nucleotide sequence obtained from each of the vestimentiferans analyzed. The strong similarity among deduced amino acid sequences of COI suggested a close relationship between vestimentiferans and pogonophorans. On the basis of the phylogenetic relationships, the analyzed vestimentiferan populations were classified tentatively as five species. The genetic differentiation of vestimentiferans was suggested to occur bathymetrically as well as being a consequence of horizontal segregation. Two of these tentatively identified species inhabit both a hydrothermal area and a cold seep area, as is the case for some species of bivalves that belong to the genera Bathymodiolus and Calyptogena. Farmland abandonment has been a significant issue in mountain regions. Thus, many studies have attempted to uncover its driving factors. Most existing studies take a large- or meso-scale view of mountain regions, and there are a few studies that provide community-level analysis. Therefore, it is unclear how ecological and sociopolitical factors are actually combined to influence farmland abandonment at a community level. Thus, we conducted a field survey in the mountain village of Ladakh, northern India, beginning in 2009 and tried to understand the farmland abandonment occurrence process from the viewpoint of traditional altitudinal models of land use. Using data collected during the survey, a relationship between altitude and farmland abandonment was established. However, this relationship was complex, combining traditional forms of land use system in the village. The altitudinal model of land use was very common in mountain regions; thus, discussions of current land use issues should be based on an understanding of such traditional altitudinal systems.  相似文献

We prepared a carbonaceous sorbent for Cr(VI) from the culm of Sasa kurilensis by dehydration with concentrated H2SO4. The removal of Cr(VI) by the sorbent was highly solution pH dependent and mainly governed by physicochemical sorption. The equilibrium data fit well in the Langmuir isotherm model and indicate the endothermic nature of the Cr(VI) sorption. The desorption experiments suggest that the Cr(VI) sorption is generally irreversible, owing to strong interaction of HCrO4 with the active sites of the sorbent.  相似文献

This paper proposes the construction of steel pipe sheet pile (SPSP) cutoff walls for promoting remediation of water-soluble toxic substances and containment of water-soluble toxic substances at landfill sites in order to maintain and ensure the environmental safety of waste landfill sites over time. It investigates environmental safety at coastal waste landfill sites by applying water cut-off and remediation promotion techniques to the joint sections of SPSP water cut-off walls that provide shore protection for waste landfills. Results from the research herein show that the construction of SPSP water cut-off walls with features such as the containment of water-soluble toxic substances and remediation promotion is possible by applying water cut-off and remediation techniques to H–H joints, which are structural joint components of SPSP walls. In addition, they show that the performance of remediation in H–H joints can be controlled by adjusting the water cut-off efficiency of the H–H joint flange.  相似文献

We characterized the ability of the cell free extract from polychlorinated dibenzo-p-dioxins degrading bacterium Geobacillus sp. UZO 3 to reduce even highly chlorinated dibenzo-p-dioxins such as octachlorodibenzo-p-dioxins in incineration fly ash. The degradation of 2,7-dichlorodibenzo-p-dioxin (2,7-DCDD) as a model dioxin catalyzed by the cell free extract from this strain implicates that the ether bonds of 2,7-DCDD molecule undergo reductive cleavage, since 4′,5-dichloro-2-hydroxydiphenyl ether and 4-chlorophenol were detected as intermediate products of 2,7-DCDD degradation.  相似文献

Present trends of urbanization are accompanied by increasing demographic and economic shrinkage of rural regions. In countries such as Japan, these rural regions trail behind metropolitan counterparts according to GDP, the conventional measure used to guide governmental policies. Yet, past research suggests that these regions may be undervalued. Further, the Inclusive Wealth Index (IWI), largely only used at the national level, may be able to capture aspects previously missed. As such, our study attempts to highlight the wealth of rural regions by comparing the inclusive wealth of Sado Island and Japan between 1990 and 2014. Minor methodological modifications were made according to data availability at the local level and to improve the accuracy of human capital estimations. Results captured the ongoing shrinkage of Sado and demonstrate the distinct potential of the IWI as a stock measure. Sado’s per capita wealth was about 10% lower than the national averages, but its natural capital was about threefold national averages. Supplementary estimations of the natural capital of fisheries and cultivated forests suggest that inclusion of additional factors in the evaluation would further increase the relative valuation of rural regions. We discuss implications of our estimations for wellbeing, and conclude with a critical appraisal of the IWI calculation towards policy implementation of the index.  相似文献
