Resolution of DNA linkage discrepancies through analysis of a VNTR locus in a family study of cystic fibrosis

First-trimester prenatal diagnosis of a fetus at 25 per cent risk for cystic fibrosis (CF) was performed by indirect linkage analysis of polymorphic markers using Southern blotting and polymerase chain reaction (PCR) amplification. The results revealed discrepancies in the allelic patterns between the father and the affected child, thereby complicating the prediction of fetal outcome. Analysis of a highly polymorphic VNTR locus within the human retinoblas-toma (RB) gene on chromosome 13 showed that the affected child and the fetus did not have the same biological father, and therefore the affected child could not be used to determine linkage of markers in the father of the fetus. The analysis of VNTR loci can be an effective method of resolving conflicting data during prenatal diagnosis of monogenic diseases.     

The importance of chorionic villus sampling after first trimester diagnosis of cystic hygroma

The prenatal diagnosis of The Turner Syndrome is described at a menstrual age of 12 weeks. Detection of cystic hygroma was followed by vaginal chorionic villous sampling (CVS) which revealed a 45,X karyotype. Early documentation of fetal karyotype in the presence of a cystic hygroma is essential for accurate diagnosis and genetic counselling.     

Status of air pollution in Beijing

A study has been made on elements organic constituents, TSP, SO2,NO2 of atmospheric pollutants in Beijing. 17 elements, and some PAHs, e. g. B(a)P, B(b, j, k)P, and B(g, h, i)P, in airborne particles by X-ray fluorescence spectroscopy and HPLC, GC/MS, have been determined respectively. It has been shown that the elements Pb, Zn, S and Cu were more enriched in fine particles and different valence states of sulfur at various sites. It was found that the concentrations of S6+ and S2-were more than 85% and less than 15% of the total sulfur respectively. Concentrations of major PAHs and sulfur-containing compounds increased in winter and in urban area. High values for Pb and Zn in city, Fe and Mn at industrial area and Cu, Al rural sites were obtained respectively. This implies the functions of different elemental sources of various sites. Thus, elements can be from distingushed anthropogenic and natural sources.The main contribution of SO2 was found of to have same seasonal variation as the anthropogenic el     

A new design of groundwater sampling device and its application

Compounds in the atmosphere contaminate samples of groundwater. An inexpensive and simple method for collecting groundwater samples is developed to prevent contamination when the background concentration of contaminants is high. This new design of groundwater sampling device involves a glass sampling bottle with a Teflon-lined valve at each end. A cleaned and dried sampling bottle was connected to a low flow-rate peristaltic pump with Teflon tubing and was filled with water. No headspace volume was remained in the sampling bottle. The sample bottle was then packed in a PVC bag to prevent the target component from infiltrating into the water sample through the valves. In this study, groundwater was sampled at six wells using both the conventional method and the improved method. The analysis of trichlorofluoromethane（ CFC-11 ） concentrations at these six wells indicates that all the groundwater samples obtained by the conventional sampling method were contaminated by CFC-11 from the atmosphere. The improved sampling method greatly eliminated the problems of contamination, preservation and quantitative analysis of natural water.     

How experienced individuals contribute to an improvement in collective performance in ants

Certain groups of organisms are capable of improving their collective performance with experience. In a recent study, we demonstrated that, over successive emigrations, colonies of the ant Temnothorax albipennis are able to improve their collective performance by reducing the time taken to complete an emigration (Langridge et al., Behav Ecol Sociobiol 56:523–529, 2004). In this paper, by recording the performance of individually marked workers during repeated emigrations, we were able to analyse some of the ways in which time gains are achieved. We found that: (1) those transporters that also transported in the preceding emigration began to transport earlier in the current emigration and, in the majority of emigrations, transported more items than those transporters that had not transported in the preceding emigration; (2) the time that elapsed before the first item was transported into the new nest reduced over successive emigrations, and this first item was, in the majority of emigrations, carried by a transporter that had also transported in the preceding emigration; (3) the number of adults that were transported reduced over successive emigrations. Our results strongly suggest that the behaviour of transporters that also transported in a preceding emigration may be modified as a result of their experience and that, consequently, their efforts in the next emigration make a major contribution to the improved performance of the colony as a whole.
