Gymnodinium nagasakiense is a noxious red tide dinoflagellate often associated with damage to fisheries in Japan. DNA synthesis and the cell cycle in this organism were investigated from 1989 to 1990 by determining relative DNA contents of individual cells using an epifluorescence microscopy-based microfluorometry system. The nuclei were stained with the DNA-specific fluorochrome 4-6-diamidino-2-phenylindole (DAPI). Because photosynthetic pigment interferes with the fluorescence from the DAPI-DNA complex, the pigment was eliminated by methanol treatment as a first step in quantitative microfluorometry. Nuclear DNA contents, cell size distribution, cell density, and frequency of paired cells were determined every 2h for 24h using cells grown on a 12h light:dark cycle. DNA synthesis and cell division were tightly phased to a particular period of the light:dark cycle. DNA synthesis (S phase) occurred from 10:00 to 22:00 hrs and was followed by cytokinesis. The presence of such a distinct S phase strongly suggests thatG. nagasakiense has a typical eukaryotic cell cycle. This type of cell cycle makes it possible to estimate speciesspecific in situ growth rate based on the diel pattern of DNA synthesis. 相似文献

Vertical distribution, life cycle, and developmental characteristics of the mesopelagic copepod Gaidius variabilis Brodsky in the Oyashio region were investigated by combining analyses of field copepodite populations with laboratory-rearing
data of egg hatching and naupliar development. Field samplings from five discrete depths between the surface and ≤2000 m were
made approximately every month for 1 year. Most populations of G. variabilis occurred between 600 and 1000 m depth. A modest degree of reversed diel vertical migration behavior and some stage-specific
depth-distribution patterns were noted. All copepodite stages were observed throughout the year, suggesting a year-round spawning
of G. variabilis. From a prominent abundance peak of Copepodite Stage 1 (C1) seen in June to August, together with development times of eggs
and nauplii obtained in laboratory-rearing experiments, the major spawning season was extrapolated to be April to June, the
phytoplankton bloom season. Tracing the peak abundance of each copepodite stage (distinguishing males and females for C4 to
C6), the generation times of males and females were deduced as 2 and 1 year, respectively. All between-stage increments in
terms of wet-, dry-, and ash-free dry weights were greatest in C3/C4, and least in C5/C6 for both males and females. The increments
in C3/C4 and C4/C5 were greater for males than for females, reflecting a longer stage duration of the males. These weights
did not increase in C5/C6 males, possibly because feeding ceased in C6 males. These results for G. variabilis are compared with those for some mesopelagic copepods previously reported from other regions.

Life cycle assessment (LCA), a quantitative method for evaluating the total environmental impact of a product, from the materials in its manufacture to its final disposal, is playing an increasingly important role in manufacturing. When the LCA method is applied to a product containing many kinds of electronic components, there is a need for life cycle inventory (LCI) data on the components. This paper provides an original calculation of the LCI data for each electronic components industry. These data show the amount of input energy and emissions into the atmosphere per yen of production yield. It is demonstrated that the magnitude of the LCI data for each industry is essentially equal to that of the other industries. Furthermore, we conclude that the LCI data for all electronic components are roughly equivalent, making it possible to calculate the LCI data of any electronic component by simply multiplying the LCI data for the industry by the price of the component. Furthermore, after comparing the materials production stages with the component manufacturing stage in the calculation, it became clear that for several component industries the materials production stage could not be omitted from the calculation. In this study, a new rubber surface pattern for a footwear sole was developed to prevent slip-related falls. This pattern shows a high static coefficient of friction (SCOF) and a high dynamic coefficient of friction (DCOF) when sliding against a liquid contaminated surface. A hybrid rubber block, in which a rubber block with a rough surface (Ra = 30.4 μm) was sandwiched between two rubber blocks with smooth surfaces (Ra = 0.98 μm), was prepared. The ratio of the rough surface area to the whole rubber block surface area r was 0%, 30%, 50%, 80%, and 100%. The coefficient of friction of the rubber blocks was measured when sliding against a stainless steel plate with Ra of 0.09 μm contaminated with a 90% aqueous solution of glycerol. While the SCOF increased with an increase of the rough surface area ratio r, the DCOF during steady-state sliding decreased with an increase of the rough surface area ratio r. The rough surface area ratio of 50% achieved a SCOF value around 0.5 or more and a DCOF value greater than 0.5. Furthermore, the difference in the value of the SCOF and DCOF was the smallest for the rubber block with r of 50%. The results indicated that the rubber block with r of 50% would be applicable to a footwear sole surface pattern to prevent slip and fall accidents on contaminated surfaces. 相似文献

We evaluated the effects of tributyltin (TBT), polychlorinated biphenyls (PCBs), and a mixture of TBT and PCBs on reproduction, gonadal histology, and sexual behavior in Japanese medaka (Oryzias latipes). We administered TBT (1 microg g(-1) body weight [b.w.] daily), PCBs (1 microg g(-1) b.w. daily), or both to medaka for 3 weeks. We assessed reproductive success during week 3 and the sexual behavior of male medaka after the exposure period. Fertilization success was significantly decreased in groups that received TBT. The profiles of fertility developed a bimodal distribution in the TBT and TBT+PCBs groups, and the numbers of males that performed "following" and "dancing" were significantly decreased in the TBT+PCBs group. The frequency of "dancing" also decreased after treatment with TBT, whereas fish that received PCBs only showed no difference in sexual behavior. Testes that had oocytes showed normal spermatogenesis, even though two of the five males treated with TBT+PCBs had some indication of testis-ova, which did not occur in any other group. Vertical distribution and population structure of Eucalanus bungii were investigated at site H in the Oyashio region (western subarctic Pacific) from September 1996 through October 1997 to evaluate the species lifecycle pattern and associated ontogenetic vertical migration. Additional temporary samplings were also made at several stations covering the entire subarctic Pacific, Okhotsk Sea and Japan Sea, as a basis for regional comparison of lifecycle features of this species. At site H, a marked phytoplankton bloom occurred from mid-March to June, and E. bungii spawned in April/May in the surface layer. Resulting nauplii and copepodite stage 1 (C1) formed a prominent abundance peak in early June. The C1 developed and reached C5 by August. The development of nauplii through C4 occurred in the surface layer. From August onwards, C5 and a small fraction of C3–C4 sank gradually deeper, and entered diapause to overwinter at >500 m depth. The C5 molted to C6 males and females in February and March, respectively. The C6 males and females mated at 250–500 m depth, and only mated C6 females ascended to the surface layer in April for spawning. Judging from the size of lipid droplets in the body, the C3–C5 specimens deposited lipids in the body through the phytoplankton bloom period, and the lipids were consumed gradually during overwintering. Taking account of sampling season, temporal changes in population structure, and vertical distribution, the data collected from the western subarctic Pacific and Okhotsk Sea are consistent with a 1-year lifecycle for the site H population, while the data from the central and eastern subarctic Pacific were consistent with a 2-year lifecycle. The populations from the southern and southeastern Japan Sea did not fit the features of either lifecycle scenario, and because of their very small population size it is suggested that they originated from the northern Japan Sea. Regional comparison of the prosome length of C6 females, including those in the Bering Sea, indicated significantly larger specimens from the Japan Sea and Okhotsk Sea, and smaller specimens in the eastern subarctic Pacific, as compared with those in the western subarctic Pacific (including site H) and Bering Sea. White-streaked grouper (Epinephelus ongus) is an important fishery target that forms fish spawning aggregations (FSAs) at specific spawning grounds. The aims of the present study were to investigate migration behavior and returning behavior in terms of FSA using acoustic telemetry. Among the 30 specimens tracked, three types of movement pattern were observed: type 1, high site fidelity at the release area before the spawning period, migration to the spawning ground during the spawning period and return to the release area just after the spawning period (17 specimens); type 2, high site fidelity at the release area, disappearance from the area during the spawning period but was not detected at the spawning ground and return to the release area (8 specimens); and type 3, high site fidelity at the release area was observed before, during and after the spawning period (5 specimens). For the 17 specimens that were categorized into type 1, the dates of departure from their release area and the dates of return to the release area of males were, respectively, significantly earlier and later than those of females. The duration between the departure date and the return date (“duration away”) of males was significantly longer than that of females. The duration away tended to be positively related to the condition factor. All 25 specimens that were categorized as type 1 and type 2 showed returning behaviors to the release area. 相似文献

This study describes the possible separation of chlorinated plastic films (PVC and PVDC) from other heavy plastic packaging waste (PPW) by selective twist formation and gravity separation. Twists formation was mechanically induced in chlorinated plastic films, whereas twist formation did not occur in PS and PET films. After twist formation, all the films had the apparent density of less than 1.0 g/cm3 and floated in water even though the true density was more than 1.0 g/cm3. However, the apparent density of the PS and the PET films increased with agitation to more than 1.0 g/cm3, whereas that of chlorinated plastic films was kept less than 1.0 g/cm3. The main reason would be the air being held inside the chlorinated plastic films which was difficult to be removed by agitation. Simple gravity separation after twist formation was applied for artificial film with 10 wt.% of the chlorinated films and real PPW films with 9 wt.% of the chlorinated films. About 76 wt.% of the artificial PPW films and 75 wt.% of real PPW films after the removal of PP and PE were recovered as settling fraction with 4.7 wt.% and 3.0 wt.% of chlorinated plastic films, respectively. These results indicate that simple gravity separation process after twist formation can be used to reduce the chlorinated plastic concentration from mixed heavy PPW films. 相似文献
