Numerous cases of hereditary diseases and disorders have been reported in wild animals bred in captivity, but little attention has been paid to the particular genetic management problems that arise when such defects occur. These problems include the obstacle of eliminating the deleterious allele(s) without contemporary loss of genetic variability. In this paper we use the statistical methods of pedigree analysis to address questions regarding a previously presumed hereditary form of blindness observed in a captive wolf population bred for conservation purposes in Scandinavian zoos. The most likely mode of inheritance coincides with an autosomal recessive allele with either a full penetrance or a reduced penetrance of 0.6 (depending on the reliability of studbook records). Using these two models of inheritance, we calculate the probability of carrying the blindness allele for each living animal. Analysis of the effect of removing high-probability carriers on founder allele survival and level of inbreeding demonstrates that the frequency of the deleterious allele can be significantly reduced without seriously affecting founder allele survival or current degree of inbreeding in the wolf population. 相似文献

Standardized tests were applied to aromatic and polar fractions of sediment extracts to determine whether polar constituents or oxidative degradation products contributed significantly to the toxicity of sediments oiled by the Exxon Valdez spill. Intertidal sediment and pore-water samples were collected in September 1990 from two heavily oiled sites and an unoiled site in Prince William Sound (PWS). Methylene chloride extracts from these samples were fractionated by liquid chromatography into aliphatic, aromatic and polar fractions, and the aromatic and polar fractions were tested for toxicity using the MicrotoxR test, bivalve larval mortality and development (Mytilus); several measures of genotoxicity in Mytilus, including SOS ChromotestR, anaphase aberrations and sister chromatid exchange; and survival, anaphase aberrations and teratogenicity in coho salmon (Onchorhynchus kisutch). MicrotoxR and SOS ChromotestR protocols were applied in a screening mode to all samples, whereas other tests were applied only to selected fractions from two sites. Samples from Bay of Isles (oiled) were consistently more toxic (usually only 2 to 5-fold) than the Mooselips Bay (unoiled) samples, which gave very low responses in all tests. for both sites, however, responses to polar and aromatic fractions were about the same in most tests, suggesting that while the overall toxicity of the oil was low in these samples, at least part of that toxicity was derived from polar constituents. Compared to the parent hydrocarbons, polar oxidation products partition preferentially into pore-water and are more rapidly diluted and dispersed in the water column. These results suggest that polar oxidation products of petroleum hydrocarbons pose little risk to marine organisms, except possibly for infauna continuously exposed to pore-water in heavily oiled sediments. Independent surveys showed that sediment toxicity in PWS declined during 1989-1991 to near background levels, in accord with previous understanding of oil weathering and toxicity. 相似文献

Canopy-forming macroalgae are key species on temperate rocky shores. However, there is a lack of understanding of how the relative balance of physical and biological factors controls the establishment and persistence of intertidal macroalgae. Here we present an integrated study of the relative importance of wave-induced forces and grazing for the recruitment and survival of the canopy-forming intertidal macroalgae Fucus vesiculosus and F. spiralis. A set of overtopped breakwaters provided a nearly unconfounded gradient in wave exposure between seaward and landward sides. A biomechanical analysis was performed based on empirical measurements of maximum drag forces in breaking waves, a model of long-term maximum wave height, and the breaking stress of Fucus spp. The estimated maximum flow speed (7-8 m/s) on the seaward side of the breakwaters was predicted to completely dislodge or prune Fucus spp. larger than approximately 10 cm, while dislodgment was highly unlikely on the landward side for all sizes. Experimental transplantation of Fucus spp. supported the biomechanical analysis but also suggested that mechanical abrasion may further limit survival in wave-exposed locations. Experimental removal of the limpet Patella vulgata, which was the principal grazer at this site, resulted in recruitment of Fucus spp. on the seaward side. We present a model of limpet grazing that indicates that limpet densities >5-20 individuals/m2 provide a proximate mechanism preventing establishment of Fucus spp., whereas wave action >2 m/s reduces persistence through dislodgment and battering. In a conceptual model we further propose that recruitment and survival of juvenile Fucus spp. are controlled indirectly by wave exposure through higher limpet densities at exposed locations. This model predicts that climate change, and in particular an increased frequency of storm events in the northeast Atlantic, will restrict fucoids to more sheltered locations. 相似文献

Counterintuitive density-dependent growth in a long-lived vertebrate after removal of nest predators
Examining the phenotypic and genetic underpinnings of life-history variation in long-lived organisms is central to the study of life-history evolution. Juvenile growth and survival are often density dependent in reptiles, and theory predicts the evolution of slow growth in response to low resources (resource-limiting hypothesis), such as under densely populated conditions. However, rapid growth is predicted when exceeding some critical body size reduces the risk of mortality (mortality hypothesis). Here we present results of paired, large-scale, five-year field experiments to identify causes of variation in individual growth and survival rates of an Australian turtle (Emydura macquarii) prior to maturity. To distinguish between these competing hypotheses, we reduced nest predators in two populations and retained a control population to create variation in juvenile density by altering recruitment levels. We also conducted a complementary split-clutch field-transplant experiment to explore the impact of incubation temperature (25 degrees or 30 degrees C), nest predator level (low or high), and clutch size on juvenile growth and survival. Juveniles in high-recruitment (predator removal) populations were not resource limited, growing more rapidly than young turtles in the control populations. Our experiments also revealed a remarkably long-term impact of the thermal conditions experienced during embryonic development on growth of turtles prior to maturity. Moreover, this thermal effect was manifested in turtles approaching maturity, rather than in turtles closer to hatching, and was dependent on population density in the post-hatching rearing environment. This apparent phenotypic plasticity in growth complements our observation of a strong, positive genetic correlation between individual body size in the experimental and control populations over the first five years of life (rG - +0.77). Thus, these Australian pleurodiran turtles have the impressive capacity to acclimate plastically to major demographic perturbations and enjoy the longer-term potential to evolve adaptively to maintain viability. 相似文献

Studies on the effect of genetically modified Bacillus thuringiensis (Bt) crops on true soil dwelling non-target arthropods are scarce. The objective of this study was to assess the influence of a 4-week exposure to two Bt maize varieties (Cry1Ab) Cascade and MEB307 on the collembolan Protaphorura armata. For comparison three non-Bt maize varieties, Rivaldo (isogenic to Cascade), Monumental (isogenic to MEB307) and DK242, and two control diets based on baker's yeast (uncontaminated and contaminated with Bt toxin Cry1Ab) were also tested. Due to a lower C:N ratio, individuals reared on yeast performed significantly better in all of the measured endpoints than those reared on maize. P. armata performed equally well when reared on two Bt and three non-Bt maize varieties. Although there were no negative effects of Bt maize in this experiment, we recommend future studies on Bt crops to focus on species interactions in long-term, multi-species experiments. 相似文献

Changes in the saturated uptake kinetics of the limiting nutrient were followed as Thalassiosira pseudonana (Clone 3 H) batch cultures entered ammonium, nitrate, silicate and phosphate starvation. Cultures starved of ammonium or phosphate developed very high specific uptake capacities over a 24 to 48 h starvation period, due to both decreases in cell quota and increases in uptake rates per cell. In particular, the cell phosphorus quota decreased ca. 8-fold during phosphate starvation and specific uptake rates exceeded 100 d-1. In contrast, cultures entering nitrate or silicate starvation underwent little or no further cell division, and the uptake capacity declined during starvation. After 24 to 48 h starvation, an induction requirement for uptake of nitrate or silicate was apparent. A study of gonadal state in 39 small specimens of Arctica islandica Linné collected in April 1976 from the Middle Atlantic shelf of North America suggests that sexual maturity is reached at a later age than has been reported for other shelf bivalves. Eight of the specimens had gonads in the undifferentiated state. On the basis of annual internal growth banding in the shells of these immature specimens, age ranged between 4 and 14 years. Average age was 9.38 years (standard deviation, s,= 3.54). Shell length ranged from 24 to 47 mm (average 38.85 mm, s=9.25). Differentiated gonads were found in specimens as young as 6 years. Age of maturity shows a wide range and may be dependent upon growth rate and locality. 相似文献

We estimated the broad-sense heritability of larval size in 20 full-sib families of the giant scallop, Placopecten magellanicus (Gmelin, 1791) grown in laboratory culture in August and September 1991. The goal was to compare scallops with other bivalves which have been shown to have significant heritabilities for larval growth. Secondly, we estimated the lipid content of occytes from female parents, since this is hypothesized to affect larval growth and survival. Finally, we estimated the among-family variation in mortality from 4 to 21 d as a test of possible genetic variation for viability among larval scallops. Significant genetic variation (h2=1.10 to 1.24) was estimated for larval shell length at 4, 14, and 21 d. There was a significant correlation (r=0.66) between larval size at 4 d and lipid content of oocytes, but only when two females with high levels of lipid oocyte–1 were excluded as outliers. There was no correlation between larval size at 14 and 21 d and lipid content of oocytes. Mortality among families from 4 to 21 d was high (69 to 97%), and was significantly different among families. These results indicate that there is significant heritability for larval growth which is largely independent of the lipid content of the oocytes. A high heritability for larval growth may indicate that this trait is only weakly correlated with fitness. 相似文献
