Evaluation of a respirometric test method to determine the heterotrophic yield coefficient of activated sludge bacteria 总被引:1,自引:0,他引:1
The heterotrophic yield coefficient of activated sludge bacteria is an important parameter in the field of wastewater degradation kinetics and the determination of degradation kinetics of defined chemicals. With the help of easily biodegradable organic compounds like glucose and acetate the respiration kinetics of degrading activated sludge bacteria was studied. The results of the respiration analysis were transformed to examine the degradation and respiration kinetics of municipal wastewaters at different food to microorganism ratios (F/M ratios). It was proved that the heterotrophic yield coefficient of aerobically degrading activated sludge bacteria could reliably be determined and that the heterotrophic yield coefficient was independent of the F/M ratio over a wide range. 相似文献

Inhibition of Na+/K+-ATPase from gill plasma membranes of the shore crab Carcinus maenas by cadmium was investigated and compared with inhibitory effects by known antagonists (ouabain and Ca2+). For comparative considerations the Cd2+-inhibition of the enzyme from dog kidney was also tested. Na+/K+-ATPase from dog kidney and from crab gill differed greatly in sensitivity against ouabain. The inhibition constant K
i of the dog enzyme amounted to 9.1 × 10−7 mol l−1, i.e. more than 300-fold smaller than the K
i of 2.9 × 10−4 mol l−1 determined for the crab enzyme. Ca2+ inhibited the activity of Na+/K+-ATPase from crab gill plasma membranes with a K
i of 4.3 × 10−4 mol l−1. The Na+/K+-ATPase from crab gill was inhibited by Cd2+ with a K
i of 9.1 × 10−5 mol l−1. Cd2+ inhibited the Na+/K+-ATPase from dog kidney with a K
i (6.4 × 10−5 mol l−1) comparable to that observed in the crab gill enzyme. Under experimental conditions Cd2+-inhibition of Na+/K+-ATPase was irreversible. Repeated washing, centrifugation and homogenization of the plasma membranes (four times) with Cd2+-free buffer did not restore any activity lost in the presence of 1 × 10−3 mol l−1 Cd2+. Since ouabain-insensitive (nonspecific) ATPases in the plasma membrane fraction of crab gills were inhibited by Cd2+ in the same way as Na+/K+-ATPase, the heavy metal is considered as an unspecific ATPase inhibitor. Comparing these results with literature data on
Cd2+-binding to electrophoretically separated proteins suggests that Na+/K+-ATPase is a Cd2+-binding enzyme. The results obtained on Na+/K+-ATPase were reflected by Cd2+-inhibition of the branchial ion-transport functions depending on this enzyme. The transepithelial short-circuit current of
isolated gill half lamellae, a direct measure of area-specific active ion uptake, and the transepithelial potential difference
of isolated, perfused whole gills, also indicative of active ion uptake, were inhibited by the heavy metal in a time- and
dose-dependent mode. Remarkably these inhibitions were also irreversible. These findings are ecologically and biomedically
significant: even when the actual environmental or tissue concentrations measured are low, biological microstructures such
as Na+/K+-ATPase may accumulate the heavy metal by tight binding over prolonged periods until the first inhibitory effects occur.

Temporal trends of five tetra- to hexabromodiphenyl ethers [BDE47, BDE99, BDE100, BDE153 and BDE154) and two methoxy-tetraBDEs [6-methoxy-2,2',4,4'- tetraBDE (6-MeO-BDE47) and 2'-methoxy-2,3',4,5'- tetraBDE (2'-MeO-BDE68)] in pike from Lake Bolmen for the years 1967-2000, are presented. All BDE congeners show increasing trends up to the mid-1980s (Sigma5PBDE from 60 to 1600 pg/g wet weight in 1989, i.e. a more than 25-fold increase), and then decrease or level off. The decreasing trends of PBDEs after the 1980s were considerably slower in the present study than was found in a study of an environmental matrix from the Baltic Proper covering the same time period. This difference suggests local sources near Lake Bolmen. The MeO-BDEs show initially decreasing concentrations, which for 6-MeO-BDE47 continues until the early 1990s. The concentrations of 6-MeO-BDE47 in herring from five locations along the Swedish coast increased from south to north in the Baltic Sea. No correlation between the concentrations of the BDE congeners and the MeO-BDEs was observed, indicating sources other than PBDEs for these compounds. The presence of MeO-BDEs in fish from lakes with different characteristics suggests a natural production not favoured by eutrophication, or dependent on sampling season and geographical location. 相似文献

A systems perspective is used to discuss the concept of sustainability. From this perspective, it is argued, sustainability can be regarded as a system state that is mediated by specific structures. This is fundamentally different from regarding sustainability merely as a normative goal, as it is presently regarded by most. Insight into the kinds of structures which mediate a system’s state open the door to proactive design of new structures and mechanisms, which are necessary for yielding effective change: in this case, promoting the sustainability agenda. The kind of change required to transform the prevailing trajectory of human affairs is presented as a second order change: a change that requires a major shift, and a complete transformation of the system itself, not only in a few aspects of its behavior. A new definition of sustainability is offered, anchored in the interaction of a population and the carrying capacity of its environment. From this definition, five core sustainability principles are derived, along with their respective policy and operational implications. Together, these principles prescribe the conditions that must be met to attain sustainability as an enduring state. The principles themselves form an integrated, systemic set, which requires them to be acted on simultaneously. A piecemeal approach—focusing on one aspect while neglecting others—is not likely to yield effective results for the whole. 相似文献

Concentrations of individual polychlorinated biphenyls (PCBs) were determined in blubber, liver, kidney and brain tissues of 10 male harbour seals (Phoca vitulina) of different ages. The animals were found dead or dying on the southern coast of Norway in 1988 during the morbilli virus epizootic. Twenty-three PCB congeners were investigated in all tissues by capillary gas chromatography with electron capture detection and comparison with standards of the respective congeners (IUPAC numbers 28, 52, 74, 99, 101, 105, 110, 114, 118, 128, 138, 141, 149, 153, 156, 157, 170, 180, 183, 187, 194, 206 and 209). In addition, three toxic coplanar congeners (IUPAC numbers 77, 126 and 169) were investigated in the blubber samples. Total PCB (sum of the determined congeners) in kidney (12-60 nmol g(-1)), liver (12-90 nmol g(-1)) and blubber (10-79 nmol g(-1)) were similar on extractable lipid basis, while total PCB in brain lipid was about 1/10 (1.6-12 nmol g(-1)). The absolute concentrations of the individual congeners in brain lipid correlated poorly with the respective concentrations in each of the other tissues, while the individual congener concentrations in the lipid fractions of kidney, liver and blubber tended to correlate with each other. The relative concentrations of the sum of the most persistent congeners (PCB-99, -128, -138, -153, -170, -180, -187 and -194) correlated in all tissues, and an increase of the relative concentrations of these congeners, most distinct in blubber, with higher carcass weight, was shown. Also, total PCB in blubber correlated with carcass weight. When comparing the relative concentration patterns of individual congeners (per cent of total) in the four tissues, six congeners differed significantly between tissues, and the relative brain concentrations showed most deviation from the other tissues. The concentration of the coplanar PCBs was 1 : 1000-1 : 1000 of the total PCB concentration. Aerosol matter in the size range <2 μm was collected in a Berner impactor and subsequently analysed by scanning electron microscopy (SEM) and energy dispersive X-ray (EDX) spectrometry. Owing to the low electron beam energy of 5 keV (occasionally 10 keV), analysis was restricted to elements with atomic numbers 20 (Ca). Sub-micrometer aerosol matter was found to contain mostly S, O, and C as well as some K and Ca. Nitrogen appeared to escape detection, probably due to bombardment-induced sublimation of NO3 and NH4. During sampling at low to moderate relative humidity (<60%) the sulphur-rich fraction of the aerosol matter (most likely sulphates) regrew in the form of microcrystals with sizes up to 10 times larger than the mean aerodynamic diameter of the respective impactor stage. By contrast, when sampling during periods in course of which the relative humidity exceeded 70%, the aerosol matter regrew in the form of extended amorphous agglomerates. The aerosol deposits also contained large numbers of carbon nanoparticles, well separated from the regrown sulphate-rich matter. The nanoparticles were similar in size (20–40 nm), much smaller than the equivalent aerodynamic diameter of the impacting particles (63 nm–2 μm). Presumably, the carbon nanoparticles constituted the core of larger air-borne particles covered with sulphates (as well as with nitrates and organic carbon). The regrown microcrystals disappeared rapidly under electron bombardment at high current density, an observation that indicates high volatility at elevated temperatures. Aerosol matter collected in the size range between 1 and 2 μm contained large fractions of particles made of O, Si, P, K, and Ca (oxides). These particles were highly resistant to electron bombardment (hard) and showed little or no evidence for agglomeration or regrowth. After removing the soluble (acidic) material from the collected aerosol matter, only carbon nanoparticles and hard coarse particles were left behind. The observation of agglomerated or crystallized “soft” aerosol matter in combination with phase separation of carbon nanoparticles lends further support to the assertion that it is not possible to collect useful quantities of fine and ultrafine aerosol particles with as-suspended morphology. Myanmar's protected area (PA) system began nearly 150 years ago under royal patronage. Park policies and practices, embodied in 19 pieces of legislation developed sporadically during and after the colonial period. As a result of the FAO-UNDP's Nature Conservation and National Parks Project (1981-1985) the government established the Nature & Wildlife Conservation Division and placed it within the Forest Department as the agency responsible for PA management. As a consequence the number of parks increased from 14 to 33. Myanmar's median park size is less than 50 km(2), but only five parks (15%) are larger than 1000 km(2). Most parks conserve terrestrial habitats; parks encompassing inland wetlands, mangrove, and marine habitats are limited in number and size. Existing PAs unequally represent Myanmar's ecosystems; the Malay Transition, Burmese coast, Burmese Transition and Cardamom Mountains bio-units are under-represented within the system. The effective total PA size (i.e., area of all parks less the area of 13 paper parks) is currently about 2.1%. Budgetary support for parks increased 11% since 1996, but is insufficient to address park needs, particularly in remote parks that are understaffed. Limited education and training of PA staff is a major factor limiting effective park management. Fifty-eight percent of park wardens are educated as foresters, and 42% have university degrees. The average posting in a park is 4 years, which is less than ideal for management continuity. Recommended actions to secure Myanmar's PAs include evaluation and reformulation of policies, increasing representation of Myanmar's habitats within the PA system, management planning, and standardizing protocols for anti-poaching patrols and other forms of law enforcement. Improved leadership training for wardens and range forest officers can also improve park management. Funding for community relations and more integrated management of parks and people can reduce conflicts, while environmental education and outreach activities can provide some needed services. Finally, new ways and means are needed to support these activities. Myanmar's PAs need creative and sustainable means of support, including partnerships with non-government organizations to supplement traditional means of support. 相似文献

Abstract The effect of intracheally administered DDT (5 mg/100 g body weight) or endosulfan (1 mg/100 g body weight) for three cosecutive days have been studied on lipid metabolism of rat lung subcellular fractions. Both the insecticides did not affect the lung weight and the protein contents of microsomes, lamellar bodies and surfactant but significantly increased the phospholipid contents of microsomal and surfactant system. Most of the neutral lipid components of lung subcellular fractions were also increased by DDT or endosulfan treatments, except that of surfactant triglycerides which were decreased by DDT treatment. DDT or endosulfan both increased the incorporation of radioactive [methyl‐3H]choline into microsomal phosphatidylcholine (PC) and surfactant dipalmitoylphosphatidylcholine (DPPC) without affecting the incorporation of radioactive [methyl‐14C]methionine, showing the increased synthesis of PC via CDPcholine pathway. The results presented in this communication showed that DDT and endosulfan, the two different chloroinsecticides have similar effects on microsomal lipid metabolism but produce different biochemical manifestations on the secretion of surfactant phospholipids. 相似文献
