An electrochemical COD (chemical oxygen demand) sensor using an electrode-surface grinding unit was investigated. The electrolyzing (oxidizing) action of copper on an organic species was used as the basis of the COD measuring sensor. Using a simple three-electrode cell and a surface grinding unit, the organic species is activated by the catalytic action of copper and oxidized at a working electrode, poised at a positive potential. When synthetic wastewater was fed into the system, the measured Coulombic yields were found to be dependent on the COD of the synthetic wastewater. A linear correlation between the Coulombic yields and the COD of the synthetic wastewater was established (10-1000 mg L(-1)) when the electrode-surface grinding procedure was activated briefly at 8 h intervals. When various kinds of wastewater samples obtained from various sewage treatment plants were measured, linear correlations (r(2)> or = 0.92) between the measured EOD (electrochemical oxygen demand) value and COD of the samples were observed. At a practical wastewater treatment plant, the measurement system was successfully operated with high accuracy and good stability over 3 months. These experimental results show that the application of the measurement system would be a rapid and practical method for the determination of COD in water industries.  相似文献

Fungal laccase-catalyzed degradation of hydroxy polychlorinated biphenyls   总被引:9,自引:0,他引:9

Hydroxy polychlorinated biphenyls (hydroxy PCBs) are toxic metabolites of PCBs. Their toxicity such as strong endocrine disruption demands effective remediation methods. Laccases from Trametes versicolor and Pleurotus ostreatus were tested to degrade hydroxy PCBs. Optimum pHs for both enzymes were around 4.0. Laccase from T. versicolor degrades hydroxy PCBs more rapidly than that from P. ostreatus. The enzymatic activities remained little changes in up to 10% organic solvents, but decreased rapidly in more than 10% acetone, acetonitrile or DMSO. Degradation rate constants decreased with increase of chlorination and no degradation was observed with tetra-, penta- and hexa-chloro hydroxy PCBs in non-mediated reactions. However, the tetra- to hexa-chloro hydroxy PCBs were degraded by laccase from T. versicolor in the presence of the mediator 2,2,6,6-tetramethylpiperidine-N-oxy radical. The other mediators, 4-ethyl-2-methoxyphenol, 2,2'-azino-bis(3-ethylbenzthiazoline sulfonic acid) diammonium salt and 1-hydroxybenzotriazole and humic acid, also enhanced degradation of all the hydroxy PCBs except 4-hydroxy-2',3,3',4',5,5'-hexachlorobiphenyl. The results showed that 3-hydroxy biphenyl was more resistant to laccase degradation than 2- or 4-hydroxy analogues. Significant linear-correlations (coefficient of determination, r2 = 0.9097 and 0.8186 for laccases from P. ostreatus and T. versicolor, respectively) were found between the ionization potentials and the removal rate constants of hydroxy PCBs.  相似文献

Stenotrophomonas maltophilia T3-c, isolated from a biofilter for the removal of benzene, toluene, ethylbenzene, and xylene (BTEX), could grow in a mineral salt medium containing toluene, benzene, or ethylbenzene as the sole source of carbon. The effect of environmental factors such as initial toluene mass, medium pH, and temperature on the degradation rate of toluene was investigated. The cosubstrate interactions in the BTEX mixture by the isolate were also studied. Within the range of initial toluene mass (from 23 to 70 pmol), an increased substrate concentration increased the specific degradation of toluene by S. maltophilia T3-c. The toluene degradation activity of S. maltophilia T3-c could be maintained at a broad pH range from 5 to 8. The rates at 20 and 40 degrees C were 43 and 83%, respectively, of the rate at 30 degrees C. The specific degradation rates of toluene, benzene, and ethylbenzene by strain T3-c were 2.38, 4.25, and 2.06 micromol/g-DCW/hr. While xylene could not be utilized as a growth substrate by S. maltophilia T3-c, the presence of toluene resulted in the cometabolic degradation of xylene. The specific degradation rate of toluene was increased by the presence of benzene, ethylbenzene, or xylene in binary mixtures. The presence of toluene or xylene in binary mixtures with benzene increased the specific degradation rate of benzene. The presence of ethylbenzene in binary mixtures with benzene inhibited benzene degradation. The presence of more than three kinds of substrates inhibited the specific degradation rate of benzene. All BTEX mixtures, except tri-mixtures of benzene, ethylbenzene, and xylene or mixtures of all four substrates, had little effect on the degradation of ethylbenzene by S. maltophilia T3-c. The utilization preference of the substrates by S. maltophilia T3-c was as follows: ethylbenzene was degraded fastest, followed by toluene and benzene. However, the specific degradation rates of substrates, in order, were benzene, toluene, and ethylbenzene.  相似文献

A four-stage biological nutrient removal (BNR) process was operated to investigate the effect of anaerobically fermented leachate of food waste (AFLFW) as an external carbon source on nutrient removal from domestic wastewater having a low carbon-to-nitrogen ratio. The BNR system that was supplemented with AFLFW showed a good performance at a sludge retention time (SRT) of 30 days, despite low temperature. With this wastewater, average removal efficiencies of soluble chemical oxygen demand (COD), total nitrogen (T-N), and total phosphorus (T-P) were 88 to 93%, 70 to 74%, and 63 to 68%, respectively. In this study, several kinds of poly-hydroxyalkanoates (PHAs) were observed in cells. These included 24% poly-3-hydroxybutyrate (PHB), 41% poly-3-hydroxyvalerate (PHV), 18% poly-3-hydroxyhexanoate (PHH), 10% poly-3-hydroxyoctanoate (PHO), 5% poly-3-hydroxydecanoate (PHD). and 2% poly-3-hydroxydodecanoate (PHDD), indicating that microorganisms could store various PHAs through the different metabolic pathways. However, breakdown of the enhanced biological phosphorus removal (EBPR) mechanism was observed when SRT increased from 30 to 50 days for the enhancement of nitrification. To study the effect of SRT on EBPR, a sequencing batch reactor (SBR) system that was supplied with glucose was operated at various SRTs of 5, 10, and 15 days. Nitrification and denitrification efficiencies increased as SRT increased. However, the content of intracellular materials such as PHAs, glycogen. and poly-P in cells decreased. From these results, it was concluded that SRT should be carefully controlled to increase nitrification activity and to maintain biological phosphorus removal activity in the BNR process.  相似文献

It is now acknowledged that bacteria from gut microbiota deeply interact with their host by altering many physiological traits. Such interplay is likely to consequently affect stress tolerance. Here, we compared cold and heat tolerance of Drosophila melanogaster flies with undisrupted (control (Co)) versus disrupted gut microbiota (dechorionated eggs (De)). The disrupting treatment strongly reduced bacterial load in flies’ guts, though 16S sequencing analysis did not evidence strong diversity changes in the remaining bacterial community. Both chill coma recovery and acute cold survival were repeatedly lower in De than in Co flies under our experimental conditions. However, heat tolerance was not consistently affected by gut disruption. Our results suggest that microbiota-related effects on the host can alter ecologically relevant traits such as thermal tolerance.  相似文献

We have developed a new nanofilter using a carbon nanotube-silver composite material that is capable of efficiently removing waterborne viruses and bacteria.The nanofilter was subjected to plasma surface treatment to enhance its flow rate,which was improved by approximately 62%.Nanoscale pores were obtained by fabricating a carbon nanotube network and using nanoparticle fixation technology for the removal of viruses.The pore size of the nanofilter was approximately 38 nm and the measured flow rate ranged from 21.0 to 97.2 L/(min·m~2)under a pressure of 1–6 kgf/cm~2 when the amount of loaded carbon nanotube-silver composite was 1.0 mg/cm~2.The nanofilter was tested against Polio-,Noro-,and Coxsackie viruses using a sensitive real-time polymerase chain reaction assay to detect the presence of viral particles within the outflow.No trace of viruses was found to flow through the nanofilter with carbon nanotube-silver composite loaded above 0.8 mg/cm~2.Moreover,the surface of the filter has antibacterial properties to prevent bacterial clogging due to the presence of 20-nm silver nanoparticles,which were synthesized on the carbon nanotube surface.  相似文献

The resin capsule system (RCS) was tested as a means of providing data on the presence and forms of volatile hydrocarbons. Results indicated that resin capsules provided data showing sensitivity to soil variables (texture and moisture content) and time. The objectives of this paper are to evaluate the RCS methodology and to determine whether carbonaceous resin capsules provide results that can be described by fundamental chemical partitioning and kinetic principles. Findings revealed a significant relationship between quantities of benzene, toluene, ethylbenzene, and xylene adsorbed on the capsule and quantities partitioned into the vapor phase. Kinetic evaluation indicated that the vapor adsorption by the resin capsule is regulated by diffusion processes. No verification of rate-limiting processes was possible due to limitations imposed by the experimental design, but it appears that during early stages, adsorption rate was limited by vapor diffusion through the soil. The resin capsule data also reflected differences that would be expected due to properties of the organic liquids present. These results provide further evidence that the RCS could be developed to suggest direct in situ monitoring to reveal quantities and nature of organic substances in soils.  相似文献

This study analyzed the concentrations of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA) and perfluorohexane sulfonate (PFHxS) in maternal and umbilical cord sera at delivery from the general population in Korea. Seventy samples were analyzed with ion-pairing and LC/MS/MS. PFOS, PFOA and PFHxS were detected in both maternal and umbilical cord sera. There was a high correlation of PFC concentrations between maternal and cord serum samples, implying transplacental transport. Ranking of transplacental transfer efficiency was PFOA > PFHxS > PFOS. Student’s t-tests revealed that concentrations of maternal PFOA were related with decreases in birth weight, birth length and ponderal index, suggesting a possible impact on fetal growth. With multiple logistic regression models, maternal PFOS concentration showed a significant inverse association with ponderal index (OR = 0.22; 95% CI, 0.05–0.90). Umbilical cord PFHxS concentration showed a significant inverse association with birth weight (OR = 0.26; 95% CI, 0.08–0.85) or a marginally significant inverse association with birth length (OR = 0.33; 95% CI, 0.09–1.17). This is the first report demonstrating an inverse association of birth outcomes with PFHxS exposure. Concentrations of maternal PFOA were decreased with parity, implying that delivery is one of the major routes for PFOA elimination in women. This study demonstrated prenatal exposure of PFCs through placental transfer which could result in possible developmental effects in the population sampled. Our results may provide data basis to conduct a larger scale investigation into developmental effects of PFCs in the future and contribute to understanding levels of PFC contaminations from a variety of populations in the globe.  相似文献

Overcrowding that happens in places like concerts, stadiums or pilgrimage locations might sometimes cause injury or loss of life. Maintaining the safety of crowd in these places is therefore very important. In addition, increasing the performance of the buildings and structures has always been an important concern. Most of the previous work focused on using new devices and methods for monitoring and management of the crowd but they rarely focus on a comprehensive and structured approach with the purpose of increasing efficiency and safety. In this paper, we explore a Russian “Theory of Inventive Problem Solving”, TRIZ, to see whether its principles can help us to solve or improve overcrowding issues. More specifically we find the contradictions that arise in the context of crowd management and we observe which of the categorical solutions suggested by TRIZ might possibly be useful for our problem. Increasing the crowd capacity in a relatively small area, which leads to overcrowding, is one of the common contradictions in crowd management. TRIZ has been suggested as a good method for solving problems which involve contradiction and thus, chosen for this purpose.  相似文献
