The accuracy of biochemical and molecular prenatal diagnoses using chorionic villi as the fetal source was assessed by seven centres participating in the NICHD collaborative study on the safety and accuracy of chorionic villus sampling (CVS) and amniocentesis. Of 601 pregnancies studied, biochemical methods were used to determine the diagnosis in 283 fetuses at risk for 35 different metabolic disorders. Fifteen different lysosomal storage diseases accounted for 81 per cent of the biochemical prenatal diagnoses performed, with 57 per cent of these pregnancies at risk for Tay-Sachs disease. No errors were made in the biochemical diagnoses that predicted affected or unaffected fetuses. However, the diagnoses of certain disorders (e.g., mucopolysacchariodosis type IH, metachromatic leukodystrophy, and Krabbe disease) occasionally required confirmatory studies in cultured amniocytes because the enzyme results were inconclusive in direct and/or cultured villi or due to the presence of a pseudodeficiency allele. Of these, only the diagnosis of a fetus at risk for Krabbe disease remained inconclusive after special studies to discriminate between mutant and pseudodeficiency alleles. Recombinant DNA techniques were used to predict the diagnosis of 318 fetuses at risk for 16 different disorders in which the defective disease gene could be detected either directly or by linkage analysis to a nearby polymorphic marker. Of these, 32 per cent were for haemoglobinopathies, 25 per cent for cystic fibrosis, 24 per cent for Duchenne or Becker muscular dystrophy, and 7 per cent for haemophilias. Pregnancies at risk for known disorders with specific molecular lesions (e.g., sickle cell disease) were accurately diagnosed in direct and/or cultured villi. Diagnoses requiring analyses with closely linked polymorphic markers were occasionally uninformative or inconclusive. Maternal contamination was not reported in any biochemical or molecular-based diagnosis. These studies document the high accuracy and rapidity of both biochemical and mutation-specific prenatal diagnoses with direct and cultured chorionic villi.  相似文献

Climate modelers often use agreement among multiple general circulation models (GCMs) as a source of confidence in the accuracy of model projections. However, the significance of model agreement depends on how independent the models are from one another. The climate science literature does not address this. GCMs are independent of, and interdependent on one another, in different ways and degrees. Aquatic floating plants on BioHaven mats were tested for their potential use as a Best Management Practice to be incorporated within existing stormwater detention ponds. Plants were analyzed for their capability to remove nutrient-pollution in parallel with the study of ecological dynamics. Experiments were carried out in cylindrical mesocosms of 5 m diameter and 1.2 m height, above-ground pools with a water volume of 14 m3. The design parameters tested were for 5% and 10% vegetated floating island coverage of the mesocosm, both with and without shoreline plants called littoral zone. This littoral shelf was 0.5 m thick, graded at a downward slope of 1:5 toward the center using loamy soil with low organic matter content, excavated from below turf grass. Endemic plant species were chosen for the experimental location in central Florida based on a wetland identification manual by the Florida Department of Environmental Protection to ensure the study was not compromised by unique climate requirements of the plants. Nutrient and aquatic chemical conditions such as pH, dissolved oxygen, temperature, turbidity, and chlorophyll a were monitored to understand their relationships to the general wetland ecosystem. Real-time polymerase chain reaction analysis identified the microbial activity near the rhizospheric zone. Logistical placement considerations were made using spatial sampling across the horizontal plane of the mesocosms, beneath and around the root zone, to determine if nutrients tend to aggregate around the floating island. This study concluded that the application of floating islands as a stormwater technology can remove nutrients through plant uptake and biological activity. The most cost-effective size in the outdoor mesocosms was 5% surface area coverage of the mat.  相似文献

Salinity, selenium, and uranium pose water‐quality challenges for the Arkansas River in southeastern Colorado and other rivers that support irrigation in semiarid regions. This study used 31 years of continuous discharge and specific conductance (SC) monitoring data to assess interannual patterns in water quality using mass balance on a 120‐km reach of river. Discrete sampling data were used to link the SC records to salinity, selenium, and uranium. Several important patterns emerged. Consumptive use reduced discharge by a median value of 33% and drove corresponding increases in salinity and uranium concentrations. Increased water availability for irrigation from rainfall and upstream snowpack in 1995–1999 flushed additional salinity and uranium into the river in 1996–2000; average annual total dissolved solids (salinity) concentrations increased 25%, and loads increased 131%. Smaller flushing events have occurred, sometimes lagging an increase in water availability by about one year. The pattern indicates flushing of salts temporarily stored, evaporatively concentrated, or of geologic origin. Mobilization of selenium from the reach was minor compared to salinity and uranium, and net selenium removal from the river was suggested in some years. Several processes related to irrigation could be removing selenium. The results provide context for efforts to improve water quality in the Arkansas River and rivers in other semiarid regions.  相似文献

The dissipation of hexazinone (Velpar) in two tropical soil types in Kenya was studied under field and semi-controlled conditions for a period of 84 days. The dissipation was found to be very rapid and this could be attributed to adverse weather conditions including high initial rainfall as well as to low soil-organic-matter content, volatilization, surface run-off and biodegradation. The DT50 values of dissipation obtained by first order kinetics were 20 days and 21.3 days in clay and loam soil types, respectively. The influence of bargasse compost (1000 μg/g dry soil) was also studied and was found to enhance dissipation to some extent, giving DT50 values of 18 days and 18.3 days in clay and loam soil types, respectively.  相似文献

It is broadly recognized that river delta systems around the world are under threat from a range of anthropogenic activities. These activities occur at the local delta scale, at the regional river and watershed scale, and at the global scale. Tools are needed to support generalization of results from case studies in specific deltas. Here, we present a methodology for quantitatively constructing an empirical typology of anthropogenic change in global deltas. Utilizing a database of environmental change indicators, each associated with increased relative sea-level rise and coastal wetland loss, a clustering analysis of 48 global deltas provides a quantitative assessment of systems experiencing similar or dissimilar sources and degrees of anthropogenic stress. By identifying quantitatively similar systems, we hope to improve the transferability of scientific results across systems, and increase the effectiveness of delta management best practices. Both K-Means and Affinity Propagation clustering algorithms find similar clusters, with relative stability across small changes in K-Means cluster number. High-latitude deltas appear similar, in terms of anthropogenic environmental stress, to several low-population, low-latitude systems, including the Amazon delta, despite substantially different climatic regimes. Highly urbanized deltas in Southeast Asia form a distinct cluster. By providing a quantitative boundary between groups of delta systems, this approach may also be useful for assessing future delta change and sustainability given projected population growth, urbanization, and economic development trends.  相似文献

In this study soils from sugarcane-cultivated fields were screened for bacterial species capable of atrazine (6-chloro-N2-ethyl-N?-isopropyl-1,3,5-triazine-2,4-diamine) degradation due to long exposure of the soils to this herbicide. To enrich for atrazine degraders, Minimal Salt Medium containing atrazine as the sole N source and glucose as the C source was inoculated with soils impacted with this herbicide and incubated. Bacterial growth was monitored by measuring optical density. The degradation of atrazine was followed by measuring residual atrazine in liquid cultures over a given time period by high performance liquid chromatography. Bacterial strains isolated from the enrichment cultures were characterized by biochemical tests and identified by 16S rRNA gene sequencing. Two bacterial strains coded ISL 8 and ISL 15 isolated from two different fields were shown to have 94 and 96% 16S rRNA gene sequence similarity to Burkholderia cepacia respectively. Another bacterial sp., ISL 14 was closely related to Enterobacter cloacae with a 96% 16S rRNA gene sequence similarity. There was not much difference between the extents of atrazine degradation by the enrichment cultures with communities (79-82% applied amount) from which pure strains were isolated and the pure strains themselves in liquid cultures that showed a degradation of 53-83% of applied amount. The study showed existence of bacterial strains in different sugarcane-cultivated fields which can use atrazine as a nitrogen source. The bacterial strains isolated can be used to enhance the degradation of atrazine in contaminated soils where atrazine is still considered to be recalcitrant.  相似文献

Recent severe drought events have occurred over the Ogallala Aquifer region (OAR) during the period 2011–2015, creating significant impacts on water resources and their use in regional environmental and economic systems. The changes in terrestrial water storage (TWS), as indicated by the Gravity Recovery and Climate Experiment (GRACE), reveals a detailed picture of the temporal and spatial evolution of drought events. The observations by GRACE indicate the worst drought conditions occurred in September 2012, with an average TWS deficit of ~8 cm in the northern OAR and ~11 cm in the southern OAR, consistent with precipitation data from the Global Precipitation Climatology Project. Comparing changes in TWS with precipitation shows the TWS changes can be predominantly attributable to variations in precipitation. Power spectrum and squared wavelet coherence analysis indicate a significant correlation between TWS change and the El Nino‐Southern Oscillation, and the influence of equatorial Pacific sea surface temperatures on TWS change is much stronger in the southern OAR than the northern OAR. The results of this study illustrate the value of GRACE in not just the diagnosis of significant drought events, but also in possibly improving the predictive power of remote signals that are impacted by nonregional climatic events (El Nino), ultimately leading to improved water resource management applications on a regional scale.
