Methylmercury is an important source of environmental contamination and the central nervous system (CNS) is one of the main target organs. Methylmercury genotoxicity was already demonstrated in peripherical tissues but was never detected in the brain. Thus, the objective of this work was to verify its genotoxic effect using brain cell lines. Glioblastoma (U373) and neuroblastoma (B103) human cell lines were exposed to methylmercury (0-10 microM). By measuring cellular viability, concentrations inducing <20% of cellular death (P<0.05) were selected: 1 and 0.1 microM. To detect micronuclei, 200,000 cells were treated with methylmercury for 24 h, and then incubated with cytochalasin B (2 microg/ml) for 72 h (U373) or 48 h (B103). The binucleation index, frequency of micronucleated cells, micronucleation index, metaphasic index and index of nucleoplasmic bridges were determined. Statistical analysis showed indices and percentages significantly higher (P<0.05) in methylmercury-treated cells. Each cell line was shown to be differently sensitive to each biomarker of genotoxic damage, which seems to indicate the existence of different mechanisms of toxicity. This work demonstrates, for the first time, MeHg ability to provoke genotoxicity in cells of brain origin with relatively low levels of exposure.  相似文献

The aim of this study was to determine the impact of specific environmental contaminants on the activities of antioxidant enzymes in terrestrial, free-living birds. To investigate this, several biomarkers and the levels of lead, mercury, and arsenic were determined in blood of white stork (Ciconia ciconia) nestlings from two different colonies located in the west of Spain that were exposed to different environmental conditions and human activities. Different biomarker and contaminant patterns were observed between the two colonies, suggesting different levels of environmental contamination. In comparison to nestlings from a natural reference area, the levels of the three hazardous elements Pb, As, and Hg were significantly higher in the colony nearby a landfill. Analysis of antioxidant biomarkers revealed significantly higher levels of glutathione and glutathione S-transferase activity in birds from the colony close to the landfill site than were found in those from the “control” area. In contrast, the levels of malondialdehyde did not significantly differ between the two sampling sites. In addition, a positive correlation between glutathione and both Hg and As has been observed. Taken together, exposure to the three hazardous elements is reflected by enhanced activity of glutathione S-transferases, and elevated levels of glutathione and malondialdehyde. White storks can therefore be considered as a suitable model species for environmental monitoring programs.  相似文献

To assess the interaction between testosterone (T) treatment and acclimation to different salinities, seawater-acclimated gilthead sea bream (Sparus auratus) were implanted with slow-release coconut oil implants alone (control) or containing T (5 μg/g body mass). After 5 days, eight fish of control and T-treated groups were sampled. The same day, eight fish of each group were transferred to low salinity water (LSW, 6 ppt, hypoosmotic test), seawater (SW, 38 ppt, control test) and high salinity water (HSW, 55 ppt, hyperosmotic test) and sampled 9 days later. Gill Na+, K+-ATPase activity increased in HSW-acclimated fish with respect to SW- and LSW-acclimated fish in both control and T-treated groups. Kidney Na+, K+-ATPase activity was also enhanced in HSW-acclimated fish, but only in T-treated group. From a metabolic point of view, most of the changes observed can be attributed to the action of salinity and T treatment alone, since few interactions between T treatment and osmotic acclimation to different salinities were observed. Those interactions included in treated fish: in the liver, decreased capacity in using glucose in fish acclimated to extreme salinities; in the gills, decreased capacity in using amino acids in HSW; in the kidneys increased capacity in using amino acids in extreme salinities; and in the brain, decreased glycogen and acetoacetate levels of fish in LSW.  相似文献

In this study, we investigate association patterns of 249 bottlenose dolphin feeding groups off Sardinia Island (Italy) from January 2000–May 2007 and describe how their association behaviour is related to their response to food patches created by a marine fin fish farm. We also tested the hypothesis that dolphins have different social structures with different feeding activities: Associations should decrease during opportunistic feeding behaviours as it is easier to capture prey, and cooperation is not as necessary. Sixteen individually identified bottlenose dolphins were observed participating in both opportunistic and not opportunistic feeding activities, with a mean of 30 ± 8 times and 9.6 ± 1 times, respectively. Bottlenose dolphins show non-random social behaviour during feeding and this behaviour differs depending on their specific foraging activity. Dolphin associations during feeding can be divided into three categories: acquaintances, affiliates, and feeding associates. Association behaviour during fish farm feeding is consistent with our hypothesis that during opportunistic behaviours, benefits from cooperation decrease, as it is easier to capture prey. Group size homogeneity in both feeding activities demonstrates that the number of dolphins engaging in foraging is not necessarily related with cooperation levels. Moreover, an adult dolphin may prefer to associate with a specific individual, independent of the sex, who shares the same foraging priorities. This study is the first to show how aquaculture is not only directly affecting marine predators but could also indirectly affect their social structure and behaviour.  相似文献

Background

This paper describes a conceptual framework for solutions-focused management of chemical contaminants built on novel and systematic approaches for identifying, quantifying and reducing risks of these substances.

Methods

The conceptual framework was developed in interaction with stakeholders representing relevant authorities and organisations responsible for managing environmental quality of water bodies. Stakeholder needs were compiled via a survey and dialogue. The content of the conceptual framework was thereafter developed with inputs from relevant scientific disciplines.

Results

The conceptual framework consists of four access points: Chemicals, Environment, Abatement and Society, representing different aspects and approaches to engaging in the issue of chemical contamination of surface waters. It widens the scope for assessment and management of chemicals in comparison to a traditional (mostly) perchemical risk assessment approaches by including abatement- and societal approaches as optional solutions. The solution-focused approach implies an identification of abatement- and policy options upfront in the risk assessment process. The conceptual framework was designed for use in current and future chemical pollution assessments for the aquatic environment, including the specific challenges encountered in prioritising individual chemicals and mixtures, and is applicable for the development of approaches for safe chemical management in a broader sense. The four access points of the conceptual framework are interlinked by four key topics representing the main scientific challenges that need to be addressed, i.e.: identifying and prioritising hazardous chemicals at different scales; selecting relevant and efficient abatement options; providing regulatory support for chemicals management; predicting and prioritising future chemical risks. The conceptual framework aligns current challenges in the safe production and use of chemicals. The current state of knowledge and implementation of these challenges is described.

Conclusions

The use of the conceptual framework, and addressing the challenges, is intended to support: (1) forwarding sustainable use of chemicals, (2) identification of pollutants of priority concern for cost-effective management, (3) the selection of optimal abatement options and (4) the development and use of optimised legal and policy instruments.

This study was based on a groundwater vulnerability assessment approach implemented for the Mexico City Metropolitan Area (MCMA). The approach is based on a fuzzy multicriteria procedure integrated in a geographic information system. The approach combined the potential contaminant sources with the permeability of geological materials. Initially, contaminant sources were ranked by experts through the Analytic Hierarchy Process. An aggregated contaminant sources map layer was obtained through the simple additive weighting method, using a scalar multiplication of criteria weights and binary maps showing the location of each source. A permeability map layer was obtained through the reclassification of a geology map using the respective hydraulic conductivity values, followed by a linear normalization of these values against a compatible scale. A fuzzy logic procedure was then applied to transform and combine the two map layers, resulting in a groundwater vulnerability map layer of five classes: very low, low, moderate, high, and very high. Results provided a more coherent assessment of the policy-making priorities considered when discussing the vulnerability of groundwater to organic compounds. The very high and high vulnerability areas covered a relatively small area (71 km2 or 1.5% of the total study area), allowing the identification of the more critical locations. The advantage of a fuzzy logic procedure is that it enables the best possible use to be made of the information available regarding groundwater vulnerability in the MCMA.  相似文献

The monetary valuation of ecosystem services has proved to be a powerful tool for influencing management decisions, as it can be used to quantify the relative importance of various ecosystem functions. However, these valuation methods are rarely implemented with adequate care; in particular, they do not consider spatial and temporal variations in the services offered and demanded. This paper examines the effect of heterogeneity on the valuation of cultural services offered by the Doñana natural protected area in Spain. We apply the zonal travel cost method at various spatial scales, and the individual travel cost method at various temporal scales. We conclude that economic valuation techniques must account for spatial and temporal heterogeneity if they are to provide accurate and realistic information.  相似文献

Surface waters located in intensive agricultural areas are more vulnerable to the pesticides contamination, which is a major concern if the water is intended to be used for human consumption. The aim of this study was to evaluate the presence and the distribution of pesticides in the Alqueva reservoir, an important source of water supply (South of Portugal), considering their representativeness in the agricultural practice of the area. For the analysis of pesticides risk impact we used the environmental quality standards in the field of water policy proposed recently by the European Commission. The pesticides belonging to the classes of phenylureas, triazines, chloroacetanilides, organophosphorous and thiocarbamates were analysed by on-line solid phase extraction-liquid chromatography-tandem mass spectrometry. The pesticides more frequently detected were atrazine, simazine, diuron and terbuthylazine. The highest levels of these pesticides were registered in spring, after pesticides treatment, namely in olive-tree and vine crops. The priority pesticides atrazine and diuron reached values above the annual average proposed in the European Union Legislation. The herbicide atrazine reached values that surpassed the proposed maximum allowable concentration (2 000 ng L? 1). The sampling stations most affected by these pesticides were Sra. Ajuda, Lucefecit and Alcarrache, located in the northern part of the reservoir, closer to Spain where the agricultural activity is more intensive.  相似文献

A gel permeation chromatographic (GPC) clean-up based method was developed for determination of selected organochlorine pesticides and polychlorinated biphenyls in human serum. The method permits automation of the sample extract clean-up stage and is designed to work with small amounts of sample. Different working variables were studied in its development, including injection volume, flow rate, and fat amount as the most representative coextract. The method provides solvent and time savings. Initial extraction was performed using 96-well solid-phase disk extraction plates, and quantification was performed by gas chromatography with electron capture detection and mass spectrometry. Recoveries of PCB congeners 28, 52, 101, 118, 138, 153, and 180 and organochlorine pesticides HCB, β-HCH, γ-HCH, heptachlor epoxide, p,p′-DDE, and p,p′-DDT at two spiking levels ranged from 55% to 115% with relative standard deviations ranging from 2.0% to 14.6%. Limits of quantification ranged from 0.06 to 0.16 ng mL−1 for PCBs and from 0.12 to 0.36 ng mL−1 for organochlorine pesticides. Finally, the method was applied to the analysis of 38 serum samples and the results were compared with those of another procedure validated at the laboratory.  相似文献
