AbstractAn industrial-scale, profitable method for production of the most widely used bioinsecticide, Bacillus thuringiensis (Bt), is challenging because of its widespread application. The aim of this study is to present a strategy to develop a low-cost, large-scale bioprocess to produce Bt H14.This study was first focused on the design of a culture medium composed of economical and available components, such as glycerol and lysed Saccharomyces cerevisiae. The production goal of 1200 ITU was achieved using a medium composed of 20:20 g L?1of glycerol:lysed yeast in batch cultures. Efforts were subsequently focused on the design of an appropriate culture system, and an original two-stage culture system was proposed. First, yeast (the primary component of the culture medium) are cultivated using a minimal mineral medium and lysed, and in the second stage, Bt is cultivated in the same bioreactor using the lysed yeasts as culture medium (supplemented with a feeding pulse of 10 g L?1 glycerol). This system was called fed batch one pot (FOP). A new inoculation strategy is also presented in this study, since these Bt cultures were inoculated directly with heat pre-treated spores instead of vegetative bacteria to facilitate the bioprocess. This study was developed from the laboratory to production-scale bioreactors (measuring from 500 mL to 2500 L), and the efficiency of the proposed strategy was evident in LD50 tests results, achieving 1796 ITU in large-scale processes. Both the use of non-conventional sources and the process development for biomass production are important for cost-effective production of Bt-based insecticides in mosquito control projects. 相似文献

Elevated concentrations of ground-level ozone (O3) are frequently measured over farmland regions in many parts of the world. While numerous experimental studies show that O3 can significantly decrease crop productivity, independent verifications of yield losses at current ambient O3 concentrations in rural locations are sparse. In this study, soybean crop yield data during a 5-year period over the Midwest of the United States were combined with ground and satellite O3 measurements to provide evidence that yield losses on the order of 10% could be estimated through the use of a multiple linear regression model. Yield loss trends based on both conventional ground-based instrumentation and satellite-derived tropospheric O3 measurements were statistically significant and were consistent with results obtained from open-top chamber experiments and an open-air experimental facility (SoyFACE, Soybean Free Air Concentration Enrichment) in central Illinois. Our analysis suggests that such losses are a relatively new phenomenon due to the increase in background tropospheric O3 levels over recent decades. Extrapolation of these findings supports previous studies that estimate the global economic loss to the farming community of more than $10 billion annually. 相似文献

Although organizational identification is founded on social identity and symbolic interactionist theories, current theories emphasize a social identity whereby organizational members categorize themselves and others based on roles and membership in an organization or work unit. In contrast symbolic interactionism, which resides in interpersonal relationships, is rarely theorized or empirically assessed in studies of organizational identification. We use survey data collected at an academic institution to explore how the strength and structure of an individual's social network both directly influences organizational identification as well as moderates the relation between social identity, or categorical, antecedents and organizational identification. Our results show that the size of an individual's network as well as the interaction between relationship strength and prestige better explain organizational identification than do antecedents based solely on categorization and social comparison processes. Thus networks of relationships, which have been a foundational but much neglected premise and process for organizational identification, are brought back into a theory of organizational identification. Cuticular hydrocarbons (CHCs) are information-rich signals in social insects that coordinate behaviors within nests. However,
in some taxa, the precise information conveyed by CHCs is poorly understood. In particular, there is a debate over whether
CHCs convey information about their bearer's dominance or fertility. Distinguishing between dominance and fertility signaling
is difficult because fertility and rank are frequently correlated within social insect colonies. This study disentangles those
relationships by examining CHCs of Polistes dominulus paper wasps during the early nest-founding stage before dominance and fertility become correlated. First, we confirm that
dominance and fertility are not associated in early spring foundresses. Then we show that CHCs are more strongly associated
with fertility than dominance. There was no relationship between cuticular hydrocarbon profiles and a wasp’s ability to dominate
rivals through aggression, suggesting that CHCs are unlikely to provide information about dominance. However, there was a
significant correlation between ovarian development and the CHC profile, suggesting that CHCs could convey valuable information
about their bearer's fertility. Furthermore, our data provide a potential mechanism for chemical signaling of fecundity, as
there is a relationship between endogenous juvenile hormone titer (a gonadotropin), degree of ovarian development, and the
CHC profile. Hormonal regulation of CHC profile expression offers a physiological mechanism to coordinate behavior, physical

The success of a recycling programme depends on the active and sustained participation of citizens in the correct separation and collection of recyclable waste. An effective study of strategies aimed at augmenting people's involvement in recycling involves understanding which factors influence the decision to co-operate with a recycling programme. This research investigates the influence of attitudes, incentives, presence of children in household and information through direct media, on households' participation in recycling. The results suggest that positive attitudes toward recycling and information are important factors in explaining recycling participation. Some guidelines that may be considered in future communication and intervention strategies designed to promote recycling participation are discussed. 相似文献

Ranked set sampling was developed for situations where measurement cost is expensive compared with unit acquisition. This
paper presents results of simulations and theory examining the impact of balanced ranked set sampling on the relative efficiencies
of the slope and intercept estimators of an ordinary least squares regression. Perfect ranking of either the independent or
the dependent variable is assumed throughout. In contradistinction to most of the published ranked set sampling work, it is
demonstrated that balanced ranked set sampling offers at most little improvement in the relative efficiencies of the slope

The Journal of Safety Research has partnered with the Injury Center at the Centers for Disease Control and Prevention (CDC) in Atlanta, Georgia, USA, to briefly report on some of the latest findings in the research community. This report is the sixth in a series of CDC articles. This conference on Hip Protector Use was held at Boston University in June, 2004 under the sponsorship of the Injury Center. Hip fractures from falls are a major threat to the health and well-being of older adults. One approach to hip fracture prevention is the use of hip protectors (HPs), soft padding or hard shells embedded within specially designed underwear. This conference brought together experts to discuss the efficacy and barriers to using HP. Conclusions from the conference included the need for HP product specifications and standards of safety and efficacy, testing protocols, and efficacy trials. While no single entity was identified to advance these initiatives, this effort should be international and should involve collaboration between researchers, health care providers and policy-makers, and HP manufacturers. Plans are already underway for a second international conference in Europe to follow-up these recommendations and develop strategies for implementation. 相似文献

Eutrophication and predator additions and extinctions are occurring in ecosystems worldwide. Although theory predicts that both will strongly alter the distribution of biomass in whole communities, empirical evidence has not been consolidated to quantitatively determine whether these theoretical predictions are generally borne out in real ecosystems. Here we analyze data from two types of trophic cascade studies, predator removals in factorial combination with fertilization and observed productivity gradients, to assess the role of top-down and bottom-up forces in structuring multi-trophic communities and compare results from these analyses to those from an extensive database of trophic cascade studies. We find that herbivore biomass declines and plant biomass increases in the presence of predators, regardless of system productivity. In contrast, while plants are increased by fertilization, this effect does not significantly increase herbivores in either the presence or absence of predators. These patterns are consistent among marine, freshwater, and terrestrial ecosystems and are largely independent of study size and duration. Thus, top-down effects of predation are transferred through more trophic levels than are bottom-up effects of eutrophication, showing strong asymmetry in the direction of control of biomass distribution in communities. 相似文献

Millions of people die every year around the world from diarrheal diseases much of which is caused by contaminated drinking water. By contrast, drinking water safety is largely taken for granted by many citizens of affluent nations. The ability to drink water that is delivered into households without fear of becoming ill may be one of the key defining characteristics of developed nations in relation to the majority of the world. Yet there is well-documented evidence that disease outbreaks remain a risk that could be better managed and prevented even in affluent nations. A detailed retrospective analysis of more than 70 case studies of disease outbreaks in 15 affluent nations over the past 30 years provides the basis for much of our discussion [Hrudey, S.E. and Hrudey, E.J. Safe Drinking Water--Lessons from Recent Outbreaks in Affluent Nations. London, UK: IWA Publishing; 2004.]. The insights provided can assist in developing a better understanding within the water industry of the causes of drinking water disease outbreaks, so that more effective preventive measures can be adopted by water systems that are vulnerable. Environmental indicators are often aggregated into a single index for various purposes in environmental studies. Aggregated indices derived from the same data set can differ, usually because the aggregated indices' sensitivities are not thoroughly analyzed. Furthermore, if a sensitivity analysis is carried out, it is not presented in a transparent fashion to policy decision-makers. This paper presents a method of generating various aggregated environmental indices and analyzing their sensitivities via the use of the fuzzy set concept. Results show that several insights into the environmental conditions of the study area (e.g., the distribution of good or bad values of indicators at a watershed and or across the region) can be revealed in the sensitivity analysis of aggregated indices. 相似文献
