Environmental exposure to silver (Ag) was assessed in occupationally non-exposed adult human population by analyzing Ag in the hair (H?·?Ag) and whole blood (WB?·?Ag). H?·?Ag was analyzed in 311 (123 men, M; 188 women, W); while WB?·?Ag was determined in 235 of these individuals (90 M, 145 W). Women had more H?·?Ag than men (M 0.05 vs. W 0.076), whereas WB?·?Ag concentrations in men and women were not significantly different. A natural distribution of the median derivatives was utilized to generate the dataset to fit the logistic sigmoid curve to assess the current human body burden of environmental Ag population exposure for M and W separately. The H?·?Ag (µg?g?1) below 0.0105 for M and 0.0145 for W, reflects low level of environmental Ag exposure. The adaptive physiological saturation phase followed where H?·?Ag rose rapidly, first for M and then for W in parallel with biological assay. Both parallel saturation curves converged and plateaued at 0.215 for M and 0.965 for W (µg?g?1). The current level of human environmental Ag exposure is low, but cases of high Ag exposure occurred sporadically. In conjunction with the medical histories overt clinical neural toxicity may be expected for H?·?Ag at 4?µg?g?1and higher. There were no significant correlation between the H?·?Ag and WB?·?Ag.  相似文献

A multimedia sampling of ambient air, wet deposition, surface water, sediment, soil and biota has been performed at Kosetice background observatory in the southern Czech Republic since 1988. An integrated monitoring approach was applied to assess the current state, anthropogenic impacts, and possible future changes of terrestrial and freshwater environments. Average PCB concentrations in the individual matrices calculated from ten years of sampling on multiple sites varied between 2 ng g(-1) in sediment and 7 ng g(-1) in soil or moss. DDT concentrations were lower in moss and needles (2 ng g(-1) and 4 ng g(-1), respectively) than in sediment (11 ng g(-1)) and soil (20 ng g(-1)), while the HCH level was higher in moss and needles (5 ng g(-1) and 6 ng g(-1), respectively) than in soil or sediment (1 ng g(-1) and 2 ng g(-1), respectively). The highest average level of PAHs was found in soil (600 ng g(-1)), while it was lower in needles (230 ng g(-1)), moss (210 ng g(-1)) or sediment (210 ng g(-1)). Time related trends of concentration levels of persistent organic pollutants in all matrices were investigated. Moss and needle trend patterns resembled those of the ambient air, showing a slight concentration decrease of all compounds, except for hexachlorobenzene. The soil, water and sediment concentrations showed a similar decrease of PAHs, PCBs, and HCHs, but there was no clear trend for DDTs and HCB.  相似文献

The aim of this study is to test the applicability of snow surveying in the collection and detailed characterization of vehicle-derived magnetic particles. Road dust extracted from snow, collected near a busy urban highway and a low traffic road in a rural environment (southern Finland), was studied using magnetic, geochemical and micro-morphological analyses. Significant differences in horizontal distribution of mass specific magnetic susceptibility (χ) were noticed for both roads. Multi-domain (MD) magnetite was identified as the primary magnetic mineral. Scanning electron microscope (SEM) analyses of road dust from both roads revealed: (1) angular-shaped particles (diameter ∼1-300 μm) mostly composed of Fe, Cr and Ni, derived from circulation of motor vehicles and (2) iron-rich spherules (d ∼ 2-70 μm). Tungsten-rich particles (d < 2 μm), derived from tyre stud abrasion were also identified. Additionally, a decreasing trend in χ and selected trace elements was observed with increasing distance from the road edge.  相似文献

Chlorpyrifos degradation in Turkish soil   总被引:12,自引:0,他引:12  
Degradation of chlorpyrifos was evaluated in laboratory studies. Surface (0-15 cm) and subsurface (40-60 cm) clay loam soils from a pesticide-untreated field were incubated in biometer flasks for 97 days at 25 degrees C. The treatment was 2 micrograms g-1 [2,6-pyridinyl-14C] chlorpyrifos, with 74 kBq radioactivity per 100 g soil flask. Evolved 14CO2 was monitored in KOH traps throughout the experiment. Periodically, soil subsamples were also methanol-extracted [ambient shaking, then supercritical fluid extraction (SFE)], then analyzed by thin-layer chromatography. Total 14C and unextractable soil-bound 14C residues were determined by combustion. From the surface and subsurface soils, 41 and 43% of the applied radiocarbon was evolved as 14CO2 during 3 months incubation. The time required for 50% loss of the parent insecticide in surface and subsurface soils was about 10 days. By 97 days, chlorpyrifos residues and their relative concentration (in surface/subsurface) as % of applied 14C were: 14CO2 (40.6/42.6), chlorpyrifos (13.1/12.4), soil-bound residues (11.7/11.4), and 3,5,6-trichloropyridinol (TCP) (3.8/4.8). Chlorpyrifos was largely extracted by simple shaking with methanol, whereas TCP was mainly removed only by SFE. The short persistence of chlorpyrifos probably relates to the high soil pH (7.9-8.1).  相似文献

Fine spatial and temporal phytoplankton variability in Mali Ston Bay has been observed for the first time based on physicochemical properties and small herbivorous zooplankton. Extensive year-through research was conducted during 2002 at Usko station which is traditionally an area of intensive shellfish farming. The Neretva River inflow, submarine springs (“vruljas”) and precipitation are additional sources of nutrients in the bay. Temperature and salinity, combined with total inorganic nitrogen (TIN) were observed to be the most important environmental factors driving the succession of phytoplankton communities. Orthophosphate was a potential limiting factor for phytoplankton development. The nanophytoplankton abundances, as well as the microphytoplankton diatoms are controlled by herbivorous zooplankton grazing (‘top-down’ control) more than other groups of microphytoplankton. Nanophytoplankton dominated phytoplankton abundance and its most intensive development was recorded in winter and spring, while increase in microphytoplankton abundance occurred in spring and autumn. Diatoms dominated microphytoplankton abundance mostly in winter and autumn, while dinoflagellates dominated in spring and summer. Considering the number of taxa and abundance, dinoflagellates were the dominant microphytoplankton group during the year and were the main component of the spring blooms. At that time, in conditions of elevated temperature (>16 °C), decreased salinity (34–36) and increased concentrations of TIN, blooms of harmful dinoflagellate Prorocentrum minimum were recorded for the first time in the bay. The results showed a significant difference in environmental conditions, as well as in the annual phytoplankton succession and community structure, as compared with studies carried out more than 20 years ago in this area.  相似文献

Mass balance of contaminants can provide useful information on the processes that influence their concentrations in various environmental compartments. The most important sources, sinks and the equilibrium or non-equilibrium state of the contaminant in individual environmental compartments can also be identified. Using the latest mercury speciation data, the results of numerical models and the results of recent studies on mercury transport and transformation processes in the marine environment, we have re-evaluated the total mercury (HgT) mass balance in the Mediterranean Sea. New calculations have been performed employing three distinct marine layers: the surface layer, the thermocline and the deep sea. New transport mechanisms, deep water formation and density-driven sinking and upwelling, were included in the mass balance calculations. The most recent data have even enabled the calculation of an approximate methylmercury (MeHg) mass balance. HgT is well balanced in the entire Mediterranean, and the discrepancies between inputs and outputs in individual layers do not exceed 20 %. The MeHg balance shows larger discrepancies between gains and losses due to measurement uncertainties and gaps in our knowledge of Hg species transformation processes. Nonetheless, the main sources and sinks of HgT (deposition and evasion) and MeHg (fluxes from sediment, outflow through the Gibraltar Strait) are in accordance with previous studies on mercury in the Mediterranean Basin. Mercury in the Mediterranean fish harvest is the second largest MeHg sink; about 300 kg of this toxic substance is consumed annually with sea food.  相似文献

Objective

Recent studies have integrated copy number variant (CNV) and gene analysis using target enrichment. Here, we transferred this concept to our routine genetics laboratory, which is not linked to centralized non-invasive prenatal testing (NIPT) facilities.

Method

From a cohort of 100 pregnant women, 22 were selected for the analysis of maternal genomic DNA (gDNA) along with fetal cell-free DNA. Using targeted enrichment, 135 genes were analyzed, combined with aberrations of chromosomes 21, 18, 13, X, and Y. The data were subjected to specificity and sensitivity analyses, and correlated with the results from invasive testing methods.

Results

The sensitivity/specificity was determined for the CNV analysis of chromosomes: 21 (80%/75%), 18 (-/82%), 13 (100%/67%), and Y (100%/100%). The gene detection was valid for maternal gDNA. However, for cell-free fetal DNA, it was not possible to determine the boundary between an artifact and a real sequence variant.

Conclusion

The target enrichment method combining CNV and gene detection seems feasible in a regular laboratory. However, this method can only be responsibly optimized with a sufficient number of controls and further validation on a strong bioinformatic background. The present results showed that NIPT should be performed in specialized centers, and that its introduction to isolated laboratories may not provide valid data.  相似文献

Severe fluid forces are believed to be a source of injury and mortality to fish that pass through hydroelectric turbines. A process is described by which laboratory bioassays, computational fluid dynamics models, and field studies can be integrated to evaluate the significance of fluid shear stresses that occur in a turbine. Areas containing potentially lethal shear stresses were identified near the stay vanes and wicket gates, runner, and in the draft tube of a large Kaplan turbine. However, under typical operating conditions, computational models estimated that these dangerous areas comprise less than 2% of the flow path through the modeled turbine. The predicted volumes of the damaging shear stress zones did not correlate well with observed fish mortality at a field installation of this turbine, which ranged from less than 1% to nearly 12%. Possible reasons for the poor correlation are discussed. Computational modeling is necessary to develop an understanding of the role of particular fish injury mechanisms, to compare their effects with those of other sources of injury, and to minimize the trial and error previously needed to mitigate those effects. The process we describe is being used to modify the design of hydroelectric turbines to improve fish passage survival.  相似文献
