Recognizing the importance of sacred groves in biodiversity conservation and management, and the recent threats to them, the
present study was conducted in Shivbari sacred grove of Himachal Pradesh. The study aimed at documenting the floral wealth
of Shivbari and promoting plantation of indigenous species in participation with local people. For this, systematic field
surveys in different seasons were conducted in Shivbari from April 2005 to November 2009, and liaison was maintained with
the local community and temple management authority for gaining insight into the history and problems of Shivbari and initiating
plantation activities. A total of 69 flowering plant species were identified inside the grove, which include 14 trees, 9 shrubs,
3 lianas and 43 herbs. This represents almost 2% of the total flowering plant species occurring in the state of Himachal Pradesh.
Mallotus philippensis followed by Putranjiva roxburghii was the most dominant tree species. Adhatoda
zeylanica was the most common shrub species, while Achyranthes aspera was the most common herb species. The grove harbours 23 plants species that are in high demand in the market, and at the
same time also influences the microclimate of the area. The temperature inside the grove was significantly lower than the
temperature outside the grove. The recent changes in socio-economic status of the local people and a shift towards market-oriented
economy have threatened the survival of Shivbari. However, the deeply held beliefs of the pilgrims, local people and priest

Bacterial strains with poly-3-hydroxybutyrate (PHB) degradation potential were isolated from waste yard soil samples of selected industrial sites in Uttarakhand, India, and two microbial consortia were developed, i.e. Consortium I comprises Pseudomonas sp. strain Rb10, Pseudomonas sp. strain Rb11 and Bacillus sp. strain Rb18, and Consortium II is composed of Lysinibacillus sp. strain Rb1, Pseudomonas sp. strain Rb13 and Pseudomonas sp. strain Rb19. The current study involved enrichment selection via liquid and semi-solid media, followed by isolation and screening of bacterial strains using PHB pellets and films. Furthermore, the identification and characterisation was done by triphasic approach. The utilisation of PHB by the characterised strains was confirmed by Fourier transform infrared spectroscopy and scanning electron microscopy. Moreover, the minimum inhibitory concentration of solubilised PHB was found to be 2.5?mg/mL, which was detected through ‘clear zone assay’. Further, the selection and biocompatibility testing of potential isolates were performed for the formation of bacterial consortia. Thus, the present work would provide direct and standardised protocol for screening and selection of potential microbiomes for biodegradation of polymers by overcoming the negative effect of organic solvents. The present study was undertaken to evaluate biochemical markers of chronic carbofuran exposure to rats in terms of lipid peroxide and intrasynaptosomal calcium levels and to correlate them with the histopathological changes in brain regions. A significant increase in lipid peroxidation (LPO) in terms of thiobarbituric acid reactive substances (TBARS) was observed in the cerebral cortex (65%) and brain stem (33%) after carbofuran exposure. This was accompanied by a significant increase (87%) in the intracellular free-Ca2+ [Ca2+]i levels. N-acetylcysteine (NAC) administration, on the other hand, reversed the carbofuran-induced increase in LPO and [Ca2+]i. Histopathological studies of carbofuran-exposed brain revealed high frequency of pyknotic neurons in the cerebral cortex and microhaemorrhages in the brain stem. NAC supplementation to carbofuran-treated animals resulted in normalisation of the brain architecture as seen by a reduction in the number of pyknotic nuclei in the cerebral cortex. These findings indicate that increased LPO and elevated [Ca2+]i levels are involved in the development of carbofuran neurotoxicity and are eventually responsible for the pathological alterations. Soil identification and chemometrics for direct determination of nitrate in soils using FTIR-ATR mid-infrared spectroscopy 总被引:2,自引:0,他引:2
The use of mid-infrared attenuated total reflectance (ATR) spectroscopy enables direct measurement of nitrate concentration in soil pastes, but strong interfering absorbance bands due to water and soil constituents limit the accuracy of straightforward determination. Accurate subtraction of the water spectrum improves the correlation between nitrate concentration and its nu3 vibration band around 1350 cm(-1). However, this correlation is soil-dependent, due mostly to varying contents of carbonate, whose absorbance band overlaps the nitrate band. In the present work, a two-stage method is developed: First, the soil type is identified by comparing the "fingerprint" region of the spectrum (800-1200 cm(-1)) to a reference spectral library. In the second stage, nitrate concentration is estimated using the spectrum interval that includes the nitrate band, together with the soil type previously identified. Three methods are compared for estimating nitrate concentration: integration of the nitrate absorbance band, cross-correlation with a reference spectrum, and principal component analysis (PCA) followed by a neural network. When using simple band integration, the use of soil specific calibration curves leads to determination errors ranging from 5.5 to 24 mg[N]/kg[dry soil] for the mineral soils tested. The cross-correlation technique leads to similar results. The combination of soil identification with PCA and neural network modeling improves the predictions, especially for soils containing calcium carbonate. Typical prediction errors for light non-calcareous soils are about 4 mg[N]/kg[dry soil], whereas for soils containing calcium carbonate they range from 6 to 20 mg[N]/kg[dry soil], which is less than four percent of the concentration range investigated. 相似文献
