Objective

The effects of mechanical stimulation in preterm amniotic membrane (AM) defects were explored.

Methods

Preterm AM was collected from women undergoing planned preterm caesarean section (CS) due to fetal growth restriction or emergency CS after spontaneous preterm prelabour rupture of the membranes (sPPROM). AM explants near the cervix or placenta were subjected to trauma and/or mechanical stimulation with the Cx43 antisense. Markers for nuclear morphology (DAPI), myofibroblasts (αSMA), migration (Cx43), inflammation (PGE2) and repair (collagen, elastin and transforming growth factor β [TGFβ1]) were examined by confocal microscopy, second harmonic generation, qPCR and biochemical assays.

Results

In preterm AM defects, myofibroblast nuclei were highly deformed and contractile and expressed αSMA and Cx43. Mechanical stimulation increased collagen fibre polarisation and the effects on matrix markers were dependent on tissue region, disease state, gestational age and the number of fetuses. PGE2 levels were broadly similar but reduced after co-treatment with Cx43 antisense in late sPPROM AM defects. TGFβ1 and Cx43 gene expression were significantly increased after trauma and mechanical stimulation but this response dependent on gestational age.

Conclusion

Mechanical stimulation affects Cx43 signalling and cell/collagen mechanics in preterm AM defects. Establishing how Cx43 regulates mechanosignalling could be an approach to repair tissue integrity after trauma.  相似文献

Objective

Magnetic resonance imaging (MRI) of placental invasion has been part of clinical practice for many years. The possibility of being better able to assess placental vascularization and function using MRI has multiple potential applications. This review summarises up-to-date research on placental function using different MRI modalities.

Method

We discuss how combinations of these MRI techniques have much to contribute to fetal conditions amenable for therapy such as singletons at high risk for fetal growth restriction (FGR) and monochorionic twin pregnancies for planning surgery and counselling for selective growth restriction and transfusion conditions.

Results

The whole placenta can easily be visualized on MRI, with a clear boundary against the amniotic fluid, and a less clear placental-uterine boundary. Contrasts such as diffusion weighted imaging, relaxometry, blood oxygenation level dependent MRI and flow and metabolite measurement by dynamic contrast enhanced MRI, arterial spin labeling, or spectroscopic techniques are contributing to our wider understanding of placental function.

Conclusion

The future of placental MRI is exciting, with the increasing availability of multiple contrasts and new models that will boost the capability of MRI to measure oxygen saturation and placental exchange, enabling examination of placental function in complicated pregnancies.  相似文献

Per-and polyfluoroalkyl substances(PFASs) are ubiquitous in sludge and water from waste water treatment plants, as a result of their incorporation in everyday products and industrial processes. In this study, we measured several classes of persistent PFASs,precursors, transformation intermediates, and newly identified PFASs in influent and effluent sewage water and sludge from three municipal waste water treatment plants in Sweden, sampled in 2015. For sludge, samples from 2012 and 2014 were analyzed as well.Levels of precursors in sludge exceeded those of perfluoroalkyl acids and sulfonic acids(PFCAs and PFSAs), in 2015 the sum of polyfluoroalkyl phosphoric acid esters(PAPs) were 15–20 ng/g dry weight, the sum of fluorotelomer sulfonic acids(FTSAs) was 0.8–1.3 ng/g,and the sum of perfluorooctane sulfonamides and ethanols ranged from non-detected to 3.2 ng/g. Persistent PFSAs and PFCAs were detected at 1.9–3.9 ng/g and 2.4–7.3 ng/g dry weight, respectively. The influence of precursor compounds was further demonstrated by an observed substantial increase for a majority of the persistent PFCAs and PFSAs in water after waste water treatment. Perfluorohexanoic acid(PFHxA), perfluorooctanoic acid(PFOA), perfluorohexane sulfonic acid(PFHxS), and perfluorooctane sulfonic acid(PFOS)had a net mass increase in all WWTPs, with mean values of 83%, 28%, 37% and 58%,respectively. The load of precursors and intermediates in influent water and sludge combined with net mass increase support the hypothesis that degradation of precursor compounds is a significant contributor to PFAS contamination in the environment.  相似文献

The effects of NiCl2 were studied in two human cell lines, HeLa and diploid embryonic fibroblasts as well as in V79 Chinese hamster cells and in L‐A mouse fibroblasts. NiCl2 produces a dose‐dependent depression of proliferation, mitotic rate, and viability, accompanied by an increasing release of lactic dehydrogenase and stimulation of lactic acid production. The plating efficiency is reduced, as are DNA and protein synthesis and, to a lesser degree, RNA synthesis.

The cytotoxicity of NiCl2 is comparable in degree to those of PbCl2 and MnCl2, but is weaker than those of HgCl2 and CdCl2. However, the different sensitivities of different cell lines must also be considered.

NiCl2 effects are more severe in serum‐free medium than in medium containing serum or serum albumin indicating that serum constituents, notably albumin, bind the metal effectively and inhibit cellular uptake; this confirms earlier reports on the serum binding and slow uptake of NiCl2.

Synchronized cells are most sensitive in the Gl and early S phases of the cell cycle. In the Painter test the depression of DNA synthesis persists following cessation of exposure to NiCI2. These findings contribute an explanation for the known genotoxic effects of nickel.  相似文献

ABSTRACT

Very important in adsorption processes is the preparation of material sorbents and from this reason, the impact of washing agent of raw sorbent was analysed. The adsorption capacity was studied in relation to the function of the equilibrium time, the amount of biosorbent and the initial metal(II) ions concentration. In this research for the sake of comparison, the sorptive properties of out-of-date coffee were compared to coffee washed with different agents (mineral acids and water). The scanning electron microscopy and Fourier transform infrared spectroscopy were used to characterise the biosorbents and to identify the functional groups that participated in metal(II) ions bonding. The obtained results clearly indicate that the out-of-date coffee are effective biosorbent for cadmium(II) and nickel(II) ions from aqueous solution. However, for metal(II) ions removal efficiency could influence sorbent preparation and nickel(II) ions were best adsorbed on coffee washed with water, but cadmium(II) ions on raw biosorbent (out-of-date coffee). The best results of sorption were achieved after the contact time of 30 and 60?min, respectively for cadmium(II) and nickel(II) ions.  相似文献

This paper focuses on comparative review and analysis of the systems of environmental protection, national environmental strategies and fundamentals of environmental legislation in Australia and the Russian Federation. These countries have many socioeconomic similarities, such as low population densities and richness in natural resources which are largely exported. The main disparity between Australia and Russia is in the types of economy. This paper ‘continues’ the sequence of recent scholarly publications on comparison of various features of environmental policies in the countries from all over the world. Comparative analysis in this research has been based on examination of scholarly publications, legislation, government documents, mass media sources and NGO responses. The analysis has revealed a number of differences including top-down implementation of policies in Russia whereas in Australia, each State (Territory) implements policies with significant independence from the Commonwealth Government. At the same time, similarities between the countries have been identified: for example, in deficits in the budget of local environmental authorities and in the presence of contradictions in legislation at national and regional levels of government. Suggestions resulting from this analysis include further integration of sustainable development strategies at all levels of Australian government to encourage further protection of the environment and, for Russia, creating a separate Ministry of Environmental Protection. These approaches should assist facilitation of sustainable development for both nations. Large-scale studies are essential to assess the emission patterns and spatial distribution of organohalogenated pollutants (OHPs) in the environment. Bird eggs have several advantages compared to other environmental media which have previously been used to map the distribution of OHPs. In this study, large-scale geographical variation in the occurrence of OHPs, such as polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and organochlorine pesticides (OCPs), was investigated throughout Europe using eggs of a terrestrial residential passerine species, the great tit (Parus major). Great tit eggs from 22 sampling sites, involving urban, rural and remote areas, in 14 European countries were collected and analysed (5-8 eggs per sampling site). The environmentally most important congeners/compounds of the analysed pollutants were detectable in all sampling locations. For PCBs, PBDEs and OCPs, no clear geographical contamination pattern was found. Sum PCB levels ranged from 143 ng/g lipid weight (lw) to 3660 ng/g lw. As expected, PCB concentrations were significantly higher in the sampled urban compared to the remote locations. However, the urban locations did not show significantly higher concentrations compared to the rural locations. Sum PBDEs ranged from 4.0 ng/g lw to 136 ng/g lw. PBDEs were significantly higher in the urbanized sampling locations compared to the other locations. The significant, positive correlation between PCB and PBDE concentrations suggests similar spatial exposure and/or mechanisms of accumulation. Significantly higher levels of OCPs (sum OCPs ranging from 191 ng/g lw to 7830 ng/g lw) were detected in rural sampling locations. Contamination profiles of PCBs, PBDEs and OCPs differed also among the sampling locations, which may be due to local usage and contamination sources. The higher variance among sampling locations for the PCBs and OCPs, suggests that local contamination sources are more important for the PCBs and OCPs compared to the PBDEs. To our knowledge, this is the first study in which bird eggs were used as a monitoring tool for OHPs on such a large geographical scale.  相似文献

PCB levels in fish (collected from local rivers), atmosphere and human milk samples have been studied to determine the exposure levels of PCBs for local residents and e-waste workers in Guiyu, a major electronic waste scrapping center in China. The source appointment and correlation analyses showed that homologue composition of PCBs in 7 species of fish were consistent and similar to commercial PCBs Aroclor 1248. PCB levels in air surrounding the open burning site were significantly higher than those in residential area. Inhalation exposure contributed 27% and 93% to the total body loadings (the sum of dietary and inhalation exposure) of the local residents, and e-waste workers engaged in open burning respectively. Total PCB concentrations in human milk ranged from N.D. to 57.6 ng/g lipid, with an average of 9.50 ng/g lipid. The present results indicated that commercial PCBs derived from e-waste recycling are major sources of PCBs accumulating in different environmental media, leading to the accumulation of high chlorinated biphenyls in human beings.  相似文献

The accumulations of both natural (U) and depleted (DU) uranium in the earthworms (Eisenia fetida) were studied to evaluate corresponding biological effects. Concentrations of metals in the experimental soil ranged from 1.86 to 600 mg kg−1. Five biological endpoints: mortality, animals’ weight increasing, lysosomal membrane stability by measuring the neutral red retention time (the NRRT), histological changes and genetic effects (Comet assay) were used to evaluate biological effects in the earthworms after 7 and 28 days of exposure. No effects have been observed in terms of mortality or weight reduction. Cytotoxic and genetic effects were identified at quite low U concentrations. For some of these endpoints, in particular for genetic effects, the dose (U concentration)-effect relationships have been found to be non-linear. The results have also shown a statistically significant higher level of impact on the earthworms exposed to natural U compared to depleted U.  相似文献
