Dissolved organic matter (DOM) is a ubiquitous constituent of natural waters and is comprised of a variety of chemically heterogeneous molecular structures and functional groups. DOM is often considered to be a major ligand for metals in most natural waters and its reactivity is thought to be strongly dependent on its chemical composition and structure. In this study, a combination of UV/visible, emission excitation matrix fluorescence (EEM) and 1H NMR spectroscopies were used to characterize DOM from the Athabasca River (Alberta, Canada). The chemical characterization of river DOM showed that the most upstream samples located in agricultural areas were blue-shifted and less aromatic and contained more hydrogens connected with oxygen functional groups than those in the wetland dominated area in the Athabasca oil sand deposit region. The presence of paramagnetic ions (Fe and Al) was not found to significantly affect the structural composition of DOM as revealed by 1H NMR. Such change in the quality of DOM may have a profound impact on metal binding in the Athabasca River watershed.  相似文献

Feed additives can change the microbiological environment of the animal digestive track, nutrient composition of feces, and its gaseous emissions. This 2-yr field study involving commercial laying-hen houses in central Iowa was conducted to assess the effects of feeding diets containing EcoCal and corn-dried distillers grain with solubles (DDGS) on ammonia (NH3), hydrogen sulfide (H2S), and greenhouse gas (CO2, CH4, and N2O) emissions. Three high-rise layer houses (256,600 W-36 hens per house) received standard industry diet (Control), a diet containing 7% EcoCal (EcoCal) or a diet containing 10% DDGS (DDGS). Gaseous emissions were continuously monitored during the period of December 2007 to December 2009, covering the full production cycle. The 24-month test results revealed that mean NH3 emission rates were 0.58 +/- 0.05, 0.82 +/- 0.04, and 0.96 +/- 0.05 g/hen/day for the EcoCal, DDGS, and Control diet, respectively. Namely, compared to the Control diet, the EcoCal and DDGS diets reduced NH3 emission by an average of 39.2% and 14.3%, respectively. The concurrent H2S emission rates were 5.39 +/- 0.46, 1.91 +/- 0.13, and 1.79 +/- 0.16 mg/ hen/day for the EcoCal, DDGS, and Control diet, respectively. CO2 emission rates were similar for the three diets, 87.3 +/- 1.37, 87.4 +/- 1.26, and 89.6 +/- 1.6 g/hen/day for EcoCal, DDGS, and Control, respectively (P = 0.45). The DDGS and EcoCal houses tended to emit less CH4 than the Control house (0.16 and 0.12 vs. 0.20 g/hen/day) during the monitored summer season. The efficacy of NH3 emission reduction by the EcoCal diet decreased with increasing outside temperature, varying from 72.2% in February 2009 to -7.10% in September 2008. Manure of the EcoCal diet contained 68% higher ammonia nitrogen (NH3-N) and 4.7 times higher sulfur content than that of the Control diet. Manure pH values were 8.0, 8.9, and 9.3 for EcoCal, DDGS, and Control diets, respectively. This extensive field study verifies that dietary manipulation provides a viable means to reduce NH3 emissions from modern laying-hen houses.  相似文献

Over three million dry metric tons of biosolids produced in the United States are land applied as Class B. Lime stabilization is employed for biosolids treatment at approximately 20% of the wastewater treatment plants because it is a simple and inexpensive process. During lime stabilization, the pH of sewage sludge is raised above 12 for pathogen inactivation and odor reduction. Lime dose and mixing have been found to greatly reduce odor generation from lime stabilized biosolids. A better quality biosolids product is less likely to create public opposition to land application programs. In this study, land application tests using Class B biosolids were conducted in order to determine whether better mixing can reduce odor generation during the land application of lime stabilized biosolids. The mixing quality of a treatment plant’s lime stabilized biosolids was improved by relocating the lime addition point, which prolonged the mixing time and produced a better mixed biosolids product. Based on field observations of land application, the poorly mixed biosolids were more odorous and offensive prior to incorporation. However, once incorporated into the soil, there was no appreciable odor difference between the biosolids. Another land application study was conducted to assess the odor of unincorporated Class A biosolids and compare it with incorporated Class A biosolids with the soil.  相似文献

In situ measurements of community respiration were made at two stations in Castle Harbor, Bermuda, during April and May, 1971. Total community respiration was 20.67 and 19.11 ml O2 m-2 h-1 at Stations 1 and 2, respectively, in April. In May, respiration increased with water temperature to 26.99 and 24.56 ml O2 m-2 h-1. Significant differences (P<0.05) existed between stations and sampling periods. Bacterial respiration was estimated from sediment treatment with streptomycin-SO4. Values ranged from 7.71 to 8.72 ml O2 m-2 h-1 in April and May, respectively. No significant difference existed between sampling periods or stations. No detectable chemical O2 demand of the sediment, determined by a formalin treatment, was found. Total community respiration was further compartmentalized into macrofaunal, meiofaunal, and microfaunal-microfloral components. Both the macrofaunal and meiofaunal respiration was negligible compared to that estimated for the microfaunal-microfloral component.Contribution No. 2708 from the Woods Hole Oceanographic Institution, and Contribution No. 552 from the Bermuda Biology Station. An Al(OH)x-montmorillonite (chlorite) complex (AM18) was prepared and 2,4-dichlorophenoxyacetic acid (2,4-D) sorbed to saturation. After several washing cycles the 'strongly sorbed' 2,4-D was 507 micrograms g-1 AM18. The bioavailability of sorbed 2,4-D was assessed in a minimal salts medium with the AM18-2,4-D as the sole C and energy source. Over a 28-day period a Pseudomonas sp. degraded 23% more of the sorbed 2,4-D than could be accounted for by desorption from AM18 in the non-inoculated controls. Possible explanations for the increase in bioavailability are presented.  相似文献

Human activities threaten the biodiversity of aquatic mammals across the globe. Conservation of these species hinges on the ability to delineate movements and foraging behaviors of animals, but gaining such insights is hampered by difficulties in tracing individuals over their lives. We determined isotope ratios in teeth (87Sr/86Sr, 13C/12C, and 18O/16O) to examine lifelong movement and resource-use patterns of a unique freshwater population of a wide-ranging pinniped species (harbor seal [Phoca vitulina]) that resides in Iliamna Lake, Alaska (U.S.A.). This population's potentially unique migratory behavior and use of different trophic resources are unknown. The isotope ratios we measured in teeth showed that seals were born in the lake, remained lifelong residents, and relied principally on resources produced from in the lake, even when seasonally abundant and nutrient-dense spawning anadromous fish (i.e., sockeye salmon [Oncorhynchus nerka]) were available in the lake. Our results illustrate how serial isotope records in teeth, particularly 87Sr/86Sr ratios, can be used to quantify how coastal mammal populations exploit both freshwater and marine ecosystems. Understanding lifelong patterns of habitat and resource use is essential information when designing effective conservation plans for threatened coastal mammals. We present the Iliamna Lake harbor seals as a unique case study into how isotope records within teeth can help reveal the cryptic ecology of such a population residing in an intact ecosystem. The results also provide critical baseline information for the Kvichak River system, which is facing an uncertain future due to proposed large-scale industrial development and a rapidly changing climate.  相似文献

 The ability to introduce and express foreign genes can greatly facilitate in vitro studies of the molecular biology of an organism. We report here the first use of modified retroviral vectors to achieve foreign gene expression in primary cultured cells from the Oka organ and ovary of the blue shrimp Penaeus stylirostris (Litopenaeus) stylirostris (Holthuis 1980). Of the heterologous promoters tested, the Moloney murine leukemia virus and Rous sarcoma virus promoters mediated the highest levels of reporter gene expression. Firefly luciferase and Escherichia coliβ-galactosidase were successfully used as reporter genes in cultured cells from both Oka organ and ovary. These in vitro systems can be exploited for the study of shrimp cellular biology and regulation of gene expression. Received: 16 January 2000 / Accepted: 21 June 2000  相似文献

This study examined the concentrations of total hydrocarbons (THC), polychlorinated biphenyls (PCB), polyaromatic hydrocarbons (PAH), and trace metals (Cu, Zn, Cd, Pb, Hg and As) in marine sediments off Scott Base (NZ) and compared them with sediments near the highly polluted McMurdo Station (US) as well as less impacted sites including Turtle Rock and Cape Evans. The Antarctic mollusc, Laternula elliptica and three common sponge species were also analysed for trace metals. The mean THC concentration in sediments from Scott Base was 3 fold higher than the pristine site, Turtle Rock, but 10 fold lower than samples from McMurdo Station. McMurdo Station sediments also contained the highest concentrations of PAHs, PCBs and the trace metals, Cu, Zn, Pb, Cd and Hg. Copper was significantly higher in bivalves from McMurdo Station than other sites. Trace metal concentrations in sponges were generally consistent within sites but no spatial patterns were apparent.  相似文献
