Behavioural ecologists attempt to predict fitness in birds from estimates of body condition and immune capacity. We investigated how the stresses associated with capture, confinement and captive-rearing of wild zebra finches (Taeniopygia guttata) affected different elements of the immune system and body condition. Wild birds had higher heterophil:lymphocyte ratios and total leucocyte counts than aviary birds, presumably an outcome of mounting specific resistance to pathogens, but this response diminished significantly within 10 days of confinement. Wild birds had lower phytohaemagglutinin-A (PHA) responses than their aviary-bred counterparts possibly because energetic costs limited a general resistance response. Wild birds were heavier and had higher haematocrits than their aviary counterparts, but had less fat, although just 10 days of captivity significantly increased fat levels. Measures of body condition were of limited use for predicting immune responsiveness. We conclude that the different elements of the immune system and body condition respond independently, and often unpredictably, to many ecological and behavioural stressors.  相似文献

Mosaicism for the Wolf-Hirschhorn syndrome, del(4)(p16), is extremely rare and has not been reported in association with a numerical chromosome abnormality. We report the prenatal diagnosis of mosaic del(4)(p16) and non-mosaic trisomy 21 in a 16-week female fetus. The pregnancy ended in spontaneous abortion at 34 weeks secondary to fetal demise. Forty-two open neural tube defects (NTDs) were identified in our series of 7440 amniocenteses tested between 11 and 15 weeks of gestation. Using a cut-off of ≥2.0 MOM, the detection rate for open NTDs was 95 per cent; 100 per cent each for anencephaly and spina bifida; and 78 per cent for encephalocele. Two encephaloceles had AFP levels less than 2.0 MOM and negative AChEs. Thirty-four (81 per cent) of these NTDs were tested between 13 and 15 weeks and 8 (19 per cent) before 13 weeks. There were 0.6 per cent false positives by AFP (excluding serious abnormalities and fetal death) and 0.1 per cent after AChE. The likelihood of an open NTD after an elevated AFP (≥2.0 MOM) was 24 and 77 per cent for any serious abnormality. These results, when combined with an earlier study, indicate that amniotic fluid AFP appears to be as sensitive a test for open NTDs between 13 and 15 weeks as between 16 and 20 weeks. Additional experience is necessary to determine this before 13 weeks.  相似文献

Herlitz junctional epidermolysis bullosa (HJEB) is a severe blistering disorder which usually results in death during infancy. We have previously shown that the anchoring filament protein laminin-5 (kalinin/nicein), which mediates keratinocyte attachment and dermal–epidermal cohesion, is abnormally expressed in individuals with HJEB. Laminin-5 was detected by Western blot analysis in amniotic fluid from 44 consecutive normal secondtrimester control pregnancies, but was undetectable in second-trimester amniotic fluid from four pregnancies with fetuses affected by HJEB. In one case of severe non-Herlitz JEB, laminin-5 was detected in both amniotic fluid and skin. In human amniotic fluid, the laminin-5 a3 subunit was processed to a major 165 kD species and a minor 145 kD species and the β2 subunit was partially processed to 105 kD. Although laminin-5 was covalently associated with laminin-6 (K-laminin) in amniotic membrane, no covalent interaction was detected in amniotic fluid. Laminin-5 from amniotic fluid strongly supported keratinocyte attachment. These results suggest that Western blot analysis of second-trimester amniotic fluid is useful in determining the prenatal diagnosis of HJEB and that laminin-5 may serve a physiologically important function in amniotic fluid.  相似文献

In recent years, an increasing number of inherited diseases in man have been recognized in which there is an impairment in the peroxisomal β-oxidation of very-long-chain fatty acids. In general, these disorders are associated with severe neurological and physical abnormalities and death within the first years of life. In this paper we describe our experience with regard to the prenatal diagnosis of a number of different inborn errors of peroxisomal β-oxidation. Eleven pregnancies at risk were monitored by measuring very-long-chain fatty acid levels as well as very-long-chain fatty acid β-oxidation in cultured chorionic villous fibroblasts and/or amniotic fluid cells. Five affected fetuses were identified. It is concluded that prenatal diagnosis in this group of diseases can be done reliably using cultured chorionic villous fibroblasts or amniotic fluid cells.  相似文献

Chorionic villi obtained in the first trimester from a pregnancy at risk for α-mannosidosis were shown to have reduced α-mannosidase (EC 3.2.1.24) activity. The pregnancy was terminated and subsequent enzyme studies of the fetal tissues were consistent with the diagnosis of α-mannosidosis. Like the enzyme in the child's fibroblast, α-mannosidase of the chorionic villi from a pregnancy at risk for α-mannosidosis was activated by high substrate concentration and by Zn2 +, and displayed a Km value two-fold higher than normal. Our results confirm that chorionic villi can be used for early prenatal diagnosis of α-mannosidosis.  相似文献

Our aim in this study was to identify the economically optimal level of use of resources for a grazing system in tropical Australia and to compare it with the biological optimum. Grazing management trials were conducted in the Ord River irrigation area of north-western Australia. Biologists at the Commonwealth Scientific and Industrial Research Organization provided the biological data. The liveweight gains of Kimberley shorthorn steers were recorded, after a 1-year fattening period, for different grazing management strategies. Five different stocking rates were used on irrigated pangola grass (Digitaria decumbens) fertilized with nitrogen at five different rates. All other were applied at fixed, non-limiting levels.The results of the trials were modelled by a liveweight gain production function. This production function was used in an income simulation model to identify economically optimal (profit maximizing) stocking rates. This was done in two stages. First, a base analysis was undertaken on the basis o the economic conditions prevailing in September 1987. Second, sensitivity analysis was applied to the results of the base analysis. The parameters varied were the rate of interest, store cattle prices, finished cattle prices, transport costs and the length of the planning horizon.The biological optimum, when defined as maximum liveweight gain ha−1, corresponds to a much higher intensity of use of resources than the economic optimum, and yields substantially lower profits or higher losses. These discrepancies increase when economic conditions worsen, or when the planning horizon of graziers is decreased from 50 to 10 years. Therefore, the adoption of the biological optimum cannot result in an econonomically stable grazing system in the medium to long term, when it is highly likely that economic parameters will fluctuate. The economic stability of the system is increased, but is still quite weak, if the economically optimal input combination is utilized. It was not possible to determine whether this economic optimum would result in long-term ecological stability because the relevant data were not collected during the trials.The relevance of management recommendations made to graziers and land administrators would be increased if these recommendations were based on an analysis of both the economic and the ecological stability of grazing systems over the medium to long term. We show in the last part of this study how this could be accomplished.  相似文献
