In this report we present several examples in which nickel(II) in combination with DNA damaging agents caused an enhanced or synergistic biological response using several different endpoints. These examples of Ni(II) toxicity represent several approaches designed to understand the genotoxicity of Ni(II) as well as several other metal ions. They are discussed in this report as a partial basis for our hypothesis that Ni(II) may alter the cellular processing of DNA damage at some common point in the pathway for DNA repair of several different agents. In cultured Chinese hamster cells DNA damage by Ni(II) ions was not readily demonstrated by the method of alkaline elution, but pretreatment of cells with Ni(II) before X‐irradiation produced an enhanced amount of strand breaks compared to the amount produced by X‐rays alone. A synergistic enhancement of cell transformation of Syrian hamster embryo cells was observed for combined treatments of Ni(II) and benzo(a)pyrene. The nickel enhancement of mutagenesis by ultraviolet light was demonstrated for the bacterial gene gpt stably integrated into the Chinese hamster V79 genome.  相似文献

A survey was conducted on the accumulation and spatial distribution of PAHs in surface soils under different land use patterns in a valley in the Yangtze Delta region with an area of 10 km2 containing 15 small copper- and zinc-smelting furnaces. Sixty-five topsoil (0–20 cm) samples were collected and 16 PAHs were determined. The average amount of all the 16 PAHs ranged from 0 to 530 μg kg−1 (oven dry basis), with a mean concentration of 33.2 μg kg−1. Benzo[a]pyrene and indeno[1, 2, 3, -cd]pyrene were the two main PAHs present at high concentrations, while pyrene and fluorene had very low concentrations. PAH concentrations were higher in uncultivated than in cultivated soils, and areas of woods and shrubbery had the␣lowest soil PAH contents. The average PAH-homologue concentrations ranked as follows: 5-rings >> 3-rings, 4-rings > 6-rings >2-rings. Much higher concentrations of PAHs were found in the southern part of the sampling area, perhaps due to deposition of airborne particles by the southeasterly winds in winter and spring. We conclude that the small smelting furnaces were the dominant source of PAHs that accumulated in the soils and the southeasterly winds led to the spatial distribution of PAHs in the topsoils. Land vegetation cover and soil utilization patterns also affected the accumulation and distribution of soil PAHs.  相似文献

In a multifactorial pot experiment, maize (Zea mays L.) with or without inoculation with the arbuscular mycorrhizal (AM) fungus Glomus mosseae BEG167 was grown in a sterilized soil spiked with three levels of zinc (0, 300 and 900 mg Zn kg−1 soil) and three levels of cadmium (0, 25 and 100 mg Cd kg−1 soil). At harvest after 8 weeks of growth, the proportion of root length of inoculated plants colonized decreased with increasing Zn or Cd additon, and was 56% in the absence of both metals and was reduced significantly to 27% in the presence of the higher levels of both metals. Mycorrhizal plants had higher biomass than non-mycorrhizal controls except at the highest soil level of Cd. Cadmium had more pronounced effects on plant biomass than did Zn at the levels studied and the two metals showed a significant interaction. The data suggest that mycorrhizal inoculation increased plant growth with enchancement of P nutrition, perhaps increasing plant tolerance to Zn and Cd by a dilution effect. AM inoculation also led to higher soil solution pH after harvest, possibly reducing the availability of the metals for plant uptake, and lowered the concentrations of soluble Zn and Cd in the soil solution, perhaps by adsorption onto the extrametrical mycelium.  相似文献

Lake sediment has long been recognized as an important source of nutrients such as phosphorus. To gain a better understanding of phosphorus flux at the sediment-water interface, it is crucial to investigate the sediment porewater. There is also growing concern and interest in identifying whether organic-rich sediment is an important source of greenhouse gases such as CO(2) and CH(4). In the present study, we took sediment samples from West Lake, a shallow hypereutrophic lake in Hangzhou, Zhejiang Province, China and incubated subsamples under anaerobic conditions at 25 degrees C for 182 d using a specially designed microcosm that permits repeated extraction of sediment porewater and sampling of headspace gases. Anaerobic phosphate fluxes and mineralization of sediment organic carbon were measured. Average diffusive flux of soluble phosphorus was 0.81 mg P m(-2) d(-1) during the initial 18 d of incubation. Decomposition of sediment organic C followed zero-order reaction kinetics and methane accounted for about 50% of the mineralization products. The results suggest that organic-rich sediments can be important sources of P and methane under anaerobic conditions. Laboratory studies simulating field conditions and field studies are necessary to determine the contribution of sediment as a source of P and greenhouse gases.  相似文献

Knowledge of the historical variability of regional climate is an essential element of successful water resource planning. Lacking such perspective, planners and managers can be deceived as to the severity of a recent climate extreme, such as drought, and place a disproportionate blame on the climate, not the integrity of the supply system should water restrictions become necessary to avoid shortages. Presented here is a vivid example of how development, a lack of adequate planning, and climate variability have converged to produce three water emergencies in Rockland County, New York, since 1995. An examination of climate data over the past century indicates that the severity of the recent droughts was well within the range of past variability. Rather than climate alone, the recent water emergencies have highlighted a significant mismatch between supply and demand that has been developing in Rockland County over the past three decades. Substantial development, largely in the form of single‐family homes, has not been matched with a corresponding enhancement of the county's water system. Realistic plans for meeting current water demand will require cooperation among all stakeholders, beginning with an acknowledgement that climate variations are inevitable, not the sole source of blame when water shortages arise.  相似文献

Marsh creation is currently receiving wide attention in the United States as an important tool for mitigating the impacts of development in coastal wetlands. The perception that there is no net loss in valuable coastal wetlands when development is mitigated by the creation of man-made marshes can have a substantial impact on the permitting and decision-making processes. The effective result may be the trading of natural salt marshes for man-made marshes.Techniques for marsh creation were developed by the US Army Corps of Engineers to enhance and stabilize dredge spoil materials. Most research sponsored by the Corps has been directed at determining whether these goals have been accomplished. A survey of the research indicates that there is insufficient evidence to conclude that man-made marshes function like natural salt marshes or provide the important values of natural marshes. It is necessary, therefore, for decision-makers to understand the limitations of present knowledge about man-made marshes, realistically evaluate the trade-offs involved, and relegate mitigation to its proper role in the permitting process—post facto conditions imposed on developments that clearly meet state qualifications and policies.  相似文献

Temporal changes in soil solution properties and metal speciation were studied in non-rhizosphere soil and in the rhizosphere of the hyperaccumulator Thlaspi caerulescens J. & C. Presl (population from Prayon, Belgium) grown in a Zn- and Cd-contaminated soil. This paper focuses on soil solution Zn and pH dynamics during phytoextraction. The concentration of Zn in both non-rhizosphere and rhizosphere soil solutions decreased from 23 mg/l at the beginning to 2 mg/l at the end of the experiment (84 days after transplanting of seedlings), mainly due to chemical sorption. There was no significant difference in overall Zn concentration between the planted and the unplanted soil solutions (P > 0.05). Soil solution pH decreased initially and then increased slightly in both planted and unplanted soil zones. From 60 to 84 days after transplanting, the pH of the rhizosphere soil solution was higher than that of non-rhizosphere soil solution (P<0.05). Zn uptake by the hyperaccumulator plants was 8.8 mg per pot (each containing 1 kg oven-dry soil) on average. The data indicate that the potential of T. caerulescens to remove Zn from contaminated soil may not be related to acidification of the rhizosphere.  相似文献

A modified glass bead compartment cultivation system is described in which glass beads continue to be used in the hyphal compartment but are replaced by coarse river sand in the compartments for host plant roots and mycorrhizal hyphae. Arbuscular mycorrhizal (AM) associations were established using two host plant species, maize (Zea mays L.) and red clover (Trifolium pratense L.) and two AM fungi, Glomus mosseae and G. versiforme. When the standard and modified cultivation systems were compared, the new method yielded much more fungal tissue in the hyphal compartment. Using G. versiforme as the fungal symbiont, up to 30 mg of fungal dry matter (DM) was recovered from the hyphal compartment of mycorrhizal maize and about 6 mg from red clover. Multi-element analysis was conducted on samples of host plant roots and shoots and on harvested fungal biomass. Concentrations of P, Cu and Zn were much higher in the fungal biomass than in the roots or shoots of the host plants but fungal concentrations of K, Ca, Mg, Fe and Mn were similar to or lower than those in the plants. There were also significant differences in nutrient concentrations between the two AM fungi and these may be related to differences in their proportions of extraradical mycelium to spores. The high affinity of the fungal mycelium for Zn was very striking and is discussed in relation to the potential use of arbuscular mycorrhiza in the phytoremediation of Zn-polluted soils.  相似文献
