The published results on nanoparticles cytotoxicity and genotoxicity such as titanium dioxide nanoparticles (TiO(2) NPs) are inconsistent, and often conflicting and insufficient. Since different parameters may have impact on the toxicity results, there is need to lay stress on detailed characterization of NPs and the use of different testing conditions for assessment of NPs toxicity. In order to investigate whether dispersion procedures influence NP cytotoxicity and genotoxicity, we compared two protocols giving TiO(2) NP dispersions with different stability and agglomeration states. Detailed primary and secondary characteristics of both TiO(2) NP dispersions in culture media were carried out before toxicological testing; TK6 human lymphoblast cells, EUE human embryonic epithelial cells and Cos-1 monkey kidney fibroblasts were used to assess cytotoxicity (by trypan blue exclusion, proliferation activity and plating efficiency assays) and genotoxicity (by the comet assay). DNA strand breaks were detected by the alkaline comet assay. DNA oxidation lesions (especially 8-oxo-7,8-dihydroguanine, 8-oxoG) were measured with a modified comet assay including incubation with specific repair enzyme formamidopyrimidine DNA glycosylase (FPG). The TiO(2) NPs dispersion with large agglomerates (3 min sonication and no serum in stock solution) induced DNA damage in all three cell lines, while the TiO(2) NPs dispersed with agglomerates less than 200 nm (foetal serum in stock solution and sonication 15 min) had no effect on genotoxicity. An increased level of DNA oxidation lesions detected in Cos-1 and TK6 cells indicates that the leading mechanism by which TiO(2) NPs trigger genotoxicity is most likely oxidative stress. Our results show that the dispersion method used can influence the results of toxicity studies. Therefore at least two different dispersion procedures should be incorporated into assessment of cyto- and genotoxic effects of NPs. It is important, when assessing the hazard associated with NPs, to establish standard testing procedures and thorough strategies to consider the diverse conditions relevant to possible exposures.  相似文献

Demographic change and economic decline produce modified urban land use pattern and densities. Compared to the beginning of the 90s after the German reunification, nowadays massive housing and commercial vacancies followed by demolition and perforation come to pass in many cities of the former GDR. In consequence, a considerable surplus of urban brownfields has been created. Furthermore, the decline in the urban fabric affects social infrastructure and urban greenery of local neighbourhoods. Here, urban planning enters into ‘uncharted territory’ since it needs to assess the socio-environmental impact of shrinkage.In order to carry out such an evaluation quantitatively, a multi-criteria assessment scheme (MCA) was developed and applied. Firstly, we identified infrastructure and land use changes related to vacancy and demolition. Secondly, demolition scenarios for the coming 20 years were applied in order to give an idea for a long-term monitoring approach at the local district level. A multi-criteria indicator matrix quantifies the socio-environmental impact on both urban greenery and residents. Using it, we set demolition scenarios against urban ‘quality of life’ targets. Empirical evidence comes from Leipzig, in eastern Germany, a representative case study for urban shrinkage processes.The results show that shrinkage implies socio-environmental changes of residential livelihoods, however, does not simply increase or decrease the overall urban quality of life. The integrated assessment of all indicators identifies environmental and social opportunities, as well as the challenges a shrinking city is faced with.  相似文献

European whelks (Buccinum undatum) have shown to accumulate high levels of arsenic. Since the accumulation process is not well understood it is necessary to gain information about the geographical variability of the arsenic concentration in them. Here we show that the mean arsenic concentrations of the whelks are site specific and vary by a factor of 3.5 in ten different geographical locations. At fishing grounds where whelks exhibited low arsenic concentrations the arsenic concentration increased linearly with size, whereas the whelks with high arsenic levels from a different location showed no correlation. Although the overall arsenic concentration in the whelks differed between 45 and 655 mg kg(-1) d.w., the inorganic arsenic concentration did not exceed 0.4 mg kg(-1) d.w. The main arsenic compound is arsenobetaine, which is widely considered as non-toxic. The exposure to toxic inorganic arsenic when eating whelks cannot be estimated from their size or their total arsenic concentration.  相似文献

Although a number of comprehensive reviews have examined global ecosystem services (ES), few have focused on studies that assess urban ecosystem services (UES). Given that more than half of the world’s population lives in cities, understanding the dualism of the provision of and need for UES is of critical importance. Which UES are the focus of research, and what types of urban land use are examined? Are models or decision support systems used to assess the provision of UES? Are trade-offs considered? Do studies of UES engage stakeholders? To address these questions, we analyzed 217 papers derived from an ISI Web of Knowledge search using a set of standardized criteria. The results indicate that most UES studies have been undertaken in Europe, North America, and China, at city scale. Assessment methods involve bio-physical models, Geographical Information Systems, and valuation, but few study findings have been implemented as land use policy.

Prescribed fire is a management tool used by wildland resource management organizations in many ecosystems to reduce hazardous fuels and to achieve a host of other objectives. To study the effects of fire in naturally accumulating fuel conditions, the ambient soil temperature is monitored beneath prescribed burns. In this study we developed a stochastic model for temperature profiles (values at 15 minute intervals) recorded at four depths beneath the soil during a large prescribed burn study. The model was used to assess the temporal fit of the data to particular solutions of the heat equation. We used a random effects model to assess the effects of observed site characteristics on maximum temperatures and to estimate risks of temperatures exceeding critical levels in future similar prescribed fires. Contour plots of estimated risks of temperatures exceeding 60°C for a range of fuel levels and soil depths indicated high risks of occurrence, especially when the moisture levels are low. However, the natural variability among sites seems to be large, even after controlling fuel and moisture levels, resulting in large standard errors of predicted risks.  相似文献

Over the next century, society will increasingly be confronted with the impacts of global change (e.g. pollution, land use changes, and climate change). Multiple scenarios provide us with a range of possible changes in socio-economic trends, land uses and climate (i.e. exposure) and allow us to assess the response of ecosystems and changes in the services they provide (i.e. potential impacts). Since vulnerability to global change is less when society is able to adapt, it is important to provide decision makers with tools that will allow them to assess and compare the vulnerability of different sectors and regions to global change, taking into account exposure and sensitivity, as well as adaptive capacity. This paper presents a method that allows quantitative spatial analyses of the vulnerability of the human-environment system on a European scale. It is a first step towards providing stakeholders and policy makers with a spatially explicit portfolio of comparable projections of ecosystem services, providing a basis for discussion on the sustainable management of Europe’s natural resources.

Reinserting thermoplastic industrial scrap materials back into the production process must be feasible and cost-effective. Unfortunately, many thermoplastic materials contain colour pigments that cause undesired colour effects and, therefore, have to be removed before recycling. The purpose of this study was to reduce the inorganic colour pigments titanium dioxide, chromium(III) oxide and iron(III) oxide from acrylonitrile butadiene styrene. Based on experiences with dissolution-based polymer recycling, the two methods for removing these colour pigments studied in this project were filtration and centrifugation. Multiple laboratory-scale experiments were performed with two solvent formulations, acetone and CreaSolv? SB. Using the filtration method, both solvent formulations achieved reduction rates of 80% for titanium and iron and 90% for chromium. While similar reduction rates were obtained with high-speed laboratory-type centrifuges for acetone solutions, the results for CreaSolv? SB solutions were considerably lower under the same experimental conditions. Increasing the temperature or centrifugation speed, however, will also increase the reduction rates. Thereby, CreaSolv? SB solutions become suitable for industrial-scale processes. This is important because industrial processes based on CreaSolv? SB solutions are significantly safer than acetone-based processes.  相似文献

What defines success and failure of river restoration measures is a strongly debated topic in restoration science, but standardized approaches to evaluate either are still not available. The debate is usually centered on measurable parameters, which adhere to scientific objectivity. More subjective aspects, such as landscape aesthetics or recreational value, are usually left out, although they play an important role in the perception and communication of restoration success. In this paper, we show that different perceptions of restoration success exist by analyzing data from 26 river restoration measures in Germany. We addressed both objective parameters, such as hydromorphological changes and changes in fish and benthic invertebrate assemblages, from field investigations, and subjective parameters, such as opinions and perceptions, from water managers via an online survey. With regard to the objective hydromorphological and biotic parameters, our results agree with many studies that have reported improvements in the hydromorphology following restoration; however, there is no similar agreement between results concerning changes in the benthic invertebrate and fish assemblages. The objective results do not correspond to the subjective parameters because self-evaluation of the restoration projects by water managers was overly positive. Indeed, 40% of the respondents admitted that their evaluation was based on gut feeling, and only 45% of the restoration measures were monitored or occasionally checked. This lack of objectively recorded data meant that the water managers were not able to reasonably evaluate restoration success. In contrast, some self-evaluation responses reflected a different perception of the restoration success that was based on landscape aesthetic values or on benefit for the public; others adopted a general "condemned to success" attitude. Based on our data, we argue (1) that goals should be thoughtfully formulated prior to restoration implementation and (2) that it is necessary to monitor river restoration success from different perspectives.  相似文献

Previous studies evaluating the success of river restorations have rarely found any consistent effects on benthic invertebrate assemblages. In this study, we analyzed data from 24 river restoration projects in Germany dating back 1 to 12 years and 1231 data sets from adjacent river reaches that lie within 0-5, 5-10, and 10-15 km rings centered on the restored sites. We calculated restoration success and recolonization potential of adjacent river reaches based on stream-type-specific subsets of taxa indicative for good or bad habitat quality. On average, the restorations did not improve the benthic invertebrate community quality. However, we show that restoration success depends on the presence of source populations of desired taxa in the surrounding of restored sites. Only where source populations of additional desired taxa existed within a 0-5 km ring around the restored sites were benthic invertebrate assemblages improved by the restoration. Beyond the 5-km rings, this recolonization effect was no longer detected. We present here the first field results to support the debated argument that a lack of source populations in the areas surrounding restored sites may play an important role in the failure to establish desired invertebrate communities by the means of river restorations. In contrast, long-range dispersal of invertebrates seems to play a subordinate role in the recolonization of restored sites. However, because the surroundings of the restored sites were far from good ecological quality, the potential for improvement of restored sites was limited.  相似文献
