The novel Serratia proteamaculans isolated from a chromium-contaminated site was tolerant to a concentration of 500 mg Cr(VI)/l. The optimum pH and temperature for reduction of Cr(VI) by S. proteamaculans were found to be 7.0 and 30 °C, respectively. The Cr(VI) reduction rate decreased with the increase in Cr(VI) concentration from 100 to 400 mg/l, suggesting the enzymatic chromium reduction. Resting and permeabilised cell assays provided the better evidence that chromate reduction in S. proteamaculans is enzymatic. Reduction by cell-free filtrate shows no extracellular chromate-reducing activity, revealing that this activity may be associated to membrane fraction and/or cytosolic fraction. Assays conducted with cytosolic and particulate fraction of S. proteamaculans confirmed the role of membrane-bound proteins in Cr(VI) reduction. Furthermore, chromium reduced by heat-treated cells suggests that membrane-associated chromate reductase activity of S. proteamaculans is preceded by its adsorption on the cell surface. 相似文献

Human serum samples (n?=?113) from Bizerte, northern Tunisia, collected between 2011 and 2012 were analyzed for 8 organochlorine pesticides (OCPs) including p,p′-dichlorodiphenyltrichloroethane (DDT) and its metabolites, hexachlorobenzene (HCB), hexachlorocyclohexane isomers, dieldrin, and heptachlor and 12 polychlorinated biphenyls (PCBs) congeners. Concentrations of these residues in serum were determined by gas chromatography with electron capture detector and total cholesterol (CHOL) and triglycerides (TG) levels were evaluated by enzymatic colorimetric method. HCB, p,p′-DDE, PCB-138, PCB-153, and PCB-180, were the most abundant organochlorine compounds (OCs) detected in >95 % of the study subjects. The mean levels of p,p′-DDE and HCB in serum were 168.8 and 49.1 ng?g?1 lipid, respectively. The sum PCBs concentrations ranged from 37.5 to 284.6 ng?g?1 lipid in the samples, with mean and median value of 136.1 and 123.2 ng?g?1 lipid, respectively. The PCB profile consisted of persistent congeners, such as PCB-138, PCB-153, and PCB-180 which contributed for approximately 82.7 % to the ∑PCBs. Statistical analysis showed that most OCs correlated significantly with age, considering all samples together or with gender differentiation. The present study shows that the levels of p,p′-DDE and ∑DDTs were significantly higher in females than in males (p?<?0.05), while PCBs levels were significantly higher in male (p?<?0.05) than in females. The effects of ten generational zinc or cadmium pre-exposure on metal tolerance among beet armyworm Spodoptera exigua individuals were compared. These effects were assessed in animals from the 11th generation, reared on a diet either uncontaminated or contaminated with metal (cadmium or zinc). The survival rate of larvae and the degree of metal accumulation (in larvae, pupae and moths; among larval organs: gut and fat body) were analysed. Catalase, superoxide dismutase and glutathione transferase activity in larval organs of individuals subjected to different metal treatments were also measured. Animals transferred from control rearing to metals (cadmium or zinc) in the 11th generation, as well as those from multigenerational zinc treatment, but not from multigenerational cadmium treatment, had a significantly lower survival rate than control animals. Insects from the groups with the high metal treatment had high bioaccumulation factors (above 3.7 and 2.3 following cadmium and zinc, respectively). Cadmium (but not zinc) pre-exposure had a significant effect on metal accumulation in larvae. Multigenerational metal pre-exposure seemed to have mainly a negative effect on glutathione transferase activity in the gut of larvae from the 11th generation, in the case of the individuals exposed to metal other than that used in pre-exposure treatment or kept in control conditions. However, in the case of zinc pre-exposure, such effect was only apparent when zinc was replaced by cadmium. The long-term effect of cadmium on catalase activity in larvae was found. 相似文献

Linear alkylbenzene sulfonate (LAS) is a synthetic anionic surfactant widely present in the environment due to its intensive production and use in the detergency field. Admitting that current procedure of risk assessment has limits in providing realistic risk assessment data and predicting the cumulative effect of the toxicant mixtures, the incorporation of information regarding the mode of action and cell response mechanism seems to be a potential solution to overcome these limits. In this regard, we investigated in this study the LAS cytotoxicity on human intestinal Caco-2 cells, trying to unveil the protein actors implicated in the cell response using proteomics approach in order to give a better understanding of the toxicological effect and allow the identification of appropriate biomarkers reflecting the mode of action associated with LAS. As results, we demonstrated that LAS induces a time- and dose-dependent cytotoxicity in Caco-2 cells accompanied by an induction of oxidative stress followed by an excessive increase of intracellular calcium level. Proteomics approach helped in discovering three informative biomarkers of effect associated with LAS cytotoxic effect, reported for the first time: calreticulin, thioredoxin, and heat shock cognate 71 (HSP7C), confirmed by real-time PCR and western blot analysis. These biomarkers could serve for more reliable future risk assessment studies that consider the toxicants mode of action in order to help in the prediction of potential cumulative effects of environmentally coexisting contaminants. 相似文献

Abstract Brain tissue samples of nice (7.5 g from 25 mouse brains and lamb (25 g) were homogenized and subcellular fractions prepared in order to assay the distribution of neurotoxic esterase (NTE) activity. The specific inhibitor, N,N‐diisopropylphosphorodiamidic fluoride (mipafox) was synthesized and purified. Maximum specific activity of NTE was reached in the microsomal fraction (110,000 g) while the enzyme activity in the soluble fraction (110,000 g) was extremely low. This subcellular distribution of NTE activity in mammal brains is an original contribution. Brain microsomal fraction is suggested to be a more reliable source for the highest activity of NTE. The specific activity of NTE of lamb brain was much higher than that of mouse brain. This night help interpretation of the characteristic species variation in susceptibility to NTE inhibitors which are known to be potent delayed neurotoxic agents. 相似文献

Abstract 14C‐DDT dissipated gradually from natural water under outdoor conditions and declined to 40% of the applied radioactivity after 5 months. The losses are due to adsorption to particulates and volatilization from the water surface. In natural water DDT undergoes gradual conversion to DDE as the major degradation product and to a lesser extent to DDD. It may be concluded that DDT dissipates and degrades fairly rapidly in subtropical natural waters. Adsorption to particulate matter contributes to partial “removal” of DDT. 相似文献

This research work consists on valorizing poultry waste by biotransformation into biofertilizers, associating this agro-industrial waste with algae (abundant natural resources) and molasses (a by-product of the sugar refining industry) ensuring a good contribution of nutritional chemical elements and obtaining a balanced formulation. A total of seven different formulations of the above three components, were examined in a simplex centroid design. A fungal inoculum of Aspergillus niger was used as a fermentation agent for better quality of biotransformation. The monitoring of this biotransformation is ensured during 15 days by following the evolution of physicochemical and microbiological parameters and, to understand the bioconversion of the simple compounds of the biofertilizer mixture such as short aliphatic chains, sugar, and amino acids into soluble mineral compounds, a Fourier transform infrared spectroscopy (FTIR) analysis was carried out before and after biotransformation. Finally, germination and fertilization tests were performed to evaluate the efficiency of the final product on a barley crop. The overall results of the present study showed that the mixture which contained 68.75% poultry waste, 12.5% molasses, and 18.75% algae presented the better microbiological and chemical safety criteria required for a good biofertilizer according to NF U44-551 standard. 相似文献

The term sustainability invites interest in the main factors that affect the deterioration of natural resources. Different hypotheses have been put forward concerning the relationship between water and its effect on different economic sectors. Several questions can be asked here, for example: Does a higher water withdrawal for one sector mean that this sector is adding more to the GDP or is it a sunk cost (it is the cost that should be ignored compared to the benefit of water withdrawal for an individual economic sector) compared with the benefits to the economy? Do social factors affect water withdrawal more than economic factors or are they both impacting equally? We aim to answer these questions and to shed light on different socio-economic factors that affect water withdrawal in different economic sectors. This study investigates, in depth, the interaction between humans and the environment and can be useful in monitoring the direct effect on water withdrawal from agricultural and non-agricultural sectors and on different national economic variables that act as an indicator for economic development and growth. We used simultaneous equation models in our analysis, both the three-stage least squares and the two-stage least squares to explore the relationships. For more credibility, we run the fixed and random effects of 2SLS. Our results showed the influence of trade openness and economic growth on water withdrawal for different economic sectors, and the effect of an increasing demand for water for non-agricultural purposes, which adds pressure on the agricultural sector and eventually may lead to rising food prices. 相似文献
