In this study, Bacillus cereus isolate from tannery effluent was employed for orange II dye decolorization in simulated minimal salt broth and textile effluent. Most of the physicochemical parameters of textile effluent were above the permissible limits. The strain was highly tolerant to dye up to 500 mg l?1. Increasing dye concentration exerted inhibitory effect on the bacterial growth and decolorization. The maximum decolorization of initial 100 mg dye l?1 was achieved at optimum pH 8.0 and 33 °C under static culture conditions during 96-h incubation. Supplementation with optimized glucose (0.4 %, w/v) and ammonium sulfate (0.1 %, w/v) with 3.0 % B. cereus inoculum further enhanced dye decolorization to highest 68.5 % within 96-h incubation. A direct correlation was evident between bacterial growth and dye decolorization. Under above optimized conditions, 24.3 % decolorization of unsterilized real textile effluent by native microflora was achieved. The effluent decolorization enhanced substantially to 37.1 % with B. cereus augmentation and to 40.5 % when supplemented with glucose and ammonium sulfate without augmentation. The maximum decolorization of 52.5 % occurred when textile effluent was supplemented with optimized exogenous carbon and nitrogen sources along with B. cereus augmentation. Gas chromatography–mass spectrometry identified sulfanilic acid as orange II degradation product. Fourier transform infra red spectroscopy of metabolic products indicated the presence of amino and hydroxyl functional groups. This strain may be suitably employed for in situ decolorization of textile industrial effluent under broad environmental conditions.  相似文献

A preliminary investigation was conducted to assess lignocellulolytic efficiency of crude extracts from three white-rot fungi, Pleurotus florida PF05 (PF), Pleurotus sajor-caju PS07 (PS) and Pleurotus eryngii PE08 (PE). The activities of CMC-ase, xylanase, beta-glucosidase, beta-xylosidase, laccase and Mn peroxidase in extracts were evaluated. PF produced its highest CMC-ase (317 UL(-1)'), beta-glucosidase (62 UL(-1)), beta-xylosidase (37 UL(-1)) and laccase (347 UL(-1)) activities while, PS produced highest xylanase (269 UL-(1)) and Mn peroxidase (69 UL(-1)) activities. In addition, crude extracts extracted were employed for their in vitro degradability assessment; and were evaluated with mono and mixed extracts separately to corn cob substrate. The losses in cell wall components and dry matter during 5 and 10 days incubations were analyzed after treatments of extracts. Maximum 8.2, 4.4 and 2.8% loss were found respectivelyin hemicellulose (HC), cellulose (C) and lignin (L) with mono extract of PF within 10 days. The influence of mono extract of each strain (PF PS and PE) and their mixed extracts (PF+PS, PF+PE, PS+PE and PF+PS+PE) on degradation of cell wall constituents were remarkably differed. The mixed extract treatment proved maximum 13.6% HC loss by PF+PS+PE extract, 9.2% loss in C by PF+PS extract and 5.2% loss of L by the PF+PS+PE extract treatment. The highest dry matter loss (8.2%) was recorded with PF+PS+PE mixed extract combination.  相似文献

The Burmese roofed turtle (Batagur trivittata) is one of the world's most endangered turtles. Only one wild population remains in Myanmar. There are thought to be 12 breeding turtles in the wild. Conservation efforts for the species have raised >700 captive turtles since 2002, predominantly from eggs collected in the wild. We collected tissue samples from 445 individuals (approximately 40% of the turtles’ remaining global population), applied double‐digest restriction‐site associated DNA sequencing (ddRAD‐Seq), and obtained approximately 1500 unlinked genome‐wide single nucleotide polymorphisms. Individuals fell into 5 distinct genetic clusters, 4 of which represented full‐sib families. We inferred a low effective population size (≤10 individuals) but did not detect signs of severe inbreeding, possibly because the population bottleneck occurred recently. Two groups of 30 individuals from the captive pool that were the most genetically diverse were reintroduced to the wild, leading to an increase in the number of fertile eggs (n = 27) in the wild. Another 25 individuals, selected based on the same criteria, were transferred to the Singapore Zoo as an assurance colony. Our study demonstrates that the research‐to‐application gap in conservation can be bridged through application of cutting‐edge genomic methods.  相似文献

The activity of microorganisms often plays an important role in dynamic natural attenuation or engineered bioremediation of subsurface contaminants, such as chlorinated solvents, metals, and radionuclides. To evaluate and/or design bioremediated systems, quantitative reactive transport models are needed. State-of-the-art reactive transport models often ignore the microbial effects or simulate the microbial effects with static growth yield and constant reaction rate parameters over simulated conditions, while in reality microorganisms can dynamically modify their functionality (such as utilization of alternative respiratory pathways) in response to spatial and temporal variations in environmental conditions. Constraint-based genome-scale microbial in silico models, using genomic data and multiple-pathway reaction networks, have been shown to be able to simulate transient metabolism of some well studied microorganisms and identify growth rate, substrate uptake rates, and byproduct rates under different growth conditions. These rates can be identified and used to replace specific microbially-mediated reaction rates in a reactive transport model using local geochemical conditions as constraints. We previously demonstrated the potential utility of integrating a constraint-based microbial metabolism model with a reactive transport simulator as applied to bioremediation of uranium in groundwater. However, that work relied on an indirect coupling approach that was effective for initial demonstration but may not be extensible to more complex problems that are of significant interest (e.g., communities of microbial species and multiple constraining variables). Here, we extend that work by presenting and demonstrating a method of directly integrating a reactive transport model (FORTRAN code) with constraint-based in silico models solved with IBM ILOG CPLEX linear optimizer base system (C library). The models were integrated with BABEL, a language interoperability tool. The modeling system is designed in such a way that constraint-based models targeting different microorganisms or competing organism communities can be easily plugged into the system. Constraint-based modeling is very costly given the size of a genome-scale reaction network. To save computation time, a binary tree is traversed to examine the concentration and solution pool generated during the simulation in order to decide whether the constraint-based model should be called. We also show preliminary results from the integrated model including a comparison of the direct and indirect coupling approaches and evaluated the ability of the approach to simulate field experiment.  相似文献

Biological treatment of high-strength nitrogenous wastewater is challenging due to low growth rate of autotrophic nitrifiers. This study reports bioaugmentation of Thiosphaera pantotropha capable of simultaneously performing heterotrophic nitrification and aerobic denitrification (SND) in sequencing batch reactors (SBRs). SBRs fed with 1:1 organic-nitrogen (N) and NH4 +-N were started up with activated sludge and T. pantotropha by gradual increase in N concentration. Sludge bulking problems initially observed could be overcome through improved aeration and mixing and change in carbon source. N removal decreased with increase in initial nitrogen concentration, and only 50–60 % removal could be achieved at the highest N concentration of 1000 mg L?1 at 12-h cycle time. SND accounted for 28 % nitrogen loss. Reducing the settling time to 5–10 min and addition of divalent metal ions gradually improved the settling characteristics of sludge. Sludge aggregates of 0.05–0.2 mm diameter, much smaller than typical aerobic granules, were formed and progressive increase in settling velocity, specific gravity, Ca2+, Mg2+, protein, and polysaccharides was observed over time. Granulation facilitated total nitrogen (TN) removal at a constant rate over the entire 12-h cycle and thus increased TN removal up to 70 %. Concentrations of NO2 ?-N and NO3 ?-N were consistently low indicating effective denitrification. Nitrogen removal was possibly limited by urea hydrolysis/nitrification. Presence of T. pantotropha in the SBRs was confirmed through biochemical tests and 16S rDNA analysis.  相似文献

The physicochemical analyses of pulp-paper mill effluent revealed that it was dark brown with 1761?±?2.3 color PtCo units having slightly alkaline pH, high biological oxygen demand and chemical oxygen demand values, and contained large quantities of organic and inorganic constituents, well above the prescribed standards. The bacterial growth, color reduction, and dechlorination were evident in all the four sets of experiments with different possible combinations of nutrient supplementation and Pseudomonas putida augmentation. A high degree of decolorization at 29.7% and 27.4% was observed by the effluent native microflora during 48 and 24 h, in unaugmented effluent supplemented with glucose + yeast extract and glucose + peptone, respectively. The extent of decolorization in glucose + yeast extract unaugmented effluent also corresponded with high degree of dechlorination (59.3%) during 60-h incubation (SET III). An appreciable level of growth, decolorization, and dechlorination was evident in nutrient unsupplemented P. putida augmented effluent as well as in the control natural effluent. However, a maximum level of growth response (OD 1.641-1.902) during 36-48 h, removal of color (39.72-48.2%) during 24-36 h, and chloride ions (80.1-83.5%) during 36 h was achieved in P. putida augmented effluent supplemented with glucose + yeast extract or peptone. Therefore, supplementation of effluent with glucose and yeast extract or peptone and concomitant augmentation with P. putida is required for efficient effluent decolorization and detoxification.  相似文献

The integrated pest management (IPM) modules of pesticide schedule on Basmati rice were validated at field experiments conducted in Northern India for consecutive 3 years (2005–2008). The pesticide residues were found below the detectable limit (<0.01–0.001 mg/kg) in soil and irrigation water samples of Kaithal region. In Dehra Dun region of Uttrakhand, the residues of carbendazim in rice grains and soil were detected below <0.01 mg/kg level. In second year experiments (2006–2007), only four non-IPM soil samples indicated the presence of chlorpyrifos and endosulfan in the range of ND <0.001 to 0.07 mg/kg, out of 45 samples analyzed. Carbendazim applied as seed treatment at Dehradun and Kaithal field trials was found below detectable limit in both IPM and non-IPM rice grains (<0.01 mg/kg) and irrigation water (0.01 μl/ml). Chlorpyrifos was detected in five water samples from Kaithal and one from Pant Nagar in the range of 0.003–0.006 μl/L, α- and β-isomer of endosulfan in the range of 0.005–0.03, and 0.005–0.02 μl/ml, respectively, in one sample from Pant Nagar and two from Kaithal, out of a total of 22 samples. In the region of Uttrakhand and Uttar Pradesh during 2007–2008, four non-IPM samples of soil indicated trace levels of endosulfan, out of 16 samples analyzed. The residues were detected below detection limit for carbendazim (<0.01 mg/kg) in soil samples of Dehradun IPM fields and for endosulfan and carbendazim (0.001–0.01 μl/L) in water samples each from IPM and non-IPM fields of Uttar Pradesh. The results of 3-year trials of IPM module indicated basmati rice as safe and economical with pesticide residue-free rice grains.  相似文献

Ground water quality of Hisar city was assessed for its suitability for drinking purposes. Samples collected from the Bore-wells (forms a part of municipal water supply) and handpumps (direct consumption) were analyzed for the various physico-chemical parameters including pH, electrical conductivity, total dissolved salts, total hardness, total alkalinity, sodium, potassium, calcium, magnesium, carbonate, bicarbonate, chloride and sulfate. The concentrations of magnesium, sodium, potassium, sulfate and especially of chloride were found moderately higher than the WHO standards for the drinking water. Further a comparison of fluoride (F) levels in groundwater of various cities and towns of Haryana state was performed. The relatively higher concentrations of F in groundwater of Haryana raise the risk of fluorosis and hence groundwater must be used with proper treatment. Promising defluoridation methods using locally available materials and technologies are discussed for the prevention and control of fluorosis. Data were assessed statistically to find the suitable markers of ground water quality as an aid to monitoring groundwater quality.  相似文献

In the present study, catalytic wet oxidation (CWO) was investigated for the destruction of organic pollutants in the thermally pretreated effluent from a pulp and paper mill under moderate temperature and pressure conditions. The thermal pretreatment studies were conducted at atmospheric pressure and 368K using copper sulfate as a catalyst. The thermal pretreatment reduced COD by about 61%. The filtrate of the thermal pretreatment step was used at pH 8.0 for CWO at 383-443K temperature and a total pressure of 0.85MPa for 4h. Catalysts used for the reaction include copper sulfate, 5% CuO/95% activated carbon, 60% CuO/40% MnO(2), and 60% CuO/40% CeO(2). Maximum COD reduction was found to be 89% during CWO step using 5% CuO/95% activated carbon with a catalyst loading of 8gl(-1) at 443K and 0.85MPa total pressure. Overall COD reduction for the pretreatment and the CWO was found to be 96%. Besides this, 60% CuO/40% CeO(2) catalyst also exhibited the similar activity as that of obtained with 5% CuO/95% activated carbon catalyst at 423K temperature and 0.85MPa total pressure. The pH of the solution during the experimental runs decreases initially due to the formation of carboxylic acid and then increases due to the decomposition of acids.  相似文献
