The microbial community cultured from the marine sponge Rhopaloeides odorabile Thompson et al. is dominated by a single bacterium, designated strain NW001. Sequence analysis of 1212 bp of the16S rRNA
gene of strain NW001 indicates that it is a member of the α-subgroup of the class Proteobacteria. The association between
this bacterium and its host sponge was observed in healthy R. odorabile collected from six different reefs in the Great Barrier Reef representing a geographic distance of 460 km, and in four collections
in different seasons in 1997–1998 at Davies Reef (18°49.6′S; 147°34.49′E). The proportion of colonies of strain NW001 in samples
from R. odorabile, expressed as a percentage of the total heterotrophic bacterial colony count, showed no significant spatial (range: 81–98%)
or temporal differences (range: 81–99%), although colony counts of strain NW001 varied by up to two orders of magnitude between
reef sites and sampling periods. The location of strain NW001 within the sponge mesohyl was visualized by in situ hybridization,
using fluorescently labeled probes based on the 16S rRNA gene sequence of this strain. Cells of strain NW001 surround the
choanocyte chambers, suggesting that these bacteria may play a role in nutrient uptake by the sponge. The absence of strain
NW001 from corresponding seawater samples indicates that it has a specific, intimate relationship with R. odorabile and is not being utilized as a food source. A unique cyanobacterium related to the genera Leptolyngbya and Plectonema was also isolated from R. odorabile and characterized by 16S rRNA gene sequencing.

The aim of this paper is to identify what constituted barriers to progress in the implementation of the Management Standards for preventing and reducing work-related stress nationally, an approach advocated by the Health and Safety Executive in Great Britain. Data were collected from more than 100 public sector organisations through inspector visits and research interviews. Findings show that under supportive contexts, organisations were able to follow the process of a stepwise method for assessing psychosocial risks and implementing interventions using HSE assessment tools and guidance. Main enabling factors included the active and visible support from senior management, human resource departments, and line managers; regular communications on progress, sufficient organisational capability in terms of resources and expertise; departmental/team level assessment as opposed to an overall corporate wide assessment, and involvement of key stakeholders (e.g. Trade Union, employees). Some of the critical barriers across many public sector organisations included in this study were: major or on-going organisational changes; lack of organisational capability; and the resource intensive aspect of the method requiring focus groups in addition to stress survey data. My concern is to question the quality and utility of science in general and ethology in particular as applied to animal welfare.
This topic has in the past provoked me to some severe criticism, for example, 'A lot of well-intended welfare research is
neither very good science nor very helpful to the animals.... Too much welfare research is (in my opinion) flawed either because
it is oversimplistic, or because it is not so much designed to test preconceptions but to reinforce prejudice' (Webster 1994).
Dawkins (1997) has recently responded to this challenge, addressing the question 'Why has there not been more progress in
welfare research?' Her response is concerned largely with applied ethology. My own criticism was not directed at ethologists
in particular. I was more concerned by the misuse of scientific method by those who seek to obtain a so-called 'objective'
measurement of something which they preconceive to be a stress (e.g. measurement of plasma concentrations of cortisol or endorphins
in animals following transportation). Here the 'objective' measure frequently becomes the test that gives the answer that
they want, and if it fails, then they seek other 'objective' markers until they achieve a set of measurements that supports
the subjective impression which they had at the outset.
My second main concern is that the welfare state of a sentient animal is a very complex affair and cannot be embraced by any
single scientific discipline, be it ethology, physiology, molecular or neurobiology. Unfortunately it is also too complex
to be embraced by a single-sentence definition. The best I can do is to suggest that it is determined by the capacity of an
animal to sustain physical fitness and avoid mental suffering. The assessment of this is necessarily multidisciplinary. 相似文献

Abstract Radiolabelled end‐use mixtures of glyphosate with and without a cationic surfactant (Ethomeen® T/25) and an organosilicone surfactant (Silwet® L‐77) were applied onto trembling aspen (Populus tremuloids Michx.) leaves at the rate of 1.0 kg of AE (acid equivalent) in 35 L/ha area of foliage. A 5‐mm rainfall with an intensity of 10 mm/h was applied at intervals of 0.5, 8, 24, 36, 48, 72, and 96 h after treatment. Glyphosate washoff was determined by liquid scintillation counting of radioactivity in the rain‐washing. At 36 h post‐treatment, both the adjuvants significantly reduced glyphosate washoff (Ethomeen by 69.6% and Silwet by 59.7%) from foliage, compared to the washoff (82.6%) when Vision alone was applied without the adjuvants. Results on the rate of plant growth indicated that with a rain‐free period of 8 h or more, the growth of most seedlings was stunted within 1 or 2 d. Percentage of foliar browning 20 d after treatment with rain‐free period of 8 to 48 h ranged from 8 to 80% for Vision alone, 75 to 100% for Vision with Ethomeen, and 85 to 100% for Vision with Silwet, respectively. Physical properties of the end‐use mixtures were measured with and without the two adjuvants to examine droplet spreading and drying rates in relation glyphosate rainfastness. The Silwet adjuvant lowered the surface tension of the end‐use mixture, but Ethomeen did not. Droplets containing Silwet were spread more than those containing Ethomeen. However, the greater area of contact caused by Silwet did not contribute to a significant increase in the translocation rate of glyphosate into untreated parts of the seedlings, and showed no relationship with rainfastness of glyphosate deposits on trembling aspen. 相似文献

The soil of some 50 km(2) around the town of Weinfelden in north-east Switzerland has been sampled and analysed to estimate and map the concentrations of heavy metals before an incinerator for the canton's waste is built. Given that the estimates are subject to error, the probabilities that true values exceed the maximum tolerable concentrations of the Swiss federal guide have also been estimated by disjunctive kriging. These may now be used by the local planners for making decisions. Of the metals examined, lead exceeded the guide value at several sampling points, and a moderate probability of excess of lead is fairly widespread. Copper exceeded the guide value in vineyards and orchards, but also in a few places elsewhere, and the estimated probabilities of excess were not negligible. The other two metals of concern, cadmium (with one exception) and zinc, had concentrations much less than the guide values. The patterns of distribution of the four metals were similar, the product-moment correlations among them are substantial, except in the vineyards and orchards, and it seems likely that they had common sources. 相似文献

Wild and farmed mussels, Mytilus edulis, coexist with salmon farms in Scottish sea lochs. A synthetic pyrethroid, cypermethrin, is licensed for use on fish farms to control sea lice infestations as a formulation called Excis. In this study, uptake of cypermethrin from Excis exposure is investigated through the use of gas chromatography with mass-spectrometry. The effects of Excis on mussels are also examined by measuring the neutral red retention time of lysosomes, aerial survival and shell closure. The isomeric ratios of cis:trans cypermethrin measured in mussels are around 80:20; a marked increase from 40:60 to which the mussels were exposed. This is most likely due to preferential metabolism of trans-isomers, as the same response is seen in vertebrates. There is a pronounced behavioural effect of shell closure, where mussels exposed to 1000 microg/l cypermethrin shut their shells within an hour of exposure. Arguments are presented for this effect being either a voluntary response on recognition of cypermethrin, or an effect arising from an involuntary action of cypermethrin on the adductor muscle. Even at 1000 microg/l cypermethrin, neutral red retention time and aerial survival are not affected. The data suggest that the responses of mussels shown here are unlikely to occur in the field, even at the concentrations of cypermethrin used in fish cages, for the treatment of sea lice. 相似文献
