Metabolic profiling in plants can be used to differentiate between treatments and to search for biomarkers for exposure. A methodology for processing Ultra-High-Performance Liquid Chromatography-Diode-Array-Detection data is devised. This methodology includes a scheme for selecting informative wavelengths, baseline removal, retention time alignment, selection of relevant retention times, and principal component analysis (PCA). Plant crude extracts from rapeseed seedling exposed to sublethal concentrations of glyphosate are used as a study case. Through this approach, plants exposed to concentrations down to 5 μM could be distinguished from the controls. The compounds responsible for this differentiation were partially identified and were different from those specific for high exposure samples, which suggests that two different responses to glyphosate are elicited in rapeseed depending on the level of exposure. The PCA loadings indicate that a combination of other metabolites could be more sensitive than the response of shikimate to detect glyphosate exposure. 相似文献

In this study, a 3D urban groundwater model is presented which serves for calculation of multispecies contaminant transport
in the subsurface on the regional scale. The total model consists of two submodels, the groundwater flow and reactive transport
model, and is validated against field data. The model equations are solved applying finite element method. A sensitivity analysis
is carried out to perform parameter identification of flow, transport and reaction processes. Coming from the latter, stochastic
variation of flow, transport, and reaction input parameters and Monte Carlo simulation are used in calculating probabilities
of pollutant occurrence in the domain. These probabilities could be part of determining future spots of contamination and
their measure of damages. Application and validation is exemplarily shown for a contaminated site in Braunschweig (Germany),
where a vast plume of chlorinated ethenes pollutes the groundwater. With respect to field application, the methods used for
modelling reveal feasible and helpful tools to assess natural attenuation (MNA) and the risk that might be reduced by remediation

Determination of emission of contaminant gases as ammonia, methane, or laughing gas from natural ventilated livestock buildings with large opening is a challenge due to the large variations in gas concentration and air velocity in the openings. The close relation between calculated animal heat production and the carbon dioxide production from the animals have in several cases been utilized for estimation of the ventilation air exchange rate for the estimation of ammonia and greenhouse gas emissions. Using this method, the problem of the complicated air velocity and concentration distribution in the openings is avoided; however, there are still some important issues remained unanswered: (1) the precision of the estimations, (2) the requirement for the length of measuring periods, and (3) the required measuring point number and location. The purpose of this work was to investigate how estimated average gas emission and the precision of the estimation are influenced by different calculation procedures, measuring period length, measure point locations, measure point numbers, and criteria for excluding measuring data. The analyses were based on existing data from a 6-day measuring period in a naturally ventilated, 150 milking cow building. The results showed that the methane emission can be determined with much higher precision than ammonia or laughing gas emissions, and, for methane, relatively precise estimations can be based on measure periods as short as 3 h. This result makes it feasible to investigate the influence of feed composition on methane emission in a relative large number of operating cattle buildings and consequently it can support a development towards reduced greenhouse gas emission from cattle production. 相似文献

Assessing the Effects of Nutrient Management in an Estuary Experiencing Climatic Change: The Neuse River Estuary, North Carolina 总被引:1,自引:0,他引:1
Eutrophication is a serious water quality problem in estuaries receiving increasing anthropogenic nutrient loads. Managers
undertaking nutrient-reduction strategies aimed at controlling estuarine eutrophication are faced with the challenge that
upstream freshwater segments often are phosphorus (P)-limited, whereas more saline downstream segments are nitrogen (N)-limited.
Management also must consider climatic (hydrologic) variability, which affects nutrient delivery and processing. The interactive
effects of selective nutrient input reductions and climatic perturbations were examined in the Neuse River Estuary (NRE),
North Carolina, a shallow estuary with more than a 30-year history of accelerated nutrient loading and water quality decline.
The NRE also has experienced a recent increase in Atlantic hurricanes and record flooding, which has affected hydrology and
nutrient loadings. The authors examined the water quality consequences of selective nutrient (P but not N) reductions in the
1980s, followed by N reductions in the 1990s and an increase in hurricane frequency since the mid-1990s. Selective P reductions
decreased upstream phytoplankton blooms, but increased downstream phytoplankton biomass. Storms modified these trends. In
particular, upstream annual N and P concentrations have decreased during the elevated hurricane period. Increased flushing
and scouring from storms and flooding appear to have enhanced nutrient retention capabilities of the NRE watershed. From a
management perspective, one cannot rely on largely unpredictable changes in storm frequency and intensity to negate anthropogenic
nutrient enrichment and eutrophication. To control eutrophication along the hydrologically variable freshwater–marine continuum,
N and P reductions should be applied adaptively to reflect point-source–dominated drought and non–point-source–dominated flood

Abstract: The Soil and Water Assessment Tool (SWAT) has been applied successfully in temperate environments but little is known about its performance in the snow‐dominated, forested, mountainous watersheds that provide much of the water supply in western North America. To address this knowledge gap, we configured SWAT to simulate the streamflow of Tenderfoot Creek (TCSWAT). Located in central Montana, TCSWAT represents a high‐elevation watershed with ~85% coniferous forest cover where more than 70% of the annual precipitation falls as snow, and runoff comes primarily from spring snowmelt. Model calibration using four years of measured daily streamflow, temperature, and precipitation data resulted in a relative error (RE) of 2% for annual water yield estimates, and mean paired deviations (Dv) of 36 and 31% and Nash‐Sutcliffe (NS) efficiencies of 0.90 and 0.86 for monthly and daily streamflow, respectively. Model validation was conducted using an additional four years of data and the performance was similar to the calibration period, with RE of 4% for annual water yields, Dv of 43% and 32%, and NS efficiencies of 0.90 and 0.76 for monthly and daily streamflow, respectively. An objective, regression‐based model invalidation procedure also indicated that the model was validated for the overall simulation period. Seasonally, SWAT performed well during the spring and early summer snowmelt runoff period, but was a poor predictor of late summer and winter base flow. The calibrated model was most sensitive to snowmelt parameters, followed in decreasing order of influence by the surface runoff lag, ground water, soil, and SCS Curve Number parameter sets. Model sensitivity to the surface runoff lag parameter reflected the influence of frozen soils on runoff processes. Results indicated that SWAT can provide reasonable predictions of annual, monthly, and daily streamflow from forested montane watersheds, but further model refinements could improve representation of snowmelt runoff processes and performance during the base flow period in this environment. 相似文献

Assessing PAH and PCB emissions from the relocation of harbour sediments using equilibrium passive samplers 总被引:3,自引:1,他引:2
Large-scale dredging of contaminated sediments is taking place in the harbor of Oslo, Norway. The dredged sediment masses are transferred into a confined aquatic disposal facility (CAD) in a natural 70-m deep basin within the Oslofjord. Currently there is no established method to determine how much dissolved contaminants are released during relocation and deposition of these sediments. For this reason we tested the use of equilibrium passive samplers consisting of 55 microm thin polyoxymethylene (POM-55) for studying the release of freely dissolved and thus bioavailable PAHs and PCBs at the disposal site, and found this to be a suitable method. In order to use POM-55 for monitoring PCBs, it was necessary to measure their POM-55/water partition coefficients, which was also presented as part of this study. Elevated turbidity (average 4.1 mg l(-1)) was observed at one side of the basin where no natural sill exists. Analysis of POM-55 at this location before and after deposition revealed that there was an increase in freely dissolved concentrations (C(W,free)) during deposition by a factor 37.5 for PAHs and a factor of 2.9 for PCBs. In addition, during deposition phenanthrene-to-anthracene aqueous concentration ratios at this location (values of 3-4) were more similar to those of the deposited sediments (approximately 2) than to those of the CAD water prior to deposition (approximately 14). This was not observed for the other locations where a natural sill exists at approximately 30 m water depth. The POM-55 equilibrium passive samplers are here shown to be useful tools for measuring and understanding the dynamics involved in the release of dissolved contaminants during sediment relocation. 相似文献

Wyoming’s Green Mountain Common Allotment is public land providing livestock forage, wildlife habitat, and unfenced solitude,
amid other ecological services. It is also the center of ongoing debate over USDI Bureau of Land Management’s (BLM) adjudication
of land uses. Monitoring resource use is a BLM responsibility, but conventional monitoring is inadequate for the vast areas
encompassed in this and other public-land units. New monitoring methods are needed that will reduce monitoring costs. An understanding
of data-set relationships among old and new methods is also needed. This study compared two conventional methods with two
remote sensing methods using images captured from two meters and 100 meters above ground level from a camera stand (a ground,
image-based method) and a light airplane (an aerial, image-based method). Image analysis used SamplePoint or VegMeasure software.
Aerial methods allowed for increased sampling intensity at low cost relative to the time and travel required by ground methods.
Costs to acquire the aerial imagery and measure ground cover on 162 aerial samples representing 9000 ha were less than $3000.
The four highest correlations among data sets for bare ground—the ground-cover characteristic yielding the highest correlations
(r)—ranged from 0.76 to 0.85 and included ground with ground, ground with aerial, and aerial with aerial data-set associations.
We conclude that our aerial surveys are a cost-effective monitoring method, that ground with aerial data-set correlations
can be equal to, or greater than those among ground-based data sets, and that bare ground should continue to be investigated
and tested for use as a key indicator of rangeland health. 相似文献

The effects of persistent organic pollutants on renal and liver morphology in farmed arctic fox (Vulpes lagopus) were studied under experimental conditions. Control animals received a diet containing pork (Sus scrofa) fat with low amounts of persistent organic pollutants, while the diet of the exposed animals contained whale blubber, 'naturally' contaminated with persistent organic pollutants. Polychlorinated biphenyls (PCB) and organochlorine pesticide (OCP) concentrations in the whale blubber were 488 and 395 ng/g wet weight, respectively. Animals were sacrificed and sampled when they were at their fattest (winter) as well as their lowest body weight (summer). The results show that PCB and OCP exposure causes renal (and probably also liver) lesions in arctic foxes. The prevalence of glomerular, tubular and interstitial lesions was significantly highest in the exposed group (chi-square: all p<0.05). The frequency of liver lesions (steatosis, intravascular granulocyte accumulations, interstitial cell infiltrations, lipid granulomas, portal fibrosis and bile duct hyperplasia) were also highest in the exposed group, although not significantly (chi-square: all p>0.05). The prevalence of lesions was not significantly different between lean (winter) and fat (summer) foxes for any of the lesions (chi-square: all p>0.05). We suggest that wild arctic foxes exposed to an environmental cocktail of persistent organic pollutants, such as PCBs and OCPs, in their natural diet are at risk for developing chronic kidney and liver damage. Whether such lesions may have an impact on age and health of the animals remains uncertain. 相似文献

Abstract: We used a retrospective approach to identify hydrologic metrics with the greatest potential for ecological relevance for use as resource management tools (i.e., hydrologic indicators) in rapidly urbanizing basins of the Puget Lowland. We proposed four criteria for identifying useful hydrologic indicators: (1) sensitive to urbanization consistent with expected hydrologic response, (2) demonstrate statistically significant trends in urbanizing basins (and not in undeveloped basins), (3) be correlated with measures of biological response to urbanization, and (4) be relatively insensitive to potentially confounding variables like basin area. Data utilized in the analysis included gauged flow and benthic macroinvertebrate data collected at 16 locations in 11 King County stream basins. Fifteen hydrologic metrics were calculated from daily average flow data and the Pacific Northwest Benthic Index of Biological Integrity (B‐IBI) was used to represent the gradient of response of stream macroinvertebrates to urbanization. Urbanization was represented by percent Total Impervious Area (%TIA) and percent urban land cover (%Urban). We found eight hydrologic metrics that were significantly correlated with B‐IBI scores (Low Pulse Count and Duration; High Pulse Count, Duration, and Range; Flow Reversals, TQmean, and R‐B Index). Although there appeared to be a great deal of redundancy among these metrics with respect to their response to urbanization, only two of the metrics tested – High Pulse Count and High Pulse Range – best met all four criteria we established for selecting hydrologic indicators. The increase in these high pulse metrics with respect to urbanization is the result of an increase in winter high pulses and the occurrence of high pulse events during summer (increasing the frequency and range of high pulses), when practically none would have occurred prior to development. We performed an initial evaluation of the usefulness of our hydrologic indicators by calculating and comparing hydrologic metrics derived from continuous hydrologic simulations of selected basin management alternatives for Miller Creek, one of the most highly urbanized basins used in our study. We found that the preferred basin management alternative appeared to be effective in restoring some flow metrics close to simulated fully forested conditions (e.g., TQmean), but less effective in restoring other metrics such as High Pulse Count and Range. If future research continues to support our hypothesis that the flow regime, particularly High Pulse Count and Range, is an important control of biotic integrity in Puget Lowland streams, it would have significant implications for stormwater management. 相似文献
