A poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) degrading bacterial strain designated as AF-111 was isolated from sewage sludge sample. The bacterium was identified by 16S rRNA gene sequencing. The results revealed that strain AF-111 showed 99 % similarity with Streptomyces althioticus strain NRRL B-3981 and designated as Streptomyces sp. strain AF-111. An extracellular PHBV depolymerase enzyme was produced under optimized conditions and purified through ammonium sulphate fractionation and column chromatography. The enzyme was purified to homogeneity, indicated by sodium dodecyl sulfate–polyacrylamide gel electrophoresis and molecular weight was found to be approximately 51 kDa. Effect of temperature, pH, metal ions and inhibitors on the PHBV depolymerase activity was determined. The enzyme was stable at wide range of temperature (35–55 °C) and pH (6–8). PHBV depolymerase was stable in the presence of different metal ions except iron and zinc which had inhibitory effect on depolymerase activity. Both ethylenediamine teteracetic acid and phenylmethyl sulphonyl fluoride strongly inhibited enzyme activity which indicates that this enzyme belongs to the serine hydrolase family like other polyhydroxyalkanoate depolymerases. The results show that a depolymerase from strain AF-111 can effectively degrade PHBV, therefore, it can be applied in the process of biochemical monomer recycling. 相似文献

Climate‐change induced uncertainties in future spatial patterns of conservation‐related outcomes make it difficult to implement standard conservation‐planning paradigms. A recent study translates Markowitz's risk‐diversification strategy from finance to conservation settings, enabling conservation agents to use this diversification strategy for allocating conservation and restoration investments across space to minimize the risk associated with such uncertainty. However, this method is information intensive and requires a large number of forecasts of ecological outcomes associated with possible climate‐change scenarios for carrying out fine‐resolution conservation planning. We developed a technique for iterative, spatial portfolio analysis that can be used to allocate scarce conservation resources across a desired level of subregions in a planning landscape in the absence of a sufficient number of ecological forecasts. We applied our technique to the Prairie Pothole Region in central North America. A lack of sufficient future climate information prevented attainment of the most efficient risk‐return conservation outcomes in the Prairie Pothole Region. The difference in expected conservation returns between conservation planning with limited climate‐change information and full climate‐change information was as large as 30% for the Prairie Pothole Region even when the most efficient iterative approach was used. However, our iterative approach allowed finer resolution portfolio allocation with limited climate‐change forecasts such that the best possible risk‐return combinations were obtained. With our most efficient iterative approach, the expected loss in conservation outcomes owing to limited climate‐change information could be reduced by 17% relative to other iterative approaches. 相似文献

We investigated selected chlorinated pollutants (β-HCH, γ-HCH, DDDs, DDEs, o,p′-DDT, p,p′-DDT, heptachlor, aldrin, dieldrin, and endrin) in the Lahore and the Sialkot districts of Pakistan, using eggs of cattle egret (Bubulcus ibis) collected during May and June 2007. The pollutant with highest level and frequency was ΣDDT, followed by β-HCH, γ-HCH, heptachlor, aldrin, dieldrin, and endrin in descending order. The concentration(s) were significantly higher in Sialkot heronry for all the pollutants (except p,p′-DDT) than in Lahore. The values for DDTs, β-HCH, γ-HCH, and heptachlor were significantly higher (p < 0.05) in the egg(s) than in sediment(s) and in the chicks’ diet, due to biomagnification. Among DDTs analogues, p,p′-DDD was the major contaminant with >60 % of total DDT burden, reflecting the widespread aged as well as recent use of DDT as well as anaerobic degradation (DDD/DDE > 1 in many cases) in the nearby paddy soils. In few samples, p,p′-DDT/(DDD + DDE) > 0.5 suggested the recent emission patterns from surrounding contaminated areas of demolished DDT units and obsolete pesticide stores. The higher levels of HCHs (i.e., β-HCH) in the samples collected from Sialkot indicate exposure from long-term agricultural use. Overall, concentrations of all studied POPs were less than the threshold levels known to affect reproduction. Nevertheless, total DDTs and/or HCHs burdens in some eggs contained concentrations of greater than what would educe adverse effects on birds. Link Emissions Models estimate traffic-related air pollution emissions at the individual road link level and inform governmental policies for air quality management. The current South Australian Link Emissions Model (CLEM) assumes constant spatiotemporal traffic flow at a single fixed mean speed, a potential limitation as the variability of exhaust emissions with vehicle speed has been established in the literature.We extend CLEM to eliminate the assumption of constant traffic flow, through the derivation of mean Australian vehicle speed distributions for different road types. Specifically, we successfully model the vehicle speed profile data from the second National In-Service Emissions study using Nearest Neighbour Kernel Density Estimation. We propose a mean speed Distribution Link Emissions Model (DLEM) for exhaust emission estimation based on the derived mean speed distributions. DLEM is an augmented, enhanced version of CLEM, accommodating a range of vehicle speeds and road types. The performance of the extended model, DLEM, is analysed in comparison to the current model, CLEM, through a case study analysis of vehicle exhaust emissions on a typical arterial road in Adelaide, South Australia. Results indicate use of DLEM and, by extension, mean vehicle speed distributions, has a strong impact on emission estimation. In particular, the fixed speed model, CLEM, may be substantially underestimating exhaust emissions of carbon monoxide, non-methane volatile organic compounds and particulate matter less than 2.5 μm in diameter. A method for the residual pendimethalin in soil and vegetable samples was developed. The method is based on extraction of pendimethalin from samples using microwave-assisted solvent extraction (MASE) with acetone, ethanol, and water as extraction solvent. Extracted pendimethalin samples were analyzed by high-performance liquid chromatography with ultraviolet detector at 240 nm. The MASE parameters, temperature, heating time, and solvent types were optimized with the feasibility of MASE application in the determination of pendimethalin extraction efficiency of pendimethalin from soil and vegetable samples. The maximum temperature that can be used during the heating for MASE is 60°C, where the recovery percentages reached 97%. Linearity for pendimethalin was found in the range of 2?C20 ??g mL???1 with limits of detection and limits of quantification of 0.059 and 0.17 ??g mL???1, respectively. 相似文献

Aerosol constituents (elemental carbon, organic carbon, soluble ions including organic acids, and selected trace metals) were investigated from samples of a field campaign taking place at Bhola Island in the Bay of Bengal (Bangladesh). The campaign took place in the pre-monsoon season (May 2001) using low volume samplers. Carbonaceous material comprised the majority of the analysed components. The average concentrations of EC and OC were 2.8 and 4.6 microg m(-3), respectively. Oxalic acid was the most abundant dicarboxylic acid (average 268 ng m(-3)) followed by malonic and malic acid. The contribution of carboxylic acids-carbon to organic carbon was 2.0%. Average concentrations observed for sulfate and nitrate were 3.7 microg m(-3) and 1.5 microg m(-3). Two different types of aerosol were identified at the rural background site on Bhola Island during southerly synoptic flow by means of trajectory analysis: air masses were transported from the Bay of Bengal to the sampling site in all cases. However, during "Period 1" they experienced longer residence times over the Indian Ocean, while the "Period 2" trajectories came along the Indian coast or passed over the Indian continent. During Period 1 the concentration levels of soluble ions were a factor of 4-6 lower than during Period 2. The concentrations of EC, OC and K differed less than a factor of 1.5 between the two periods. The Period 1 aerosol showed similarities to the haze layers observed during winter-monsoon conditions south of India during the INDOEX experiment. Based on EC/TC and K/EC ratios we find that around 80% of the carbonaceous aerosol from Period 1 in Bhola is from fossil fuel and only around 50% from Period 2. Absolute concentrations of carbonaceous species, soluble ions and trace metals indicate that the background site on Bhola Island is affected by emissions from urbanized regions of Southeast Asia. 相似文献

Using nitric acid-perchloric acid wet digestion based FAAS method ten selected metals (Cd, Co, Cr, Fe, K, Mn, Na, Ni, Pb and Zn) were determined in the scalp hair of male Pakistani and Libyan donors (n = 62), between 3–54 years age and residing in typical urban areas. The study was taken up to identify sources of metal distribution in two diverse population segments exposed to different environments. Sodium emerged with the highest mean hair concentration of 511.0 μg/g and 496.9 μg/g for Libyan and Pakistani donors respectively, followed by K, at 266.2 μg/g and 242.4 μg/g for the two donor categories. Cadmium showed the lowest mean hair concentration for both Pakistani (0.380 μg/g) and Libyan (0.530 μg/g) donors. Levels of Na, K, Pb, Cr and Cd were higher in hair of Libyan donors compared with Pakistani counterparts which showed higher comparative levels of Zn, Fe, Co, Ni and Mn. Strong metal-to-metal correlations were found between Na–K (r = 0.638) and Cd–Co (r = 0.574) for Pakistani donors, while for Libyan donors, Na–K (r = 0.680) and Fe–Mn (r = 0.624) correlations emerged as strongly significant. Only K was found to have a strong positive correlation (r = 0.611) with age for Pakistani donors while this correlation was significantly negative (r = −0.500) for Libyan donors, probably arising from individual food habits. Other metals showed no viable relationship with age. Principal Component Analysis (PCA) and Cluster Analysis (CA) of the data evidenced food, industrial emissions and automobile emissions as possible sources of metal distribution.
