The paper describes an experimental set-up designed to measure the volatilization of different pesticides after application under full-scale field conditions. The pesticides were sprayed around the circumference of a circle and measurements of meteorology and air concentrations of pesticides were taken in the centre of the circle. The paper describes how the dimensions like the radius of the circle and height of the mast in the experimental set-up are found from theoretical considerations. A method to increase the confidence of the flux calculations is described. It utilizes that several substances were applied simultaneously and that a certain parameter in the fitted concentration profiles depends on turbulence only and thus have the same value for all substances. A conceptual model that simulates the formation and fate of odorous substances in branched collection systems is presented. The model predicts the activity of the relevant biomass phenotypes under aerobic, anoxic, and anaerobic conditions in force mains and gravity sewers. The formation and fate of individual, malodorous substances in the bulk water, biofilms, and sediments are modeled. The release of odorous compounds from the bulk water to the sewer gas phase, their fate in the gas phase, and their subsequent release into the urban atmosphere is simulated. The continuously growing global demands on a finite land resource will require better strategic policies and management of trade-offs to avoid conflicts between different land-use sectors. Visions of the future can support strategic planning by stimulating dialogue, building a consensus on shared priorities and providing long-term targets. We present a novel approach to elicit stakeholder visions of future desired land use, which was applied with a broad range of experts to develop cross-sectoral visions in Europe. The approach is based on (i) combination of software tools and facilitation techniques to stimulate engagement and creativity; (ii) methodical selection of stakeholders; (iii) use of land attributes to deconstruct the multifaceted sectoral visions into land-use changes that can be clustered into few cross-sectoral visions, and (iv) a rigorous iterative process. Three cross-sectoral visions of sustainable land use in Europe in 2040 emerged from applying the approach in participatory workshops involving experts in nature conservation, recreation, agriculture, forestry, settlements, energy, and water. The three visions—Best Land in Europe, Regional Connected and Local Multifunctional—shared a wish to achieve a land use that is sustainable through multifunctionality, resource use efficiency, controlled urban growth, rural renewal and widespread nature. However, they differ on the scale at which land services are provided—EU-wide, regional or local—reflecting the land-sparing versus land-sharing debate. We discuss the usefulness of the approach, as well as the challenges posed and solutions offered by the visions to support strategic land-use planning.
Flow through artificial macropores may occur as a water film along the macropore walls (film flow) or as moving water segments separated by air bubbles (pulse flow). To investigate the effect of macropore flow pattern (i.e., film and pulse flow) on the interaction of solutes with macropore walls, we studied orthophosphate (P) transport and sorption in artificial macropores. The experimental setup consisted of a column (height = 20 cm, diameter = 20 cm) homogenously packed with glass beads and fitted at outflow with a vertical artificial macropore placed below the column. The artificial macropore consisted of ceramic tubes (3 or 8 mm i.d.; 31.5 cm long) coated on the inside with iron oxide serving as phosphate sorbents. An orthophosphate solution containing 0.04 mg P L(-1) was applied at a rate of 9 to 12 mm h(-1) to the column, eventually causing macropore flow. In the 8-mm-i.d. tubes only film flow occurred. Pulse flow was dominating in the 3-mm-i.d. tubes. Generally, the flow patterns were reproducible and seldom did pulse flow replaced film flow or vice versa. During film flow, a significantly larger decrease in macropore P concentration per tube was observed relative to that with pulse flow events. However, pulse and film flow lead to almost the same amounts of P sorbed per unit surface area when exposed to the same solute P concentration. Comparison with P sorption capacity experiments indicated that the sorption rate, rather than the sorption capacity, controls the amount of sorbed P during macropore flow in the studied system. 相似文献
A method for direct detection of antibiotic resistance genes in soil samples has been developed. The tetracycline resistance gene, tet(M), was used as a model. The method was validated on Danish farmland soil that had repeatedly been treated with pig manure slurry containing resistant bacteria. The tet(M) gene was directly detected in 10-80% of the samples from the various farmland soils and could be detected in all samples tested after selective enrichment. To validate the obtained results, the method was applied to garden soil samples where lower prevalence of resistance was found. RESULT: A detection limit of 10(2)-10(3) copies of the tet(M) gene per gram of soil (in a Bacillus cereus group bacterium) was achieved. tet(M) gene was detected in soil samples with the highest prevalence on farmland treated with pig manure slurry. 相似文献
The presence of erm genes was investigated among macrolide-resistant Gram-positive bacteria isolated from soil samples collected from four Danish farms that had been treated with animal waste. Soil samples were collected before, a few days after spread and 1 1/2 months and 5 months later. In 33% (9/27) of these isolates, an erm gene was detected using PCR. Eight isolates were positive for erm(B) and one isolate was positive for erm(C). No isolates contained erm(A), erm(D) or erm(F). The positive isolates were identified to genus level. Two erm(B) positive isolates were identified as Enterococcus spp., and the erm(C)-positive isolate as a Streptococcus spp., probably indicating survival from animal waste. The remaining six erm(B) positive isolates all belonged to the Bacillus cereus group. The erm(B) gene has not previously been identified in B. cereus but is frequently found among enterococci. This result could indicate horizontal transfer from bacteria of animal origin to indigenous soil bacteria. 相似文献
We aimed to compare cell-based NIPT (cbNIPT) to chorionic villus sampling (CVS) and to examine the test characteristics of cbNIPT in the first clinical validation study of cbNIPT compared to cell-free NIPT (cfNIPT).
Material and Methods
Study 1: Women (N = 92) who accepted CVS were recruited for cbNIPT (53 normal and 39 abnormal). Samples were analyzed with chromosomal microarray (CMA). Study 2: Women (N = 282) who accepted cfNIPT were recruited for cbNIPT. cfNIPT was analyzed using sequencing and cbNIPT by CMA.
Results
Study 1: cbNIPT detected all aberrations (32/32) found in CVS: trisomies 13, 18 and 21 (23/23), pathogenic copy number variations (CNVs) (6/6) and sex chromosome aberrations (3/3). cbNIPT detected 3/8 cases of mosaicism in the placenta. Study 2: cbNIPT detected all trisomies found with cfNIPT (6/6) and had no false positive (0/246). One of the three CNVs called by cbNIPT was confirmed by CVS but was undetected by cfNIPT, two were false positives. cbNIPT detected mosaicism in five samples, of which two were not detected by cfNIPT. cbNIPT failed in 7.8% compared to 2.8% in cfNIPT.
Conclusion
Circulating trophoblasts in the maternal circulation provide the potential of screening for aneuploidies and pathogenic CNVs covering the entire fetal genome. 相似文献
In a previous article, Beschta et al. (Environ Manag 51(2):474–491, 2013) argue that grazing by large ungulates (both native and domestic) should be eliminated or greatly reduced on western public lands to reduce potential climate change impacts. The authors did not present a balanced synthesis of the scientific literature, and their publication is more of an opinion article. Their conclusions do not reflect the complexities associated with herbivore grazing. Because grazing is a complex ecological process, synthesis of the scientific literature can be a challenge. Legacy effects of uncontrolled grazing during the homestead era further complicate analysis of current grazing impacts. Interactions of climate change and grazing will depend on the specific situation. For example, increasing atmospheric CO2 and temperatures may increase accumulation of fine fuels (primarily grasses) and thus increase wildfire risk. Prescribed grazing by livestock is one of the few management tools available for reducing fine fuel accumulation. While there are certainly points on the landscape where herbivore impacts can be identified, there are also vast grazed areas where impacts are minimal. Broad scale reduction of domestic and wild herbivores to help native plant communities cope with climate change will be unnecessary because over the past 20–50 years land managers have actively sought to bring populations of native and domestic herbivores in balance with the potential of vegetation and soils.
