Dihydropteridine reductase (DHPR) is an enzyme involved in the recycling of tetrahydrobiopterin (BH4), which is an obligate co-factor of the aromatic amino acid hydroxylases. DHPR deficiency is a rare, autosomal recessive disorder caused by mutations in the QDPR gene. DHPR-deficient patients are diagnosed by a lack of response to a low phenylalanine diet and by severe neurological symptoms. Final diagnosis is made by measurements of neurotransmitters and pterin metabolites in cerebrospinal fluid (CSF) and urine, in addition to DHPR enzyme activity, which can be assessed in whole red blood cells. Treatment of DHPR deficiency can be difficult and the outcome is not always satisfying, even if all treatment strategies are followed. Therefore prenatal diagnosis is of great importance in affected families. Prenatal diagnosis is possible by measuring DHPR activity in different cell types but this is time consuming. More than 25 different mutations have to date been identified in the QDPR gene and direct identification of a mutation in a fetus would be easy and rapid. We have developed a method based on denaturing gradient gel electrophoresis (DGGE) for the analysis of the QDPR gene. The method is useful for rapid and simultaneous scanning of all exons and flanking intronic sequences of the QDPR gene. We describe the first prenatal diagnosis conducted using this method. The paper reports heavy metal accumulation in algae collected at four stations and in sediments at three stations on the Black Sea coast of Turkey. The metals analysed are Cd, Co, Cr, Cu, Fe, Pb, Sb and Zn. The metal content of algae increased generally (with some exceptions) from 1991 till 1993 in ?ile and Sinop. In the sediments Sb in ?ile, As in Riva, Fe, Zn in Sinop are high. According to these findings the metal pollution increased in Turkish area of the Black Sea during the years investigated.  相似文献

The kinetics of the photocatalytic degradation of 10 different dichlorophenols and dichloroanilines have been investigated experimentally and theoretically. With the intention of finding certain molecular indices in order to determine the degradation rates of aromatic pollutants, geometry optimizations of the compounds have been performed with the semiempirical PM3 method. The molecular orbital calculations have been carried out by an SCF method using the RHF formalism. The correlations between the apparent first-order rate constants and the calculated molecular properties of the compounds have been examined. Four different structure–activity relationships have been developed expressing the logarithms of the rate constants in terms of the Hammett constants, electron densities of the substituents, the coefficients and the energies of the highest occupied molecular orbitals HOMO, and 1-octanol/water partition coefficients log?K OW.  相似文献

This study aims to investigate the phytotoxic effects of both Punica granatum peel extract directed copper oxide nanoparticles (CuONPs) and copper nitrate on Elodea canadensis. The CuONPs were characterised by UV–visible spectroscopy (UV-Vis), Dynamic Light Scattering (DLS), X-ray Diffraction (XRD), Scanning Electron Microscope (SEM), and Fourier Transform Infrared (FT-IR) Spectroscopy. The ratio of photosynthetic pigments, total soluble protein, malondialdehyde (MDA), catalase (CAT), ascorbate peroxidase (APX), and copper accumulation in the E. canadensis was determined to evaluate the phytotoxicity of the CuONPs and copper nitrate on E. canadensis. The significant improvements were observed in the growth rate of E. canadensis, chlorophyll a and b, carotenoid contents, and total soluble protein in the E. canadensis when it was exposed to the CuONPs. Additionally, CAT and APX activities and MDA contents were enhanced by accumulation of the CuONPs into the E. canadensis. These results suggested that oxidative stress was alleviated, owing to the result of an increase in the antioxidants such as CAT and APX, with an increase in the concentration of the CuONPs. Finally, the findings of this study show that copper nitrate has many negative effects on E. canadensis compared bio-synthesised CuONPs.  相似文献

In a previous study, we found that juvenile northern wheatears (Oenanthe oenanthe) exposed to a magnetic displacement to the west of their natural migration route increased their body mass. The total intensity and inclination used for the western displacement may also have been interpreted as northern compared to the experimental site (stronger total field intensity and steeper inclination angle). In order to investigate whether the fuelling response was a response to an unexpected magnetic field or specific to the northern magnetic field, we conducted a new experiment. Juvenile wheatears from the same study population were magnetically displaced to southwestern magnetic fields, exposing the birds to unexpected magnetic combinations, but eliminating the possible effect of a northern magnetic field. A control group was kept in the local geomagnetic field in Sweden for comparison. There was no difference in body mass increase between treatments, suggesting that the fuelling response previously found was not a simple response to an unexpected magnetic field, but rather a specific response to the northern magnetic field. Juvenile wheatears may have developed a fuelling response to northern magnetic fields in order to enable a successful flight towards the migration goal.  相似文献

Fuel cell (FC) hybrid vehicle power trains are an attractive technology especially for automotive applications because of their higher efficiency and lower emissions compared to conventional vehicles. This study focuses on the design of an FC hybrid power train system and evaluation of its simulations for a given speed profile through two alternative power management algorithms (PMAs). Parameters suitable for a small vehicle were taken into consideration in the mathematical model of the vehicle. The proposed hybrid power train consists of an energy storage system, composed of a 4-kg battery pack (either lithium-ion (Li-ion) battery, nickel metal hydride, or nickel–cadmium) and a direct methanol fuel cell (DMFC) as the range extender. The PMAs basically aim to fulfill the power requirements of the vehicle and decide how to command the power split between the battery and the FC. The model comprising a DMFC, a battery, and PMAs was developed in MATLAB/Simulink environment. The polarization curve of the FC was obtained using a one-dimensional DMFC model. Vehicle power requirements for a drive cycle were calculated using the equations of longitudinal dynamics of vehicle, and the results were integrated into MATLAB/Simulink model. As a result of the simulations, methanol consumption, state of charge of the battery, and power output of the FC were compared for the PMAs. This comparison shows the effect of PMAs on the hybrid vehicle performance for three battery types. The results indicate that the vehicle range could be increased when proper strategy is used as PMA.  相似文献
