We present a spatiotemporal evaluation of water quality incidents in Japan considering incident numbers, incident causes, pollutant categories, and pollution effects. Water pollution incidents in first-class river systems almost tripled to about 1487 in the 12 years from 1996 to 2007. In addition, oil makes up the largest proportion of pollutants nationwide (76.61%) and the major source of pollution for each region in Japan. Moreover, every category shows a growth trend, especially since 2005. The main cause of incidents was “Unknown” (43%), followed by “Poor working practice” (24%), and then by “Accident” (10%) and “Other” (10%). In Hokuriku, however, the main cause of incidents was “Poor working practice” (36%), which is greater than “Unknown” (30%). Finally, waterworks (approximately 60%) was the largest of four kinds of water supply infrastructure affected by pollution incidents, followed by simplified waterworks. The population affected by offensive odors and tastes peaked in 1990 and has been decreasing. Overall, the results show the characteristics of incidents from 1996 to 2007, with significant implications for adaptation measures, strategies and policies to reduce water quality incidents.  相似文献

Livestock manure is suitable for use as a composting material. However, various intestinal microbes, such as Escherichia coli, are significant components of such manures. Thus, it is desirable that the level of intestinal microbes, and particularly opportunistic pathogens, in compost is inspected and counted regularly. The sensitivity and specificity of detection of E. coli in compost have been improved by selective cultivation followed by colony polymerase chain reaction (PCR) using the ECO primer. Indeed, the sensitivity of this method is higher than that of DNA extraction from compost and PCR. In this study, changes in numbers of E. coli present in a field-scale composting process over time was assessed using selective cultivation and colony PCR. Numbers of ECO-positive colonies after 24 h decreased, with a concomitant rise in compost temperature. ECO-positive colonies were not detected from 33 to 48 h. However, ECO-positive colony numbers increased beginning on day 4 and continuing until day 42. Thus, it seems likely that the high temperatures reached during the composting process did not affect E. coli numbers in the final compost. Additionally, selective cultivation followed by colony PCR using specific primers is an appropriate method of determining levels of cultivable pathogens in composted materials.  相似文献

The effectiveness of genetic bioaugmentation relies on efficient plasmid transfer between donor and recipient cells as well as the plasmid’s phenotype in the recipient cell. In the present study, the effects of varying organic carbon substrates, initial recipient-to-donor cell density ratios, and mixtures of known recipient bacterial strains on the conjugation and function of a TOL plasmid were tested in sterile soil slurry batch reactors. The presence of soil organic carbon was sufficient in ensuring TOL plasmid transconjugant occurrence (up to 2.1 ± 0.5%) for most recipient strains in soil slurry batch mating experiments. The addition of glucose had limited effects on transconjugant occurrence; however, glucose amendment increased the specific toluene degradation rates of some Enterobacteriaceae transconjugants in soil slurry. Initial cell density ratios and mixtures of recipient strains had smaller impacts on plasmid conjugation and resulting phenotype functionality. These observations suggest that genetic bioaugmentation may be improved by minimal altering of environmental conditions.  相似文献

We have developed a system that streams and archives live sound from remote areas across Japan via an unmanned automatic camera. The system was used to carry out pilot bird censuses in woodland; this allowed us to examine the use of live sound transmission and the role of social media as a mediator in remote scientific monitoring. The system has been streaming sounds 8 h per day for more than five years. We demonstrated that: (1) the transmission of live sound from a remote woodland could be used effectively to monitor birds in a remote location; (2) the simultaneous involvement of several participants via Internet Relay Chat to listen to live sound transmissions could enhance the accuracy of census data collection; and (3) interactions through Twitter allowed members of the public to engage or help with the remote monitoring of birds and experience inaccessible nature through the use of novel technologies.  相似文献

In order to promote pollutant monitoring and preservation of water resources, we evaluate the spatiotemporal trends in recent water quality conditions in Japanese rivers. Trend analysis is conducted on the 92 major rivers in Japan using the available water quality data recorded from 1992 to 2005 and the characteristics of major pollutants in these rivers are analyzed. Spatial and temporal analysis of trends for six water quality indicators is conducted using the Mann Kendall test, a non-parametric statistical method. The indicators analyzed are biochemical oxygen demand (BOD), chemical oxygen demand (COD), dissolved oxygen (DO), total nitrogen (TN), total phosphorus (TP) and pH. The majority of sampling locations monitoring BOD, COD, TN and TP show trends toward decreasing concentrations over time. Many sampling locations show increasing DO concentrations. Our results show that water quality in Japanese rivers has improved dramatically over the past decade, although there are still problems in some places, most notably in the Hokkaido, Kanto, Kinki and Kyushu regions. The improvements seen in water quality appear to be the result of improved wastewater treatment and other water quality improvement efforts achieved through government initiative.  相似文献

An electron microscopic DOPA reaction test of fetal skin was used for the prenatal diagnosis of tyrosinase-negative oculocutaneous albinism (OCA). The subject was a 34-year-old Japanese woman in her second pregnancy. Her first child, born in 1982, had been previously examined and confirmed to have tyrosinase-negative OCA. The parents requested a prenatal diagnosis and we sampled skin from the upper trunk of the fetus. On conventional electron microscopy, the development of melanosomes in interfollicular melanocytes had progressed no further than stage II. Fetal skin samples incubated with L-DOPA solution indicated a lack of tyrosinase activity and showed that the melanosomes had not progressed beyond stage II. In skin samples from the trunks of three Japanese fetuses aborted for other reasons at 19–20 weeks of gestation, most premature melanosomes were further melanized to stage IV after incubation with L-DOPA solution. A prenatal diagnosis of tyrosinase-negative OCA was made. The parents requested a termination and skin biopsies of the abortus confirmed the diagnosis. This study shows that tyrosinase is normally present in melanocytes of the fetal epidermis at 20 weeks' gestation, and that the electron microscopic DOPA reaction test of a fetal skin biopsy specimen is safe and practical, and provides reliable information for making a prenatal diagnosis of tyrosinase-negative OCA in the second trimester.  相似文献

To better understand the role of soil organic matter in terrestrial carbon cycle, carbon isotope compositions in soil samples from a temperate-zone forest were measured for bulk, acid-insoluble and base-insoluble organic matter fractions separated by a chemical fractionation method. The measurements also made it possible to estimate indirectly radiocarbon ((14)C) abundances of acid- and base-soluble organic matter fractions, through a mass balance of carbon among the fractions. The depth profiles of (14)C abundances showed that (1) bomb-derived (14)C has penetrated the first 16cm mineral soil at least; (2) Delta(14)C values of acid-soluble organic matter fraction are considerably higher than those of other fractions; and (3) a significant amount of the bomb-derived (14)C has been preserved as the base-soluble organic matter around litter-mineral soil boundary. In contrast, no or little bomb-derived (14)C was observed for the base-insoluble fraction in all sampling depths, indicating that this recalcitrant fraction, accounting for approximately 15% of total carbon in this temperate-zone forest soil, plays a role as a long-term sink in the carbon cycle. These results suggest that bulk soil organic matter cannot provide a representative indicator as a source or a sink of carbon in soil, particularly on annual to decadal timescales.  相似文献

Hydrogen sulfide (H2S) is one of the most toxic and offensively odorous gases and is generated in anaerobic bioreactors. A middle-thermophilic sulfur-oxidizing bacterium (SOB), Thiomonas sp. strain RAN5, was isolated and applied for H2S removal from both artificial and anaerobically digested gas. When a bioreactor containing medium inoculated with RAN5 was aerated continuously with artificial gas (containing 100 ppm H2S) at 45 degrees C for 156 hr, the H2S concentration in the vented gas was reduced by 99%. This was not affected by the presence of other microbes in the bioreactor The H2S removal efficiency of the RAN5 bioreactor for anaerobically digested gas was greater than 99% at influent H2S concentrations ranging from 2 to 1800 ppm; the efficiency decreased to 90% at influent H2S concentrations greater than 2000 ppm. Thiomonas sp. strain RAN5 cannot survive at room temperature, and thus its leakage from a wastewater treatment plant would not damage sewage systems. These data suggest that Thiomonas sp. strain RAN5 may be a useful microorganism for H2S removal.  相似文献
