Fine particulatematter (PM2.5) is associated with increased risks of Alzheimer’s disease (AD),yet the toxicologicalmechanisms of PM2.5 promoting AD remain unclear. In this study,wildtype
and APP/PS1 transgenic mice (AD mice) were exposed to either filtered air (FA) or PM2.5 for eight weeks with a real-world exposure system in Taiyuan, China (mean PM2.5 concentration
in the cage was 61 μg/m3). We found that PM2.5 exposure could remarkably aggravate AD mice’s ethological and brain ultrastructural damage, along with the elevation of the pro-inflammatory cytokines (IL-6 and TNF-α), Aβ-42 and AChE levels and the decline of ChAT levels in the brains. Based on high-throughput sequencing results, some differentially expressed (DE) mRNAs and DE miRNAs in the brains of AD mice after PM2.5 exposure were screened.Using RT-qPCR, seven DEmiRNAs (mmu-miR-193b-5p, 122b-5p, 466h-3p, 10b-5p, 1895, 384–5p, and 6412) and six genes (Pcdhgb8, Unc13b, Robo3, Prph, Pter, and Tbata)

A new strategy for the bioproduction of all ketohexoses was developed using hexitols as intermediates. Biocatalysts used to employ the strategy were D-tagatose 3-epimerase, which epimerizes ketohexoses at the C-3 position, and oxidoreductases, which catalyze oxidation-reduction reactions between ketohexoses and the corresponding hexitols. Arranging all the ketohexoses and hexitols in a symmetric ring and connecting them with 20 biochemical reactions, I was able to construct a design for the bioproduction of all the rare ketohexoses. Various aldose isomerases transform ketohexoses into the corresponding aldohexoses, so the strategy is useful for the bioproduction of all the rare hexose sugars. Although forest conservation activities, particularly in the tropics, offer significant potential for mitigating carbon (C)
emissions, these types of activities have faced obstacles in the policy arena caused by the difficulty in determining key
elements of the project cycle, particularly the baseline. A baseline for forest conservation has two main components: the
projected land-use change and the corresponding carbon stocks in applicable pools in vegetation and soil, with land-use change
being the most difficult to address analytically. In this paper we focus on developing and comparing three models, ranging
from relatively simple extrapolations of past trends in land use based on simple drivers such as population growth to more
complex extrapolations of past trends using spatially explicit models of land-use change driven by biophysical and socioeconomic
factors. The three models used for making baseline projections of tropical deforestation at the regional scale are: the Forest
Area Change (FAC) model, the Land Use and Carbon Sequestration (LUCS) model, and the Geographical Modeling (GEOMOD) model.
The models were used to project deforestation in six tropical regions that featured different ecological and socioeconomic
conditions, population dynamics, and uses of the land: (1) northern Belize; (2) Santa Cruz State, Bolivia; (3) Paraná State,
Brazil; (4) Campeche, Mexico; (5) Chiapas, Mexico; and (6) Michoacán, Mexico.
A comparison of all model outputs across all six regions shows that each model produced quite different deforestation baselines.
In general, the simplest FAC model, applied at the national administrative-unit scale, projected the highest amount of forest
loss (four out of six regions) and the LUCS model the least amount of loss (four out of five regions). Based on simulations
of GEOMOD, we found that readily observable physical and biological factors as well as distance to areas of past disturbance
were each about twice as important as either sociological/demographic or economic/infrastructure factors (less observable)
in explaining empirical land-use patterns.
We propose from the lessons learned, a methodology comprised of three main steps and six tasks can be used to begin developing
credible baselines. We also propose that the baselines be projected over a 10-year period because, although projections beyond
10 years are feasible, they are likely to be unrealistic for policy purposes. In the first step, an historic land-use change
and deforestation estimate is made by determining the analytic domain (size of the region relative to the size of proposed
project), obtaining historic data, analyzing candidate baseline drivers, and identifying three to four major drivers. In the
second step, a baseline of where deforestation is likely to occur–a potential land-use change (PLUC) map—is produced using
a spatial model such as GEOMOD that uses the key drivers from step one. Then rates of deforestation are projected over a 10-year
baseline period based on one of the three models. Using the PLUC maps, projected rates of deforestation, and carbon stock
estimates, baseline projections are developed that can be used for project GHG accounting and crediting purposes: The final
step proposes that, at agreed interval (e.g., about 10 years), the baseline assumptions about baseline drivers be re-assessed.
This step reviews the viability of the 10-year baseline in light of changes in one or more key baseline drivers (e.g., new
roads, new communities, new protected area, etc.). The potential land-use change map and estimates of rates of deforestation
could be re-done at the agreed interval, allowing the deforestation rates and changes in spatial drivers to be incorporated
into a defense of the existing baseline, or the derivation of a new baseline projection. 相似文献

Nine amino acids, i.e., alanine, threonine, cysteine, glutamic acid, arginine, proline, tryptophan, phenylalanine and tyrosine, were irradiated with UV light in water containing nitrite or nitrate ion under neutral conditions. The mutagenicities of the ether extracts and the residual water layers of the reaction mixtures were assayed with and without S-9 mix using TA98 and TA100. Three aromatic amino acids, tryptophan, phenylalanine and tyrosine, were found to give direct-acting and frameshift mutagens by irradiation in aqueous nitrite solution. Among them, the ether extract of tryptophan exhibited the strongest mutagenicity toward TA98. In the case of irradiation in aqueous nitrate solution, only the ether extract of tryptophan exhibited weak mutagenicity toward TA98 without S-9 mix. As part of the California Mobile Home Study, over 250 mobile homes from throughout the state were monitored for nitrogen dioxide (NO2) concentrations. Week-long average measurements were taken with Palmes tubes in the kitchen and bedroom of each mobile home during the summer of 1984 and the winter of 1985. The study was conducted entirely by mail with the participants providing all the necessary information. Mobile homes using gas for cooking had significantly higher indoor NO2 levels than those using electricity. Mobile homes located in the Los Angeles basin had significantly higher indoor NO2 concentrations than did mobile homes in the rest of the state. Gas cooking, the inverse of the house volume and geographic location (as a surrogate of outdoor NO2) were the most important variables identified by multiple linear regression. 相似文献

During live fire training exercises, large amounts of explosives are consumed. Low order detonations of high explosive payloads result in the patchy dispersal of particles of high explosive formulations over large areas of firing range soils. Dissolution of explosives from explosive formulation particles into soil pore water is a controlling factor for transport, fate, and effects of explosive compounds. We developed an empirical method to evaluate soils based on functionally defined effective dissolution rates. An automated Accelerated Solvent Extractor was used to determine the effective elution rates under controlled conditions of RDX and TNT from soil columns containing particles of Comp B. Contrived soils containing selected soil geosorbants and reactive surfaces were used to quantitatively determine the importance of these materials. Natural soils from training ranges of various soil types were also evaluated. The effects of geosorbants on effective elution rates were compound- and sorbent-specific. TNT elution was less than that of RDX and was greatly slowed by humic acid. Iron and iron-bearing clays reduced the effective elution rates of both RDX and TNT. This empirical method is a useful tool for directly generating data on the potential for explosives to leach from firing range soils, to identify general bulk soil characteristics that can be used to predict the potential, and to identify means to engineer soil treatments to mitigate potential transport. 相似文献

Heterotrophic activities were investigated by measuring 3H-leucine incorporation to bacterial protein and 14C-glucose turnover in surface flow constructed wetlands receiving woodwaste leachate. No significant longitudinal variation was found in heterotrophic activities of bacterioplankton. An open wetland, a vegetated wetland, and a fertilized vegetated wetland were used to examine the effects of vegetation and ammonium nitrate amendment. There was not a significant difference in treatment performance among the three wetlands, except for a significant pH increase and more efficient volatile fatty acids removal in the fertilized wetland. The fertilized wetland had the highest leucine incorporation rate and shortest glucose turnover time accompanied by the lowest glucose mineralization percentage, followed by the open wetland, then the vegetated wetland. Planktonic and sedimentary bacteria contributed to the majority of the total heterotrophic activities; epiphytic bacteria played a minor role. In an earlier paper examining inherited tolerance to Pb, the shell growth of laboratory-bred offspring of Helix aspersa from contaminated sites was compared with that ofjuveniles from naive populations on dosed and undosed diets. Eight-week-old snails were fed either 500 microg g(-1) Pb or a control food in competitive trials between two populations. In the first series of trials, a parental history of exposure to Pb did not confer any advantage to either of two populations (BI and MI) competing with a na?ve population (LE). whether Pb was present in the diet or not. However, in the analysis of their metal concentrations reported here, LE are found to retain higher levels of Pb in the soft tissues than either BI or MI. Compared to their siblings on the unleaded diet, dosed LE and BI juveniles had lower soft tissue concentrations of Ca and Mg. Although the growth in shell height is unaffected by diet, LE and BI juveniles build lighter shells on the Pb-dosed diet, achieving around 75% of the shell mass of their controls. In contrast, the shell weights of dosed MI juveniles are depressed by only 15% and show no change in the essential metal concentrations of their soft tissues. A second experiment using five populations fed only the dosed food show that the shell weight/soft tissue weight ratios are comparable to the dosed snails of the previous experiment. Building a lighter shell thus appears to be the common response of all Helix populations to a high Pb diet, at least amongst juveniles. The reduction in its mass means that less Ca and Mg is added to the shell and, along with the lowered soft tissue concentrations observed in some populations. may be a consequence of an increased effort to excrete Pb. The possibility that the MI population shows a genotypic adaptation. perhaps as some form of modification of its Ca metabolism, is briefly discussed. 相似文献
