While the World Health Organization 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) equivalency factors are useful estimates of relative potencies of mixtures when conducting risk assessments, they are not useful when comparing the results of bioassays such as the H4IIE-luc to concentrations of TCDD equivalents calculated from instrumental analyses. Since there are thousands of dioxin-like compounds (DLCs), one use of screening assays is to determine if all of the aryl hydrocarbon receptor (AhR) active DLCs in a mixture have been accounted for in instrumental analyses. For this purpose, bioassay-specific relative potency (ReP) values are needed. RePs of 21 polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans, and dioxin-like polychlorinated biphenyls that exhibit effects mediated through the AhR were determined by use of the H4IIE-luc assay. Different values of RePs are derived, depending on the statistical, curve-fitting methods used to derive them from the dose–response relationships. Here, we discuss the various methods for deriving RePs from in vitro data and their assumptions and effects on values of RePs. Full dose–response curves of 2,3,7,8-TCDD and other representative DLCs were used to estimate effective concentrations at multiple points (e.g., EC20-50-80), which were then used to estimate ReP of each DLC to 2,3,7,8-TCDD. 相似文献

Most studies on insect sperm motility have been conducted in vitro using artificial environments outside the animal’s body.
Only little is known about the function of motile insect sperm at different sites within the male or female genital tracts.
We dissected genital tracts of female rove beetles (Drusilla canaliculata) to show that spermatozoa use their own motility to migrate from the spermatophore into the spermatheca. Our dissection method
allowed direct observation and filming of the spermathecal filling process inside the female’s genital tract. Spermatozoa
were found to enter the spermatheca individually, sometimes in groups of two or three. Although exhibiting only weak motility
and no progressive motion in buffer solution, the spermatozoa inside the female show vigorous lashing and reach an average
velocity of 47.5 μm s−1. To gain mobility and speed, the spermatozoa likely utilize the relatively small diameter of the spermathecal duct to push
themselves off the duct walls, rather than swimming freely in seminal fluid. The spermatozoa (approximately 1,250 μm) are
considerably longer than the distance they have to travel along the spermathecal duct (approximately 800 μm). Our study provides
the first direct observation of active sperm migration within the female of an insect stressing the importance of the genital

Gas tracer tests can be used to determine gas flow patterns within landfills, quantify volatile contaminant residence time, and measure water within refuse. While gas chromatography (GC) has been traditionally used to analyze gas tracers in refuse, photoacoustic spectroscopy (PAS) might allow real-time measurements with reduced personnel costs and greater mobility and ease of use. Laboratory and field experiments were conducted to evaluate the efficacy of PAS for conducting gas tracer tests in landfills. Two tracer gases, difluoromethane (DFM) and sulfur hexafluoride (SF6), were measured with a commercial PAS instrument. Relative measurement errors were invariant with tracer concentration but influenced by background gas: errors were 1-3% in landfill gas but 4-5% in air. Two partitioning gas tracer tests were conducted in an aerobic landfill, and limits of detection (LODs) were 3-4 times larger for DFM with PAS versus GC due to temporal changes in background signals. While higher LODs can be compensated by injecting larger tracer mass, changes in background signals increased the uncertainty in measured water saturations by up to 25% over comparable GC methods. PAS has distinct advantages over GC with respect to personnel costs and ease of use, although for field applications GC analyses of select samples are recommended to quantify instrument interferences. 相似文献

Objectives The aim of this study was to evaluate the role of nasal bone assessment in first-trimester screening for Down syndrome (DS) in the Korean population. Methods From July 2004 to March 2006, we prospectively evaluated the fetal nasal bones at 11–14 weeks' gestation in the Korean population. Results A successful evaluation was possible in 6490 of 6787 fetuses (95.6%). Absent, hypoechoic, and short nasal bones were seen in 4 (26.7%), 4 (26.7%), and 1 (6.7%) of 15 fetuses with DS, respectively, whereas in 5 (0.1%), 11 (0.2%), and 246 (3.8%) of 6456 normal fetuses. The incidence of absent and hypoechoic nasal bone showed significant differences between normal fetuses and fetuses with DS (P < 0.0005, both). Screening for DS using an absent or hypoechoic nasal bone resulted in a sensitivity of 53.3%, a specificity of 99.8%, a positive likelihood ratio of 215.2, and a negative likelihood ratio of 0.5. Conclusion Our study showed that nasal bone abnormality at 11–14 weeks of gestation had a high association with DS in the Korean population. TACOM (TAsk COMplexity) is a measure for evaluating the complexity of tasks prescribed for emergency situations in nuclear power plants. Five sub-measures constituting TACOM represent five different aspects of the task complexity exhibited in operating procedures for emergency situations. The practicality of TACOM has been verified through a series of empirical studies. However, tasks designed for abnormal situations that can significantly affect the safety of nuclear power plants, also need a proper measure for evaluating their complexity. TACOM provides a process, a systematic cognitive task analysis method and a set of guidelines to support its application. Therefore, although the characteristics of abnormal task situations are not the same as those of emergency situations, TACOM seems to be reasonably applied to abnormal situations or at least to offer meaningful insights for developing a measure for evaluating the complexity of tasks in abnormal situations. Thus this study examined the applicability of TACOM to abnormal situations through case studies. Particular attention was paid to the sufficiency and appropriateness of the three methodological tools, which are the process, the cognitive task analysis method and the set of guidelines. Collective consideration of the case studies and the characteristics of tasks prescribed for abnormal situations led us to draw the conclusion that TACOM could be reliably used for abnormal situations as well. This paper reports the process of how to apply TACOM to the tasks of abnormal situations and discusses some lessons learned through this application. 相似文献

Together with other variables, human factors play a central role in the safety of highly complex technical systems such as nuclear power plants. However, despite the unquestionable importance of human factors, little information is available about relevant ability requirements for control room jobs in nuclear power plants. The purpose of this study was to close this gap, to provide specific information about ability requirements for such jobs, and to evaluate how several hypothesized factors (ability domain, type of job, and operating condition) contribute to ability requirements. We found that high levels of cognitive as well as social/interpersonal abilities are needed for control room jobs, and that ability requirements increase with the hierarchical job level for these two domains but decrease for psychomotor/physical abilities and for sensory/perceptual abilities. Furthermore, specifically concerning jobs with a leadership function, we found some differences between incidents and normal operations regarding requirements for social/interpersonal abilities, indicating that the former require a different leadership style than the latter. 相似文献
