A simple and fast solid phase extraction procedure was developed to pre-concentrate traces of Cu and Zn prior to their determination in honey samples by flame atomic absorption spectrometry. The sample preparation included dissolution of honey samples and the passage (at 20 ml/min) of resulting 10 % m/v solutions (100 ml) through Dowex 50W?×?8-400 resin beds in order to quantitatively retain Cu and Zn and separate them from the glucose and fructose matrix. Enriched Cu and Zn traces were recovered with 5.0 ml of a 3.0 mol/l HCl solution and quantified by flame atomic absorption spectrometry. The procedure proposed was used to analyze sixty nine commercially available and freshly ripened honey samples coming from the Lower Silesia region (Poland). It enabled to measure Cu and Zn within the range of 0.01–1.42 and 0.03–15.38 μg/g, respectively, with precision better than 4 %. Accuracy, assessed on the basis of the recovery test and the comparison of results with those obtained using wet digestion and inductively coupled plasma optical emission spectrometry, was ranged from ?4 % to +6 %. Detection limits of Cu and Zn achieved with this method were 5 and 7 ng/g, respectively. 相似文献

Edible and medicinal macrofungi used in human diet represent not only important sources of nutritive elements but toxic substances as well (heavy metals and radionuclides). Radioactivity levels of four radionuclides (40K, 137Cs, 226Ra, 228Ra) were determined in the basidiomata (fruiting bodies of a Basidiomycetes) of six lignicolous (Fomitopsis pinicola, Ganoderma applanatum, Hericium clathroides, Megacollybia platyphylla, Pluteus cervinus, Trametes gibbosa) and three mycorrhizal (Boletus luridus, Boletus sp. 1, Boletus sp. 2) species as well as their soil (wood) substrates by gamma spectrometry (high-resolution high-purity germanium (HPGe) detector). The aim was to investigate their ability for radionuclide absorption according to transfer factors (from soil and wood), to predict potential bioindicator species as well as species with potential risk for human use. Samples were taken during years 2011 and 2012, at two sites in forest ecosystem of Tara Mountain (Serbia). Observed concentration ranges per dry weight were as follows: 29–3,020 Bq/kg (40K), 21.9–735 Bq/kg (137Cs), 3–39 Bq/kg (226Ra), and 2.0–18 Bq/kg (228Ra). Obtained results indicate that the type of basidiome (fleshy/tough), most likely due to a different metabolic rate, has a very important role in radionuclide accumulation. The highest activity concentrations of all analyzed radionuclides were found in species with fleshy basidiomata—P. cervinus, H. clathroides, M. platyphylla, and Boletus species. A species-specific influence on radionuclide uptake was more prominent comparing to habitat differences and the role of fungal trophic mode. No significant variations were observed regarding radionuclide activity among the same fungal species from different sampling sites. 相似文献

There is a growing interest for the application of biomakers to field-collected earthworms. Therefore we have evaluated the usability of native populations of endogeic, widely distributed earthworm Aporrectodea caliginosa in the assessment of soil genotoxicity using the Comet assay. Validation of the Comet assay on earthworm coelomocytes has been established using commercially available Eisenia fetida exposed to copper, cadmium, and pentachlorophenol, along with A. caliginosa exposed to copper in a filter paper contact test. Neutral red retention time (NRRT) assay was conducted on copper exposed and field-collected earthworms. Significant DNA and lysosomal damage was measured using Comet and NRRT assays in native populations of A. caliginosa sampled from the polluted soils in the urban area in comparison to the earthworms from the reference site. The results of this study confirm the employment of A. caliginosa as a suitable species for the in situ soil toxicity and genotoxicity field surveys. 相似文献

Kelp and other seaweeds are traditionally used in many parts of the world as a soil amendment on arable fields. Seaweeds contain
biochemical compounds that can act as plant growth regulators in terrestrial plants. In a low-intensity arable grassland in
northwest Scotland an organic fertilizer, kelp (Laminaria digitata) has been used for hundreds of years, due to its anticipated positive effect as a soil conditioner and provider of plant
nutrients. In this study the effects of kelp on germination and rooting of crops and native plants from this area were investigated
in soil-free media. Germination was studied by incubation in the presence of kelp solutions. Rooting of plant cuttings was
assessed after a pulse treatment with kelp solutions, and indole-3 acetic acid (IAA) as a reference plant growth regulator.
Germination percentage of Plantago lanceolata, Trifolium repens and Avena strigosa seeds increased significantly when incubated with 0.05% kelp solutions. Total root weight and the individual weight of roots
produced in cuttings of Vigna radiata and P. lanceolata were significantly increased when exposed to a 0.5% solution of kelp. Plant vigour, assessed visually, decreased significantly
for P. lanceolata exposed to kelp at concentrations of 0.5 and 5.0% indicating the presence of a threshold level for an inhibitory effect of
kelp at these concentrations, which may be due to high salinity. The results confirmed the presence of plant growth regulators
in kelp, and indicates that amendment with kelp may potentially affect plant community composition. The threshold levels where
some plants responded negatively to kelp amendment were close to or lower than the theoretical concentrations of kelp in soil
water at field conditions with the current doses used on the machair, indicating that care should be taken in either administering
kelp at the appropriate dose or leaching out salt before application. 相似文献

The effect of nonuniform alternating fields on neutral or charged particles is named dielectrophoresis in contrast to the action of a uniform steady field on charged particles, named electrophoresis. Dielectrophoresis exhibits e.g. collection of the cells at the electrodes, pearl chain formation everywhere in the experimental chamber, cord formation with ramification, orientation of asymmetric cells, rotation and membrane fusion. In the presented experiments frequencies between 100 Hz and 5 MHz and voltages up to 70 V between electrodes of a few millimeter distance were applied to yeast cells. Dielectrophoresis may offer an interesting biophysical and microbiologic tool for collecting cells and discriminating between living and dead ones, for investigation of influence of drugs on the status of cells, for experiments on membrane or cell fusion and for the study of surface charges in different cells. The key factors that influence the population growth rate can be identified with sensitivity analysis. Caswell presented an algorithm based on vector calculus that speeds up, and improves the practicality of the application of the sensitivity analysis, by calculating the derivatives of the explicit elements from the population Leslie matrix. Despite the usefulness of the sensitivity analysis, it is still barely applied to empirical data, partially due to requirements of high quality demographic data and failure of compatibility between time intervals of field data collection and organism’s life stage timing. This can result in the absence of a certain vital rate as a separated element in the population projection matrix, and thus intricacy in obtaining its sensitivity. In this note we call the attention for the applied value of sensitivity analysis, and also we point to a simple formula that is easily implemented in Caswell Matlab algorithm, to calculate the sensitivity of a vital rate that is not explicit as a matrix element. 相似文献
