Systematic sampling and analysis were performed to investigate the dynamics and the origin of suspended particulate matter smaller than 2.5 μm in diameter (PM(2.5)), in Beijing, China from 2005 to 2008. Identifying the source of PM(2.5) was the main goal of this project, which was funded by the German Research Foundation (DFG). The concentrations of 19 elements, black carbon (BC) and the total mass in 158 weekly PM(2.5) samples were measured. The statistical evaluation of the data from factor analysis (FA) identifies four main sources responsible for PM(2.5) in Beijing: (1) a combination of long-range transport geogenic soil particles, geogenic-like particles from construction sites and the anthropogenic emissions from steel factories; (2) road traffic, industry emissions and domestic heating; (3) local re-suspended soil particles; (4) re-suspended particles from refuse disposal/landfills and uncontrolled dumped waste. Special attention has been paid to seven high concentration "episodes", which were further analyzed by FA, enrichment factor analysis (EF), elemental signatures and backward-trajectory analysis. These results suggest that long-range transport soil particles contribute much to the high concentration of PM(2.5) during dust days. This is supported by mineral analysis which showed a clear imprint of component in PM(2.5). Furthermore, the ratios of Mg/Al have been proved to be a good signature to trace back different source areas. The Pb/Ti ratio allows the distinction between periods of predominant anthropogenic and geogenic sources during high concentration episodes. Backward-trajectory analysis clearly shows the origins of these episodes, which partly corroborate the FA and EF results. In agriculture, occupational injuries are common, and several of them lead to permanent physical disability. The objective of this case study was to assess the strain and the ergonomic needs of four farmers (aged 34–49 years) with physical disabilities. A maximal bicycle ergometer test or an arm-crank test was done to assess their maximal heart rate (HR max) and maximal oxygen consumption (V02max). The strain at work was analyzed by measuring heart rate (HR), muscle activity (EMG), and the rating of perceived exertion (RPE). The farmers were interviewed as to possible and impossible work tasks and the ergonomic redesign measures taken to improve the work environment. The work tasks performed were mainly light or moderate work for the cardiorespiratory system according to mean HR (88-102 beats/min), the percentage of HR range (17–31% HRR), and the relative V02 (22–46% V02max). The mean activity of the trapezius muscles was 0.4-9% of the maximal voluntary contraction (%MVC). All the participants had work tasks they were unable to perform. They had made ergonomie redesign changes mainly to the tractor. This case study showed that some agricultural work tasks were possible for farmers with physical disabilities and that the physical strain associated with these tasks was mainly light or moderate. 相似文献

Toxicity of two azo dyes (Reactive Orange 16 (RO16); Congo Red (CR)) and two anthraquinone dyes (Remazol Brilliant Blue R (RBBR); Disperse Blue 3 (DB3)) were compared using bacterium Vibrio fischeri, microalga Selenastrum capricornutum and ciliate Tetrahymena pyriformis. The following respective endpoints were involved: acute toxicity measured as bacterial luminescence inhibition, algal growth inhibition, and the effects on the protozoa including viability, growth inhibition, grazing effect and morphometric effects. In addition, mutagenicity of the dyes was determined using Ames test with bacterium Salmonella typhimurium His(-). DB3 dye was the most toxic of all dyes in the bacterial, algal and protozoan tests. In contrast to other dyes, DB3 exhibited mutagenic effects after metabolic activation in vitro in all S. typhimurium strains used. Of the methods applied, the algal test was the most sensitive to evaluate toxicity of the dyes tested. 相似文献

A new heterogeneous photocatalyst was prepared by oxidative polymerization of the thiophene with ferric chloride in the ZSM-5 zeolite type. The synthesized polythiophene absorbs radiation in the visible range of the electromagnetic spectrum and by illumination with visible light generates reactive oxygen species (ROS) in water medium. During illumination reactive hydroxyl radical was detected by the spin trapping EPR method. A novel bioassay with the human adrenocortical carcinoma cell line H295R can be used to screen for endocrine disrupting chemicals that affect the expression of genes important in steroidogenesis. This assay was employed to study the effects of organic contaminants associated with the freshwater pond sediments collected in the Ostrava-Karvina region, Czech Republic. The modulation of ten major genes involved in the synthesis of steroid hormones (CYP11A, CYP11B2, CYP17, CYP19, 17betaHSD1, 17betaHSD4, CYP21, 3betaHSD2, HMGR, StAR) after exposure of H295R cells to sediment extracts was investigated using quantitative real-time polymerase chain reaction (PCR). Crude sediment extracts, containing high concentrations of polycyclic aromatic hydrocarbons (PAHs) and moderate amounts of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) significantly stimulated expression of the CYP11B2 gene (up to 10-fold induction), and suppressed expression of 3betaHSD2 and CYP21 genes. A similar pattern was observed with the extracts after treatment with concentrated sulfuric acid to remove labile chemicals (including PAHs) leaving only persistent PCBs, OCPs and potentially PCDD/Fs. Comparison of the results with other mechanistically based bioassays (arylhydrocarbon receptor, AhR, mediated responses in H4IIE-luc cells, and estrogen receptor mediated effects in MVLN cells) revealed significant endocrine disrupting potencies of organic contaminants present in the sediments (most likely antiestrogenicity). Pronounced effects were observed particularly in sediment extracts from the Pilnok Pond which harbors an unusual intersexual population of the narrow-cawed crayfish Pontastacus leptodactylus (Decapoda, Crustacea). This pilot study provided the first experimental evidence of the wider application of the H295R bioassay for screening complex environmental samples, and the results support the hypothesis of chemical-induced endocrine disruption in intersexual crayfish. 相似文献

The (90)Sr and (137)Cs uptake by the plant Helianthus annuus L. was studied during cultivation in a hydroponic medium. The accumulation of radioactivity in plants was measured after 2, 4, 8, 16 and 32 days of cultivation. About 12% of (137)Cs and 20% of (90)Sr accumulated during the experiments. We did not find any differences between the uptake of radioactive and stable caesium and strontium isotopes. Radioactivity distribution within the plant was determined by autoradiography. (137)Cs was present mainly in nodal segments, leaf veins and young leaves. High activity of (90)Sr was localized in leaf veins, stem, central root and stomata. The influence of stable elements or analogues on the transfer behaviour was investigated. The percentage of non-active caesium and strontium concentration in plants decreased with the increasing initial concentration of Cs or Sr in the medium. The percentage of (90)Sr activity in plants decreased with increasing initial activity of the nuclide in the medium, but the activity of (137)Cs in plants increased. The influence of K(+) and NH(4)(+) on the uptake of (137)Cs and the influence of Ca(2+) on the uptake of (90)Sr was tested. The highest accumulation of (137)Cs (24-27% of the initial activity of (137)Cs) was found in the presence of 10 mM potassium and 12 mM ammonium ions. Accumulation of about 22% of initial activity of (90)Sr was determined in plants grown on the medium with 8 mM calcium ions. 相似文献
