Bacterial strain RV9 recovered from greengram nodules tolerated 2400 μg/mL of hexaconazole and was identified by 16 S rDNA sequence analysis as Bradyrhizobium japonicum(KY940048). Strain RV9 produced IAA(61.6 μg/mL), ACC deaminase(51.7 mg/(protein·hr)), solubilized TCP(105 μg/mL), secreted 337.6 μg/mL EPS, and produced SA(52.2 μg/mL) and 2,3-DHBA(28.3 μg/mL). Exopolysaccharides produced by strain RV9 was quantified and characterized by SEM, AFM, EDX and FTIR. Beyond tolerance limit,hexaconazole caused cellular impairment and reduced the viability of strain RV9 revealed by SEM and CLSM. Hexaconazole distorted the root tips and altered nodule structure leading thereby to reduction in the performance of greengram. Also, the level of antioxidant enzymes, proline, TBARS, ROS and cell death was increased in hexaconazole treated plants.CLSM images revealed a concentration dependent increase in the characteristic green and blue fluorescence of hexaconazole treated roots. The application of B. japonicum strain RV9 alleviated the fungicide toxicity and improved the measured plant characteristics. Also,rhizobial cells were localized inside tissues as revealed by CLSM. Colonization of B.japonicum strain RV9 decreased the levels of CAT, POD, APX, GPX and TBARS by 80%, 5%,13%, 13% and 19%, respectively over plants grown at 80 μg/(hexaconazole·kg) soil. The ability to detoxify hexaconazole, colonize plant tissues, secrete PGP bioactive molecules even under fungicide pressure and its unique ability to diminish oxidative stress make B.japonicum an attractive choice for remediation of fungicide polluted soils and to concurrently enhance greengram production under stressed environment. 相似文献

Endosulfan is an organochlorine pesticide that is toxic to aquatic life. Endosulfan might hamper the reproductive health of indigenous fish in agricultural areas of Pakistan where this pesticide is sprayed widely. The aim of the current study is to investigate the toxic effects of endosulfan on selected reproductive parameters of male freshwater fish, Cyprinion watsoni. Two concentrations of endosulfan (0.5 and 1 ppb for 30 days exposure) were tested for their effects on body weight, body length, and testicular weight, length, and width. Testicular testosterone was assayed from tissue extracts using enzyme immunoassay (EIA). A significant increase in the mortality rate was observed in both treated groups during both spawning and quiescent seasons. The overall behavior of fish in the aquarium was normal in all control and treated groups. However, the treated fish exhibited anxiety after treatment with endosulfan. The body weight and length, and testicular weight, length and width were not significantly different to the control group. The testicular testosterone concentrations were significantly lower in both endosulfan-treated groups compared to the control. The decrease was dose-dependent, with a significant difference between the two treated groups. The histomorphological results demonstrated various testicular alterations in the treated groups. These alterations included an increase in interlobular areas and clumping patterns in spermatocytes/spermatids. Because spermatids eventually differentiate into sperms, their low count will directly result in lower sperm count. Taken together, these results suggest that endosulfan is a toxicant that at least disturbs testosterone levels (possibly others) and negatively impacts the reproductive health of male freshwater fish. 相似文献

The drinking water of Naranji village and surrounding areas of the NorthWest Frontier Province (NWFP), Pakistan, was analyzed for its fluoride content. The fluoride content of water samples from the springs in the Naranji village goes up to 13.52 mg kg(-1), which exceeds the permissible limit (1.5 mg kg(-1)) set by the World Health Organization (WHO). This study explores the source of fluoride contamination and attributes it to the alkaline rocks of the Ambela granitic complex and the Koga complex. The low-lying areas towards the south have a fluoride content within the permissible limit. The Naranji area therefore needs urgent remedial measures. 相似文献

ABSTRACT: This paper presents a simple methodology, using the entropy concept, to estimate regional hydro logic uncertainty and information at both gaged and ungaged grids in a basin. The methodology described in this paper is applicable for (a) the selection of the optimum station from a dense network, using maximization of information transmission criteria, and (b) expansion of a network using data from an existing sparse network by means of the information interpolation concept and identification of the zones from minimum hydrologic information. The computation of single and joint entropy terms used in the above two cases depends upon single and multivariable probability density functions. In this paper, these terms are derived for the gamma distribution. The derived formulation for optimum hydrologic network design was tested using the data from a network of 29 rain gages on Sleeper River Experimental Watershed. For the purpose of network reduction, the watershed was divided into three subregions, and the optimum stations and their locations in each subregion were identified. To apply the network expansion methodology, only the network consisting of 13 stations was used, and feasible triangular elements were formed by joining the stations. Hydrologic information was calculated at various points on the line segments, and critical information zones were identified by plotting information contours. The entropy concept used in this paper, although derived for single and bivaviate gamma distribution, is general in type and can easily be modified for other distributions by a simple variable transformation criterion. 相似文献

Chemically modified pullulan was evaluated for its sorption efficiency and selectivity to remove cadmium(Cd) from spiked high-hardness groundwater(GW). Pullulan esterified with succinic anhydride using dimethylaminopyridine showed a fairly high degree of substitution value as confirmed by1 H NMR spectroscopy. Pullulan succinate(Pull-Suc) was converted into the sodium salt(Pull-Suc-Na). The effect of contact time(5–200 min) and p H(2–8) on Cd-uptake by the sorbent(Pull-Suc-Na) was investigated. The sorbent showed more than 90% Cd-removal in first 15 min from distilled water(DW) and GW solution,respectively. Comparison of Pull-Suc-Na with other polysaccharidal sorbents suggested its high efficiency(DW 476.2 mg/g and GW 454.5 mg/g) and selectivity for the removal of Cd by an ion exchange mechanism, which is further supported by the negative Gibbs free energy values calculated from Langmuir isotherms. A Langmuir isotherm kinetic model provided the best fit for the sorption of Cd using Pull-Suc-Na. The sorbent showed a negligible decrease in Cd-uptake over three regeneration cycles. The thermal stability testing of the sorbents indicated that Pull-Suc-Na(sorbent) is more stable than Pull-Suc. 相似文献

Co-composting of poultry manure and rubber wood sawdust was performed with the ratio of 2:1 (V/V) for a period of 60 days. An investigation was carried out to study the extracellular enzymatic activities and structural degradation utilizing Fourier transform infrared spectroscopy (FT-IR), thermogravimetry and differential thermal analysis (TG/DTA) and scanning electron microscopy (SEM). The microbial succession was also determined by using denaturing gel gradient electrophoresis (DGGE). The compost was able to reach its highest temperature of 71°C at day 3 and stabilized between 30 and 40°C for 8 weeks. CMCase, FPase and β-glucosidase acted synergistically in order to degrade the cellulosic substrate. The xylanase activities increased gradually during the composting and reached the peak value of 11.637 U/g on day 35, followed by a sharp decline. Both LiP and MnP activities reached their peak values on day 35 with 0.431 and 0.132 U/g respectively. The FT-IR spectra revealed an increase in aromaticity and a decrease in aliphatic compounds such as carbohydrates as decomposition proceeded. TGA/DTG data exhibited significant changes in weight loss in compost samples, indicating degradation of organic matter. SEM micrographs showed higher amounts of parenchyma exposed on the surface of rubber wood sawdust at day 60, showing significant degradation. DGGE and 16S rDNA analyses showed that Burkholderia sp., Pandoraea sp., and Pseudomonas sp. were present throughout the composting process. Ornithinibacillus sp. and Castellaniella ginsengisoli were only found in the initial stage of the composting, while different strains of Burkholderia sp. also occurred in the later stage of composting. 相似文献

This study was aimed to establish background and reference values of total heavy metals in soils from a representative area of Albania (Tirana). Thirty-eight soil samples collected from genetic horizons of major soil types of Tirana were analyzed for important physicochemical properties by standard methods and for total contents of Cd, Cr, Ni, Pb, Zn, and Cu by atomic absorption spectrometer, after extraction with aqua regia. The results showed that the total contents of Cd, Cr, Ni, Pb, Zn, and Cu in surface horizons varied widely with respective mean values of 0.3 (??0.6), 174.2 (??63.7), 305.9 (??133.0), 19.7 (??12.4), 95.5 (??26.3), and 42.7 (??6.8) mg/kg. The highest metal contents were found in two soils developed in limestone. The depth distribution of metals showed a tendency for accumulation of Cd and Pb in the surface horizons of three soils, suggesting that these metals partially come from anthropogenic inputs. Correlation analysis indicated that the metal contents of soils were controlled by soil properties, including pH, CaCO3, clay, organic matter, cation exchange capacity, and Fe oxides. The background values (given as the 90th percentile) were much higher than those reported in the literature, showing that the levels of respective metals were naturally higher. The total metal contents of some soils were above background levels, suggesting metal pollution. The reference values for all the analyzed metals were quite consistent with those of the Dutch system. The proposed background and reference values can be used to evaluate the soil pollution with these elements. 相似文献
