The storage of nitrate by phytoplankton cells during the early phases of upwelling was studied in coastal stations off northern
Spain (southern Bay of Biscay) between 1990 and 1994. In this region, a persistent upwelling during summer is characterised
by intermittent pulses of variable intensity, and increased nutrient concentrations in the surface layer. The main effect
of an upwelling pulse on phytoplankton distribution is the shifting of the chlorophyll a and primary production maxima to near the surface. When the upwelling relaxes, thermal stratification of the water column
occurs, and a distinct subsurface chlorophyll maximum develops below the production maximum. An accumulation of intracellular
nitrate characterized the early phases of upwelling (mean = 2.73 μmol N m−3), maximum concentrations being attained at depths where biomass and production values were moderate. In contrast, phytoplankton
cells from non-upwelling situations contained significantly lower concentrations of intracellular nitrate (mean = 0.17 μmol
N m−3). The variations in the intracellular pool of nitrate may result from the differential allocation of resources within the
cell as a result of variations in the energy available, since the uptake and assimilation of nitrate is a relatively expensive
process involving several enzymatic systems. We hypothesize that nitrate storage by phytoplankton cells is characteristic
of early phases of upwelling and is linked to patterns of carbon fixation. Average nitrogen budgets for upwelling and non-upwelling
situations indicate that intracellular nitrate reserves are not responsible for maintaining high phytoplankton growth rates,
since they only account for <2% of daily primary production during upwelling events.

A study has been made of the leaching of Cd, Zn, Pb, Cu, Tl and As in three representative soils of the zone affected by the spill from a pyrite mine in Aznalcollar (Sevilla, Spain) employing packed soil columns. According to the breakthrough and cumulative leaching curves, the relative mobilities of the different toxic elements in the columns are as follows: Tl > Cd > Zn > As > Cu > Pb. The results also show that the soils themselves have a good capacity for immobilizing the soluble fraction of the elements from the spilled mud, this capacity varying as follows: clayey soil with a high carbonate content > clayey soil with a moderate carbonate content > sandy‐clay loam soil with a low carbonate content. Given the wide industrial use of chromium (Cr) and its environmental contamination, chromium represents a risk to humans exposed to the metal. Considering that Cr(VI) is a potent oxidizing agent that increases intracellular oxidation and DNA damage, it would be worth considering the pretreatment of cells with antioxidants as a means of preventing Cr(VI)-induced toxicity. The objective of this study was to pretreat yeast cells with the water-soluble vitamin E analogue Trolox in an effort to increase cell tolerance against reactive chromium and reactive oxygen species formed during Cr(VI) reduction. Results revealed a decrease in Cr(VI)-induced cytotoxicity and mitotic gene conversions in Trolox-pretreated cells. The protective effect of Trolox in Cr(VI) induced genotoxicity was confirmed also with the prokaryotic Salmonella typhimurium SOS/umu test. Pretreatment of cells with Trolox (1) increased total Cr bioaccumulation, (2) decreased Cr(VI)-induced intracellular oxidation, (3) decreased Cr(V) persistence and (4) increased OH? formation in yeast extracts. These findings might be useful in directing future investigations concerning the use of Trolox as a human antioxidant supplement, and in clinical applications related to Cr-induced genotoxicity in occupational and environmental situations where chromium is a problem. 相似文献

Photosynthetic sulfur bacteria oxidize the sulfide produced by dissimilatory sulfate-reducing bacteria, thus preventing the occurrence of pollution by these compounds in the oxygenic zone of aquatic systems. Zinc is one of the most abundant heavy metals. Its toxic effects have been documented in various organisms that can photosynthesize, but a lack of information prevails in this respect, about photosynthetic sulfur bacteria. The aim of the present study was to evaluate the toxic effects of zinc on green sulfur bacteria (Chlorobiaceae) and purple sulfur bacteria (Chromatiaceae) in a microcosm as well to explore the possible relationships between the bacterial population growths with the physicochemical properties of the metalimnion of Zimapan Reservoir (Mexico) during a 1-year period. Hydrosoluble organic carbon (HOC) and light availability in the metalimnion were the limiting factors for growth in these bacteria. Higher HOC levels occurred in Spring, at the end of the wet season and in Winter. Current zinc levels in the reservoir seem to elicit toxic effects. At lethal zinc concentrations, the best concentration-response relationship was shown. The biomarker for purple sulfur bacteria was the bacteriochlorophyll content, while total reducing sugar was the best biomarker for green sulfur bacteria. At sublethal concentrations, zinc alters adenosine triphosphate levels, total reducing sugar, total protein content, and bacteriochlorophyll content. The observed responses indicate that green sulfur bacteria are more sensitive than purple sulfur bacteria and are suitable target organisms for use in establishing the accepted threshold concentration of zinc (NOEC 0.056–0.097 mg L?1 of Zn) in anoxic deep water. 相似文献

We determined the normal levels of butyrylcholinesterase (BChE), carboxylesterase (CbE), and glutathione S-transferases (GST) activities in three South American toad species in order to establish reference values for field pesticide monitoring purposes. Interspecies variations in B-esterase and GST activities were examined according to body mass. In addition, comparative inhibition of BChE and CbE activities using malaoxon, and chemical reactivation of malaoxon-inhibited BChE activity using pyridine-2-aldoxime methochloride (2-PAM) were investigated. Bufo fernandezae had average activity values for BChE: 17.31 mmol min?1 ml?1; CbE: 621.49 nmol min?1 ml?1 and GST: 1.94 mmol min?1 ml?1 while B. arenarum enzymatic average activities were BChE: 9.51 mmol min?1 ml?1; CbE: 270.07 nmol min?1 ml?1, and GST: 1.59 mmol min?1 ml?1; finally Bufo schneideri had enzymatic mean values of BChE: 2.08 mmol min?1 ml?1; CbE: 301.95 nmol min?1 ml?1, and GST: 1.60 mmol min?1 ml?1. Moreover, we found an allometric relationship between plasma BChE and CbE activities and body size for the three toad species. We suggest that B. fernandezae would be the species with a higher tolerance capacity to organophosphorous insecticides compared to the other toad species, while B. schneideri may be the most vulnerable toad species to field pesticide exposure, although some other factors (e.g., brain AChE sensitivity or pesticide metabolism by phosphotriesterases) should be also taken into account. The malaoxon-inhibited BChE activity of the three toad species was reactivated in the presence of 2-PAM, and it is recommended as a specific and sensitive methodology in the assessment of field exposure to OP insecticides together to compare BChE activity levels between OP-exposed and nonexposed individuals. 相似文献

An automated method for the indirect determination of fluoride by inductively coupled plasma atomic emission spectrometry using discontinuous-flow analysis has been developed. A modified flow injection analysis system was used for sample preparation. The method is based on the determination of excess cerium(III) ion after precipitation of CeF3. Separation of the precipitate is performed online on a syringe membrane filter that is cleaned each analysis cycle by 1 M nitric acid, ensuring a long life-time for the filter. As a result of the short reaction time, a detection limit of 9.5 mg L?1 was obtained. The precision (RSD) of the proposed method is 1.02% at fluoride concentration of 28 mg L?1. The applicability of the technique for groundwater monitoring of fluoride-contaminated area is demonstrated. 相似文献

The interaction between 33 commercial pesticides with hydroxypropyI‐β‐cyclodextrin (HPBCD) was studied by charge‐transfer reversed‐phase thin‐layer chromatography using aqueous sodium chloride solutions as eluents. Each pesticide interacted with HPBCD, their lipophilicity linearly decreased with increasing HPBCD concentration, the pesticide ‐ HPBCD complex (probably inclusion complex) always being more hydrophilic than the uncomplexed pesticide. In many cases the retention of pesticide increased with increasing concentration of salt in the eluent. This effect can be tentatively explained by the suppression of the dissociation of the polar groups in the solute molecules resulting in increased apparent lipophilicity. No significant correlation was found between lipophilicity and complex stability or between salting‐out effect and lipophilicity, that is other than hydrophobic forces are involved in the pesticide ‐ HPBCD interaction. 相似文献
