Biochemical and genetic studies of xenobiotic metabolism in the model plant Arabidopsis have significant potential in providing information for phytoremediation. This paper presents the toxicity of 2,6-dinitrotoluene (2,6-DNT) to Arabidopsis under axenic conditions, the fate and transformation of 2,6-DNT after uptake by the plant, and the effect of a putative glutathione S-transferase (GST), which is highly induced by 2,4,6-trinitrotoluene (TNT) in the previous study, on the detoxification of 2,6-DNT. 2,6-DNT had toxic effects on the growth of Arabidopsis based on whole seedling as well as root growth assays. Using [U- 14C]2,6-DNT, the recovery was over 87% and less than 2% accounted for the mineralization of 2,6-DNT in axenic liquid cultures during the 14d of exposure. About half (48.3%) of the intracellular radioactivity was located in the root tissues in non-sterile hydroponic cultures. 2-Amino-6-nitrotoluene (2A6NT) and two unknown metabolites were produced as transformation products of 2,6-DNT in the liquid media. The metabolites were further characterized by proton NMR spectra and the UV-chromatograms when the plant was fed with either 2,6-DNT or 2A6NT. In addition, polar unknown metabolites were detected at short retention times from radiochromatograms of plant tissue extracts. The GST gene of the wild-type of Arabidopsis in response to 2,6-DNT was induced by 4.7-fold. However, the uptake rates and the tolerance at different concentrations of 2,6-DNT and TNT were not significantly different between the wild-type and the gst mutant indicating that induction of the GST gene is not related to the detoxification of 2,6-DNT. 相似文献

The objective of this research is to characterize the variability of trace metals in the Rio Santa watershed based on synoptic sampling applied at a large scale. To that end, we propose a combination of methods based on the collection of water, suspended sediments, and riverbed sediments at different points of the watershed within a very limited period. Forty points within the Rio Santa watershed were sampled between June 21 and July 8, 2013. Forty water samples, 36 suspended sediments, and 34 riverbed sediments were analyzed for seven trace metals. The results, which were normalized using the USEPA guideline for water and sediments, show that the Rio Santa water exhibits Mn concentrations higher than the guideline at more than 50% of the sampling points. As is the second highest contaminating element in the water, with approximately 10% of the samples containing concentrations above the guideline. Sediments collected in the Rio Santa riverbed were heavily contaminated by at least four of the tested elements at nearly 85% of the sample points, with As presenting the highest normalized concentration, at more than ten times the guideline. As, Cd, Fe, Pb, and Zn present similar concentration trends in the sediment all along the Rio Santa.The findings indicate that care should be taken in using the Rio Santa water and sediments for purposes that could affect the health of humans or the ecosystem. The situation is worse in some tributaries in the southern part of the watershed that host both active and abandoned mines and ore-processing plants. 相似文献

Tools for estimating pollen dispersal and the resulting gene flow are necessary to assess the risk of gene flow from genetically modified (GM) to conventional fields, and to quantify the effectiveness of measures that may prevent such gene flow. A mechanistic simulation model is presented and used to simulate pollen dispersal by wind in different agricultural scenarios over realistic pollination periods. The relative importance of landscape-related variables such as isolation distance, topography, spatial configuration of the fields, GM field size and barrier, and environmental variation are examined in order to find ways to minimize gene flow and to detect possible risk factors. The simulations demonstrated a large variation in pollen dispersal and in the predicted amount of contamination between different pollination periods. This was largely due to variation in vertical wind. As this variation in wind conditions is difficult to control through management measures, it should be carefully considered when estimating the risk of gene flow from GM crops. On average, the predicted level of gene flow decreased with increasing isolation distance and with increasing depth of the conventional field, and increased with increasing GM field size. Therefore, at a national scale and over the long term these landscape properties should be accounted for when setting regulations for controlling gene flow. However, at the level of an individual field the level of gene flow may be dominated by uncontrollable variation. Due to the sensitivity of pollen dispersal to the wind, we conclude that gene flow cannot be summarized only by the mean contamination; information about the frequency of extreme events should also be considered. The modeling approach described in this paper offers a way to predict and compare pollen dispersal and gene flow in varying environmental conditions, and to assess the effectiveness of different management measures. 相似文献

A series of Large Eddy Simulations (LES) are performed to investigate the penetration of starting buoyant jets. The LES code
is first validated by comparing simulation results with existing experimental data for both steady and starting pure jets
and lazy plumes. The centerline decay and the growth rate of the velocity and concentration fields for steady jets and plumes,
as well as the simulated transient penetration rate of a starting pure jet and a starting lazy plume, are found to compare
well with the experiments. After validation, the LES code is used to study the penetration of starting buoyant jets with three
different Reynolds numbers from 2000 to 3000, and with a wide range of buoyancy fluxes from pure jets to lazy plumes. The
penetration rate is found to increase with an increasing buoyancy flux. It is also observed that, in the initial Period of
Flow Development, the two penetrative mechanisms driven by the initial buoyancy and momentum fluxes are uncoupled; therefore
the total penetration rate can be resolved as the linear addition of these two effects. A fitting equation is proposed to

The katydid Neoconocephalus triops exhibits in North America substantial developmental plasticity of male mating calls. The AM rate of the summer calls is significantly
faster than that of the winter calls at the same temperature. In the tropics, where N. triops originated, males express only the fast summer-call phenotype. We tested two alternative hypotheses: (1) call plasticity
in the population from North America evolved in response to selection by female preference after N. triops colonized North America, or (2) call plasticity evolved before N. triops expanded into North America and its expression in the novel environment led to adaptive change of female preferences. First,
we tested whether call plasticity was present in tropical populations of N. triops. Tropical males expressed the winter-call phenotype when reared under winter conditions, indicating that call plasticity
did not evolve in response to temperate climates. Second, we compared female preferences among temperate and tropical populations.
We found that the temperature dependence of preferred AM rate was significantly steeper in temperate N. triops than in a tropical population of N. triops. Third, we compared temperature dependence of female preference of the N. triops populations to three Neoconocephalus species without call plasticity. Only temperate N. triops had significantly steeper temperature dependence than the other species. This steeper temperature dependence matched female
preference to the fast summer call at high temperatures and to the slow winter call at low temperatures in temperate populations.
These results support the hypothesis that female preference changed in N. triops in North America to compensate for the plasticity of male calls. 相似文献

The desorption of 20 chlorinated organics from sediments has been studied using a nitrogen purge/Tenax trap system for separating the “dissolved” and “sorbed” fractions in sediment/water slurries. The desorption partition coefficient, KD, was found to decrease with increasing temperature and suspended sediment concentration. While some differences in KD and desorption rates were observed for the study chemicals, considering their wide range of physical/chemical properties such as KOW, these changes were small. Desorption half-lives averaged about 60d at 4°C, 40d at 20°C and 10d at 40°C under continuous gaseous purging. Estimates of the loadings of chemicals via desorption from bottom sediments in Lake Ontario are compared to loadings of these chemicals to the lake from the Niagara River. 相似文献

The Terai-Duar savanna and grasslands, which once extended along most of the Himalayan foothills, now only remain in a number
of protected areas. Within these localities, grassland burning is a major issue, but data on frequency and distribution of
fires are limited. Here, we analysed the incidence of active fires, which only occur during the dry season (Nov.–Mar.), within
a significant area of Terai grasslands: the Manas National Park (MNP), India. We obtained locations of 781 fires during the
2000–2008 dry seasons, from the Fire Information for Resource Management System (FIRMS) that delivers global MODIS hotspot/fire
locations using remote sensing and GIS technologies. Annual number of fires rose significantly from around 20 at the start
of the study period to over 90 after 2002, with most (85%) detected between December and January. Over half of the fires occurred
in tall grasslands, but fire density was highest in wetland and riverine vegetation, dry at the time. Most burning took place
near rivers, roads and the park boundary, suggesting anthropogenic origins. A kernel density map of all recorded fires indicated
three heavily burnt areas in the MNP, all within the tall grasslands. Our study demonstrates, despite some technical caveats
linked to fire detection technology, which is improving, that remote fire data can be a practical tool in understanding fire
concentration and burning temporal patterns in highly vulnerable habitats, useful in guiding management. 相似文献
