为了解盐生杜氏藻(Dunaliella salina)主要光捕获蛋白LHCB蛋白的功能,将已获得的盐藻Lhcb3基因构建到原核表达载体pET32a-DsLhcb3,通过优化表达条件,建立了高效的重组系统.pET32a-DsLhcb3在大肠杆菌中的优化表达条件为1 mmol/L IPTG在37℃下诱导4 h.采用镍离子亲合层析纯化获得LHCB3蛋白,并以此为抗原制备了多克隆抗体,经琼脂糖扩散检测效价,在1:16处有明显沉淀.提取盐藻总蛋白,经过制备的LHCB3抗体杂交,在29 000处获得两条明显的杂交条带,为进一步研究盐藻LHCⅡ蛋白表达机理奠定了基础. 相似文献

紫叶李叶片色素含量及叶绿素荧光动力学参数对SO2胁迫的响应 总被引:1,自引:0,他引:1
以盆栽紫叶李的2年生嫁接苗为试材,采用人工静态熏气装置分析了不同浓度SO2(0~22.8 mg·m-3)胁迫条件下(16d)紫叶李叶片光合色素含量及荧光动力学参数的变化情况.研究结果显示:在SO2浓度为5.7Mg·m-3的胁迫下,叶绿素a(Chla)、叶绿素b(Chlb)、Chla/Chlb、总叶绿素和类胡萝卜素(Car)含量均比对照增加,在其它浓度处理下均减小,且与对照间差异显著(11.4mg-3m胁迫下叶绿素a和类胡萝卜素除外).在SO2浓度小于11.4mg·m-3时,紫叶李叶片花青苷和类黄酮含量随着胁迫强度的增强而增加(与对照间差异显著,P<0.01);而在SO2浓度大于11.4rag·m-3时则随着胁迫强度的增强而降低(与对照间差异显著,P<0.01).荧光动力学参数中,PSⅡ原初光能转化效率Fv/Fm随着胁迫强度的增强呈现先增加后降低的变化趋势,PSlI潜在活性Fv/F.、光化学淬灭效率qP随着胁迫强度的增强而减小,而荧光非光化学淬灭效率gN 却随着胁迫强度的增强而增加.研究结果表明,紫叶李苗木在低浓度SO2 (小于11.4mg·m-3)胁迫下通过自身较强的保护系统使机体免受伤害;而在高浓度SO2(大于11.4rag·m-3)胁迫下通过增加qN耗散过剩的光能和提高qbPSⅡ消耗多余能量来保护光合器官免受损伤,但叶片的光合色素含量和其它荧光参数均受到不同程度的影响,紫叶李叶片受到伤害与PsⅡ反应中心失活有关. 相似文献

为开发微生物油脂资源,从富含油脂的土样中分离获得一株产油脂微生物——深黄伞形霉(Umbelopsisisabellina)华2-1,油脂含量达48.60%.采用单因素和正交实验方法,对深黄伞形霉华2-1的发酵培养条件进行优化研究.优化的培养条件为:葡萄糖100 g/L,酵母粉3 g/L,接种量为20%,初始pH值为5.0~6.0,MgSO4.7H2O和KH2PO4的添加浓度分别为0.5 g/L和2 g/L,培养温度为31℃,最佳发酵培养时间为168 h.华2-1在优化发酵条件下可获得菌体生物量、油脂产量和油脂含量分别为45.86 g/L、24.47 g/L和59.53%,较优化前分别提高了21.48%、33.42%和22.49%.因此,深黄伞形霉华2-1作为微生物油脂生产的新资源具有广阔的应用前景. 相似文献

以Al2O3负载Cu2O为催化剂,研究芴酮废水在臭氧条件下的氧化降解反应,为芴酮废水的治理提供一种新的处理方法。研究表明,Al2O3负载Cu2O的催化剂加速了臭氧氧化反应,使芴酮废水的氧化降解加快。影响芴酮废水氧化降解的主要因素有臭氧流量、废水的pH值、催化剂用量以及反应器的高度等。加大臭氧流量及增加催化剂用量,均有利于芴酮废水的降解。处理3 L芴酮废水时适宜的反应条件:臭氧流量为25 mg/h,催化剂用量为20 g,反应器的高度H为1 600 mm,废水初始pH为11。在该条件下1.75 h,废水的脱色率为91.2%,COD去除率为90.5%。 相似文献

Adsorption enhancement of laterally interacting phenol/aniline mixtures onto nonpolar adsorbents 总被引:1,自引:0,他引:1
Adsorption equilibria of phenol and aniline onto nonpolar macroreticular adsorbents were investigated in single and binary-solute aqueous systems at 293 K and 313 K. All adsorption isotherms can be well represented by the Langmuir equation. Larger uptake of aniline than phenol onto all the adsorbents probably results from the higher hydrophobicity of the former compound as well as the greater electronic density of the aromatic ring of aniline. It is interestingly observed that at a relatively high loading, the total uptake of phenol and aniline in a binary system is remarkably higher than those in a single system. Such uptake difference was elucidated by the cooperative effect arising from the lateral acid–base interaction between the loaded phenol and aniline molecules. Moreover, larger average pore size of the adsorbent is found to result in a greater cooperative coefficient, as observed from the equimolar phenol/aniline adsorption system. 相似文献

利用傅立叶变换红外光谱仪,固体制样方式,对北京市中关村地区的气溶胶样品中的无机组成进行了分析,并对气溶胶样品中的SO4^2-,NH4^+和NO3^-进行了定量测定,为检验FTIR测定法的可靠性,将FTIR法与离子色谱的测定结果进行了对照分析,结果表明,SO4^2-和NH4^的FTIR测定结果与离子色谱有较好的相关性。 相似文献
