A series of miscible-displacement column experiments were conducted under saturated flow conditions to systematically investigate the influence of physical and biological complexity on bacterial activity and fate in the presence and absence of a non-sorbing growth substrate, salicylate. Bacterial elution was monitored for three different systems; System I--a sterilized, inoculated, well-sorted sand, System II--a sterilized, inoculated, heterogeneous loamy sand (Hayhook), and System III--two different unsterilized loamy sands (Hayhook and Vinton) each with their associated indigenous microbial community. Results show that System I behaved ideally with respect to both cell and substrate transport, wherein: (1) growth occurred in response to substrate addition, (2) cell elution increased in response to the substrate pulse, and (3) breakthrough curves were reproducible for both substrate and cell elution. In contrast, System II showed ideal behavior with respect to substrate transport but showed variable behavior for cell transport. Further, there was no measurable growth in response to substrate addition and no increase in cell elution during the salicylate pulse. System III exhibited non-ideal behavior for both substrate and cell transport. Of particular interest is the fact that the indigenous communities of the two soils behaved differently. Specifically, for the Hayhook soil, an increased elution response was observed for the heterotrophic population while the salicylate-degrading community was preferentially retained in the column. In contrast for the Vinton soil, the substrate pulse did not elicit an elution response from either the heterotrophic or salicylate-degrading community from the culturable, indigenous Vinton microorganisms. For Systems II and III, the observed variability appears to be associated with the biological component of the system, since sterile controls were reproducible. This type of systematic study is critical for understanding cell and substrate transport behavior in complex, heterogeneous systems, and illustrates the potential uncertainty associated with measurements in such systems.  相似文献

Sulfonylurea herbicides are widely used in crop production on the Canadian prairies and a portion of these herbicides applied to cropland are inevitably lost to surrounding aquatic ecosystems. Little is known regarding the presence of sulfonylurea herbicides in wetlands located amongst cropland. This paper describes a new analytical method for the extraction and the determination of seven sulfonylurea herbicides (thifensulfuron-methyl, tribenuron-methyl, ethametsulfuron-methyl, metsulfuron-methyl, rimsulfuron, nicosulfuron and sulfosulfuron) in wetland sediment. The method provided > 85% analyte recovery from fortified sediment for six of the seven sulfonylurea herbicides with a limit of quantification (LOQ) of 1.0 μ g kg? 1. Tribenuron-methyl had significantly lower recovery compared to the other six sulfonylurea herbicides (LOQ = 2 μ g kg? 1). Mean recovery standard deviations were < 10%. This methodology was used to quantify sulfonylurea herbicide residues in sediment samples collected from prairie wetlands situated within the agricultural landscape of Saskatchewan and Manitoba, Canada. This is the first-known detection of sulfonylurea herbicide residues in prairie wetland sediments. Ethametsulfuron-methyl, sulfosulfuron and metsulfuron-methyl, the three most environmentally persistent of the seven sulfonylurea herbicides monitored in the surveillance component of this study, were most frequently detected in wetland sediment with mean concentrations ranging from 1.2 to 10 μ g kg? 1.  相似文献

Rhamnolipids produced by Pseudomonas aeruginosa have been proposed as soil washing agents for enhanced removal of metal and organic contaminants from soil. A potential limitation for the application of rhamnolipids is sorption by soil matrix components. The objective of this study is to empirically determine the contribution of representative soil constituents (clays, metal oxides, and organic matter) to sorption of the rhamnolipid form most efficient at metal complexation (monorhamnolipid). Sorption studies show that monorhamnolipid (R1) sorption is concentration dependent. At low R1 concentrations that are relevant for enhancing organic contaminant biodegradation, R1 sorption followed the order: hematite (Fe(2)O(3))>kaolinite>MnO(2) approximately illite approximately Ca-montmorillonite>gibbsite (Al(OH)(3))>humic acid-coated silica. At high R1 concentrations, relevant for use in complexation/removal of metals or organics, R1 sorption followed the order: illite>humic acid-coated silica>Ca-montmorillonite>hematite>MnO(2)>gibbsite approximately kaolinite. These results allowed prediction of R1 sorption by a series of six soils. Finally, a comparison of R1 and R2 (dirhamnolipid) shows that the R1 form sorbs more strongly alone than when in a mixture of both the R1 and R2 forms. The information presented can be used to estimate, on an individual soil basis, the extent of rhamnolipid sorption. This is important for determining: (1) whether rhamnolipid addition is a feasible remediation option and (2) the amount of rhamnolipid required to efficiently remove the contaminant.  相似文献

Abandoned mine tailings sites in semiarid regions remain unvegetated for extended periods of time and are subject to eolian dispersion and water erosion. This study examines the potential phytostabilization of a lead-zinc mine tailings site using a native, drought-tolerant halophyte, quailbush [Atriplex lentiformis (Torr.) S. Wats.]. In a greenhouse study germination, growth, and metal uptake was evaluated in two compost-amended mine tailings samples, K4 (pH 3) and K6 (pH 6) at 75, 85, 90, 95, and 100% mine tailings, and two controls, off-site and compost. Microbial community changes were monitored by performing MPN analysis of iron- and sulfur-oxidizing bacteria as well as heterotrophic plate counts. Results demonstrate that germination is not a good indicator for phytostabilization since it was only inhibited in the unamended K4 treatment. Plant growth was significantly reduced in 95 and 100% mine tailings, while growth in 75, 85, and 90% treatments was similar to the off-site control. Quailbush accumulated elevated levels of the nutrient metals Na, K, Mn, and Zn in the shoot tissues; however, metal accumulation was generally below the domestic animal toxicity limit. Initially, autotrophic population estimates were four to six logs higher than heterotrophic counts, indicating extremely stressed conditions. However, post-harvest, heterotrophic bacterial counts increased to normal levels (approximately 10(6) CFU g-1 dry tailings) and dominated the rhizosphere. Therefore, with compost amendment, quailbush has good potential as a native species candidate for phytostabilization of mine tailings in semiarid environments.  相似文献

Few effective strategies exist for remediating and restoring metal-contaminated soils. We have evaluated the potential of two environmentally compatible, nondestructive, biological soil-washing agents for remediating aged, lead-contaminated soils. Two contaminated soils were washed with 10 mM rhamnolipid biosurfactant and 5.3% carboxymethyl-beta-cyclodextrin (CMCD). The metal removal efficiency of these agents was compared with 10 mM diethylenetriamine pentaacetic acid (DTPA) and 10 mM KNO3. Lead removal rates by both soil-washing agents exceeded the removal by KNO3, but were an order of magnitude less than removal by the synthetic chelator, DTPA. Analysis of soil extractions revealed that the Pb in the first soil (3780 mg kg(-1)) was primarily associated with the soluble, exchangeable, oxide, and residual fractions while the Pb in the second soil (23 900 mg kg(-1)) was found in the soluble, exchangeable, carbonate, and residual fractions. After 10 consecutive washes, rhamnolipid had removed 14.2 and 15.3% of the Pb from the first and second soils, respectively, and CMCD had removed 5 and 13.4% from the same two soils. The Pb removal rate by both agents either increased or was consistent throughout the 10 extractions, indicating a potential for continued removal with extended washing. Significant levels of Cu and Zn in both soils did not prevent Pb removal by either agent. Interestingly, the effectiveness of each agent varied as a function of Pb speciation in the soil. Rhamnolipid was more effective than CMCD in removing Pb bound to amorphous iron oxides, while both agents demonstrated similar potential for removing soluble, exchangeable, and carbonate-bound Pb. Neither agent demonstrated potential for the complete remediation of metal-contaminated soils.  相似文献

A novel methodology combining digital imaging, conventional fixed visibility monitors, and solar radiation monitors has been developed to characterize the visual air quality of the El Paso and Ciudad Juarez urban vista. The authors have found that the digital image quality is reproducible and useful for quantitative analysis of visibility conditions. Regions of interest were selected in images along view paths of interest and values for a contrast variable of interest, typically the coefficient of variation or contrast ratio (CR) for discrete targets, were computed. Both of these indices are bounded at 0 and 1 and are scaled to the "clean day" maxima for a given date, time, and selected view paths. This produces a relative visibility index for various view paths. With the siting of a Belfort (6230A) visibility monitor at a central location, it has been possible to initiate contrast analysis of various targets in current and archived camera images obtained near this monitor. For uniformly "clean" days, as indicated by fine particulate matter observations and visual inspection, the authors have been able to use the extinction coefficient (Bext) derived from the 6230A to put the relative visibility index, based on CR, on an absolute basis in terms of an ideal target located at a given distance. This permits the generation of contrast extinction, Bext/C, for each view path that is independent of the actual target intrinsic contrast (within limits) and allows the comparison of Bext/C along different view paths with other air quality indices. Multiple linear regression was used to derive a relationship between the CR-based Bext/C value and air quality parameters. Visibility attenuation because of sulfate particles was found to have the highest correlation with Bext/C. In addition, solar radiation was observed to be a significant predictor of visibility in the urban region.  相似文献

The role of science in environmental foreign policymaking has received little attention in the international environmental politics literature, and systematic, in-depth case studies of the influence of science on environmental foreign policy with respect to persistent organic pollutants (POPs) are entirely missing. We present a case study of the influence of science on Canada's foreign policy on POPs from the discovery of the transboundary nature of the POPs problem around 1985 to the signing of the United Nations Stockholm Convention on Persistent Organic Pollutants in 2001. Influence was analyzed in terms of a knowledge–action methodology that focused on the types of scientific knowledge and the actions taken by scientists that were influential. Data were collected through interviews and extensive document analysis. We conclude that, while individual knowledge types and actions were influential during various periods (single-factor explanations), it was an increasingly layered and integrated “package” of knowledge types and actions, with human health impacts as its touchstone and partnering between scientists and non-scientists as its watchword, that propelled and sustained Canada's foreign policy on the POPs issue.  相似文献

Glyphosate [N-(phosphonomethyl)glycine] is the active ingredient of several herbicide products first registered for use in 1974 under the tradename Roundup. The use of glyphosate-based herbicides has increased dramatically over the last two decades particularly in association with the adoption of glyphosate-tolerant crops. Glyphosate has been detected in a range of surface waters but this is the first study to monitor its fate in prairie wetlands situated in agricultural fields. An ephemeral wetland (E) and a semi-permanent wetland (SP) were each divided into halves using a polyvinyl curtain. One half of each wetland was fortified with glyphosate with the added mass simulating an accidental direct overspray. Glyphosate dissipated rapidly in the water column of the two prairie wetlands studied (DT(50) values of 1.3 and 4.8 d) which may effectively reduce the impact of exposure of aquatic biota to the herbicide. Degradation of glyphosate to its major metabolite aminomethylphosphonic acid (AMPA) and sorption of the herbicide to bottom sediment were more important pathways for the dissipation of glyphosate from the water column than movement of the herbicide with infiltrating water. Presently, we are not aware of any Canadian guidelines for glyphosate residues in sediment of aquatic ecosystems. Since a substantial portion of glyphosate entering prairie wetlands will become associated with bottom sediments, particularly in ephemeral wetlands, guidelines would need to be developed to assess the protection of organisms that spend all or part of their lifecycle in sediment.  相似文献

Two approaches were used to investigate the influence of dissolved oxygen (DO) and substrate availability on the formation and dynamics of "bioactive zones" in a water-saturated porous medium. A bioactive zone is defined as a region where a microbial community is sufficiently active to metabolize bioavailable substrates. In the first approach, microbial activity was characterized by monitoring the spatial and temporal variability of DO and aqueous substrate (salicylate and naphthalene) concentrations during miscible-displacement experiments. In the second approach, microbial activity was monitored using multiple fiber optics emplaced in the porous medium to detect luminescence produced by Pseudomonas putida RB1353, a bioluminescent reporter organism that produces light when salicylate (an intermediate of naphthalene degradation) is present. The results of both approaches show that the location and size of the bioactive zones were influenced by in situ DO and substrate availability. When DO was not a limiting factor (i.e., lower substrate input concentrations), the bioactive zone encompassed the entire column, with the majority of the microbial activity occurring between the inlet and midpoint. However, as the availability of DO became limiting for the higher substrate input experiments, the size of the bioactive zone shrank and was ultimately limited to the proximity of the column inlet.  相似文献

The growing industrial demand for sustainable materials has led to a paradigm shift in the focus from synthetic polymers towards natural fibres. This paper deals with the challenges and opportunities associated with the use of natural fibre-reinforced polymer composites in various industrial applications. Natural fibres being biodegradable, light in weight, cost-effective and environment friendly are good candidate materials for modern industrial applications. Use of natural fibres in various industries with a focus on automotive and furniture industry has been discussed. The commonly used natural fibres in polymer composites including jute, hemp, sisal, kenaf, bamboo, cotton, flax, abaca, coir etc. have been dealt with in this paper. The literature revealed that tensile strength and other mechanical properties of these fibres are comparable to synthetic fibres like glass or carbon fibres. However, the temperature stability of polymers limits their extensive use and remains an issue to be addressed.  相似文献
