The information on bacterial community composition (BCC) in Portuguese water bodies is very scarce. Cértima River (central western Portugal) is known to have high levels of pollution, namely organic. In the present work, the BCC from a set of 16 water samples collected from Cértima River Basin and its main tributaries was characterized using 16S rDNA–denaturing gradient gel electrophoresis, a culture-independent molecular approach. Molecular data were related to environmental parameters through multivariate analysis to investigate potential impact of water pollution along the river. Principal component analysis using environmental data showed a water quality gradient from more pristine waters (at the mountain tributaries) to waters with increasingly eutrophic potential (such as Fermentelos Lake). This gradient was mainly defined by factors such as organic and inorganic nutrient sources, electrical conductivity, hydrogen carbonate concentration, and pH. Molecular results showed variations in BCC along Cértima River Basin but in the main river section, a Bacteroidetes phylotype (Flavobacterium sp.) proved to be dominant throughout the river course. Multivariate analysis suggests that spatial variation of BCC along the Cértima River Basin depended mainly on parameters such as Chl a, total suspended solid (TSS), total organic carbon, electrical conductivity, and HCO _boxclose^-_{3}^{-} levels. Bacteroidetes phylotypes were all related to higher electrical conductivity and HCO3-_{3}^{-} levels although some of these were also correlated with high SO42-_{4}^{2-} and others with high soluble reactive phosphorus, nitrate, TN, and Kjeld-N levels. The Gammaproteobacteria occurrence was correlated with high SO42-_{4}^{2-} levels. One of the Betaproteobacteria phylotypes showed to correlate with low redox potential (Eh) and high temperature, pH, TSS, and Chl a levels while another one showed a negative correlation with Chl a values.  相似文献

Cyanobacteria dominance and cyanotoxin production can become major threats to humans and aquatic life, especially in warm shallow lakes, which are often dominated by cyanobacteria. This study investigates the occurrence and distribution of microcystins (MCYST) in water, cell-bound and in the tissues of the commercial mugilid Liza sp. in the largest, coastal, Spanish Mediterranean lake (Albufera of Valencia). This is the first report concerning microcystin accumulation in tissues of mugilid fish species. Considerable amounts of microcystins were found in the water and seston, which correlated with development of Microcystis aeruginosa populations in the lake. The MCYST concentrations found in Lake Albufera (mean 1.7 and 17 μg/L and maximum 16 and 120 μg/L in water and seston, respectively) exceeded by one to two orders of magnitude the guideline levels proposed by the World Health Organization and were higher than that reported in other lakes of the Mediterranean zone. The presence of MCYST was found in all the fishes studied and accumulated differently among tissues of the commercial species Liza sp. Toxin accumulation in fish tissues showed that although the target organ for MCYST was the liver, high concentrations of microcystins were also found in other analysed tissues (liver>intestine>gills>muscle). Human tolerable daily intake for microcystins is assessed relative to the WHO guidelines, and potential toxicological risks for humans, wildlife and related ecosystems of the lake are discussed.  相似文献

According to Directive 2008/50/EC, all equipment used for NO(x) fixed measurements must comply with the reference method or equivalent by 11 June 2013. Up to that date, non-type-approved equipment can continue to be used in air monitoring networks and will probably also be used in air pollution studies even after that date. In this context, it is advisable to know how these "old" measuring instruments respond to the requirements of European Standard EN 14211: 2005: Ambient Air Quality--Standard method for the measurement of the concentration of nitrogen dioxide and nitrogen monoxide by chemiluminescence. In this work, we carry out a thorough performance evaluation of two commercial NO(x) analysers, both purchased before 2010, according to the mentioned Standard. Results show that water vapour interference is one of the most important disconformities with this Standard. The expanded uncertainty estimation of both analysers was below the quality objective of the above-mentioned Directive (15%); however, this calculation was performed by applying some modifications to the proposed model of Standard EN 14211. We study opportunities for CO2 sequestration in geological formations of the state North Rhine Westphalia in Germany. Simulations are performed for evaluating a potential site within the Bunter sandstone formation near the town of Minden in a depth?of around 3,000 m using the numerical simulator TOUGHREACT. Our focus is on three CO2 storage mechanisms: (1) hydrodynamic trapping, (2) dissolution trapping, and (3) mineral trapping. The results show that due to buoyancy the injected CO2 phase initially migrates towards the top of the reservoir and is hydrodynamically trapped beneath the confining layer of the cap rock. Then, the CO2 spreads laterally and dissolves partially in the formation water. The dissolution of CO2 results in an increase of the density of the brine causing a downward migration until it settles after 10,000 years at the bottom of the reservoir. The simulations indicate that after 10,000 years, 15% (17 Mt) from a total of 114 Mt injected CO2 are trapped hydrodynamically, 20% (23 Mt) are trapped by dissolution, and 65% (74 Mt) are fixed in newly formed carbonates such as dawsonite, ankerite, and siderite. Within our study pressure increases near the injection well by a factor of 1.1 which is lower than the upper limit usually accepted in gas storage operations. The mineral reactions cause a net decrease of porosity and in turn a decrease of permeability down to 9% of the initial value in parts of the reservoir.  相似文献

Vegetation in the upper catchment of Yellow River is critical for the ecological stability of the whole watershed. The dominant vegetation cover types in this region are grassland and forest, which can strongly influence the eco-environmental status of the whole watershed. The normalized difference vegetation index (NDVI) for grassland and forest has been calculated and its daily correlation models were deduced by Moderate Resolution Imaging Spectroradiometer products on 12 dates in 2000, 2003, and 2006. The responses of the NDVI values with the inter-annual grassland and forest to three climatic indices (i.e., yearly precipitation and highest and lowest temperature) were analyzed showing that, except for the lowest temperature, the yearly precipitation and highest temperature had close correlations with the NDVI values of the two vegetation communities. The value of correlation coefficients ranged from 0.815 to 0.951 (p?<?0.01). Furthermore, the interactions of NDVI values of vegetation with the climatic indicators at monthly interval were analyzed. The NDVI of vegetation and three climatic indices had strong positive correlations (larger than 0.733, p?<?0.01). The monthly correlations also provided the threshold values for the three climatic indictors, to be used for simulating vegetation growth grassland under different climate features, which is essential for the assessment of the vegetation growth and for regional environmental management.  相似文献

Paraíba do Sul River is located at a very densely inhabited region of Brazil crossing the three most industrialized states of the country (S?o Paulo, Minas Gerais and Rio de Janeiro states). As a result, industrial and farming residues as well as urban sewage are frequently disposed without appropriate treatment. The current study aimed at investigating the water quality in three reservoirs along the Paraíba do Sul River (Ilha dos Pombos, Santa Cecília and Santa Branca), through physiological, morphological, biochemical, and genetic biomarkers. The bioindicator chosen was the catfish Pimelodus maculatus, sampled during the dry (June 2008) and rainy (February 2009) seasons. Also, some water physicochemical parameters were analyzed from the sampling sites, but displayed no alterations according to the Brazilian Agency for Water Quality Legislation. Branchial carbonic anhydrase activity was inhibited in the dry season, while renal carbonic anhydrase activity was inhibited in the rainy season in the Santa Branca reservoir, indicating disturbance of osmoregulatory and acid-base regulation processes. Histopathological alterations were observed in the gills (neoplasic and tissue hyperplasia processes) and liver (necrosis), indicating serious damage to the functional integrity of these organs. A high incidence of melanomacrophage centers was observed in the liver, suggesting an intense immune response in all reservoirs. Acetylcholinesterase and catalase activity showed also differences corroborating some morphological results. Likewise, the induction of the micronucleus and DNA damage indicate genotoxicity, but mainly in the Santa Branca reservoir. Thus, the health status of P. maculatus warrants caution in the use of the water from the 3 reservoirs for direct human consumption, particularly after the accidental spill of endosulfan in November 2008, three months before the rainy season sampling.  相似文献

Insufficient wastewater remediation in small communities and nonpoint source pollution are the key factors in determining the water quality of small streams in an agricultural landscape. Despite the current extensive construction of municipal wastewater treatment facilities in small communities, the level of organic substances and nutrients in the recipient catchments has not decreased in many areas. This paper analyzes the changes in the water quality of the small streams after the deployment of wastewater treatment plants that were designed to address sources of pollution from small municipalities. The analysis is based on the results from a water quality monitoring network in the small watersheds in the Czech Republic. Five rural catchments with one dominant municipal pollution source, where a wastewater treatment plant was deployed during the monitoring period, were selected according to a predefined set of criteria, from a series of 317 profiles. Basic water quality indicators were selected for the assessment: O2, BOD-5, COD, TOC, conductivity, NH4-N, NO2-N, NO3-N, PT, and PO4-P. Results of the analysis showed that the simple deployment of the water treatment facilities at these streams often did not lead to a reduction of contamination in the streams. The expected post-deployment changes, namely, a significant and permanent reduction of stream contamination, occurred only in one catchment, whereas in the remainder of the catchments, only marginal changes or even increased concentrations of the contaminants were detected. As the critical factors that determined the efficiency of wastewater treatment were studied, the need for the consideration of the local conditions during the design of the facility, particularly regarding the size of the catchments, initial level of contamination, proper system of operation, and process optimization of the treatment facility, emerged as the important factor.  相似文献

Physicochemical characteristics of sediment and benthic communities were studied in the proximity of seven sewage outfalls with differences in flow and wastewater treatment in the Western Mediterranean Sea. Redox potential was the only abiotic parameter which showed a pattern related with distance to outfalls, whereas granulometry, percentage of organic matter, metal concentrations or pH did not show changes related with outfall presence. Benthic community analysis proved to be the most suitable monitoring tool. The results showed that the highest impacted stations corresponded with those closest to outfall with the highest flow and only pre-treatment, whilst a decrease of this tendency was detected in the locations where secondary treatment takes place. Meta-analysis showed a decrease of amphipods and tanaids abundance as well as redox potential, as the indicators with the clearest response to sewage presence.  相似文献

The effect of two fly ashes as soil amendment on the adsorption–desorption of metolachlor [2-chloro-N-(2-ethyl-6-methylphenyl)-N-(2-methoxy-1-methylphenyl)] and atrazine (2-chloro-4-ethylamino-6-isopropylamino-1,3,5-triazine) was studied in alluvial and laterite soils. The adsorption data for both the herbicides fitted well the Freundlich equation, and Freundlich adsorption coefficient (K f) increased with an increase of fly ash amount. Both the fly ashes differed in their extent to increase herbicide sorption, and the effect was different in different soils. Atrazine was sorbed more in the soils/soils?+?fly ash mixtures than the metolachlor. The K f values showed significant correlation with the amount of fly ash amendment (correlation coefficient, R?>?0.982). The desorption isotherms also fitted the Freundlich equation, and desorption showed hysteresis which increased with an increase in the content of fly ash amendment. The free energy change (ΔG) indicated that the sorption process is exothermic, spontaneous, and physical in nature. The study has shown that fly ash as soil amendment significantly increased the sorption of metolachlor and atrazine, but the effect is soil- and fly ash-specific.  相似文献

Enteric viruses monitoring in surface waters requires the concentration of viruses before detection assays. The aim of this study was to evaluate different methods in terms of recovery efficiencies of bacteriophage PP7 of Pseudomonas aeruginosa, measured by real-time PCR, using it as a viral control process in water analysis. Different nucleic acid extraction methods (silica–guanidinium thiocyanate, a commercial kit (Qiagen Viral RNA Kit) and phenol–chloroform with alcohol precipitation) exhibited very low recovery efficiencies (0.08–4.18 %), being the most efficient the commercial kit used for subsequent experiments. To evaluate the efficiency of three concentration methods, PBS (as model for clean water) and water samples from rivers were seeded to reach high (HC, 106 pfu ml?1) and low concentrations (LC, 104 pfu ml?1) of PP7. Tangential ultrafiltration proved to be more efficient (50.36?±?12.91, 17.21?±?9.22 and 12.58?±?2.35 % for HC in PBS and two river samples, respectively) than adsorption–elution with negatively charged membranes (1.00?±?1.34, 2.79?±?2.62 and 0.05?±?0.08 % for HC in PBS and two river samples, respectively) and polyethylene glycol precipitation (15.95?±?7.43, 4.01?±?1.12 and 3.91?±?0.54 %, for HC in PBS and two river samples, respectively), being 3.2–50.4 times more efficient than the others for PBS and 2.7–252 times for river samples. Efficiencies also depended on the initial virus concentration and aqueous matrixes composition. In consequence, the incorporation of an internal standard like PP7 along the process is useful as a control of the water concentration procedure, the nucleic acid extraction, the presence of inhibitors and the variability of the recovery among replicas, and for the calculation of the sample limit of detection. Thus, the use of a process control, as presented here, is crucial for the accurate quantification of viral contamination.  相似文献
