Immunoassay methods are generally used for measuring of allergenic substances. However, they need special facilities, skilled handling, and time-consuming procedure. In this work, a fiber-optic immunoassay system which could measure allergen by fluorescent intensities of immune complexes formed by allergens and fluorescently labeled antibodies was established. Immune complexes absorbed on the optical fiber probe surface, and excitation light was injected into the probe, then evanescent field is created in the proximity of the probe. The fluorophores were excited by the evanescent light, and fluorescence was detected by a photo diode. The target allergen detected by our system was Der f1 derived from Dermatophagoides farinae that is one of the house dust mite and major source of inhaled allergens. The fluorophore used labeling on detecting antibody was cyanine 5. The system enabled to detect and quantitatively determine of Der f1. The measurement range was from 0.24 to 250?ng/ml, and the result competes with ELISA. The measurement time was 16?min/sample. The immunoassay system was applied to measurement of Der f1 from actual dust samples. Calculated values of Der f1 showed good correlations between the fiber-optic fluoroimmunoassay and ELISA. 相似文献

This paper examines effectiveness of a regulatory enforcement organization (Major industrial Accident Prevention Center, MAPC), and a grading system for implementation of the Process Safety Management (PSM) regulation in Korea. A lot of chemical installations have been built in Korea since the 1960s. The frequent occurrence of major industrial accidents had made people's concerns grow. The Korean government enacted PSM regulations in 1996 in order to curb these accidents.However, a key question is how to make sure companies comply with the PSM regulations. In order to improve company’ compliance with PSM regulations the Ministry of Employment and Labor (MOEL) responsible for the regulation introduced a grading management system in 2001 and then established special supervisory centers for enforcement of PSM regulations in 2005. This paper reviews the role and effectiveness of the system in term of PSM enforcement. The author found that the grade-based approach has encouraged employers to implement the requirements of the PSM regulations. MAPCs play an effective role in enhancing enforcement performance. Although the more chemical plants have been established in Korea, the fewer major industrial accidents have occurred since the introduction of the system. Evolution of caste is a central issue in the biology of social insects. Comparative studies on their morphology so far suggest
the following three patterns: (1) a positive correlation between queen–worker size dimorphism and the divergence in reproductive
ability between castes, (2) a negative correlation among workers between morphological diversity and reproductive ability,
and (3) a positive correlation between queen–worker body shape difference and the diversity in worker morphology. We conducted
morphological comparisons between castes in Pachycondyla luteipes, workers of which are monomorphic and lack their reproductive ability. Although the size distribution broadly overlapped,
mean head width, head length, and scape length were significantly different between queens and workers. Conversely, in eye
length, petiole width, and Weber’s length, the size differences were reversed. The allometries (head length/head width, scape
length/head width, and Weber’s length/head width) were also significantly different between queens and workers. Morphological
examinations showed that the body shape was different between queens and workers, and the head part of workers was disproportionately
larger than that of queens. This pattern of queen–worker dimorphism is novel in ants with monomorphic workers and a clear
exception to the last pattern. This study suggests that it is possible that the loss of individual-level selection, the lack

To examine the levels of PCDD/DFs pollution in environmental samples in the vicinity of various incinerators, the levels of PCDD/DFs in air and soil samples collected near 17 incinerators and stack emission gases were investigated between 2003 and 2006. A total of 434 soil, 28 stack emission gas, and 38 air samples were analyzed for their PCDD/DFs concentrations. The PCDD/DFs concentrations in the flue gas samples ranged from 0.02 to 16.41 ng I-TEQ/Sm(3), with an arithmetical mean value of 3.13 ng I-TEQ/Sm(3). The PCDD/DFs concentrations in the air samples ranged from 0.032 to 0.965 pg I-TEQ/Sm(3). The soil samples contained between N.D. and 153.23 pg I-TEQ/g-dry, with an average of 7.36 pg I-TEQ/g-dry. These levels were generally consistent with or lower than many previous studies. The average PCDD/DFs levels in the soil samples decrease with increasing distance from the incinerator. From the PCDD/DFs level gradient from each plant, a distance of 500 m is suggested as being under the influence of an incinerator. 相似文献

The behavior and mechanism of Li leaching from lithium aluminum silicate glass-ceramics which can be used as a secondary source of Li using aqueous NaOH solution was investigated. The Li leaching efficiency is increased with increasing concentration of NaOH, specific surface area, and reaction temperature. When leached under optimum conditions, 2 mol/L NaOH, 53 μm particle undersize, 1:10 solid/liquid ratio, 250 r/min stirring speed, 100°C reaction temperature, 12 hr, the Li leaching efficiency was approximately 70%. However, when the leaching experiment was performed for 48 hr, the concentration of Li+ ions contained in the leach liquor decreased from 1160 to 236 mg/L. To investigate the origin of this phenomenon, the obtained leach residue was analyzed by X-ray diffraction, scanning electron microscopy, and energy-dispersive X-ray spectroscopy. These analyses show that zeolite was formed around the lithium aluminum silicate glass-ceramics, which affected the leaching of by adsorbing Li+ ions. In addition, using the shrinking-core model and the Arrhenius equation, the leaching reaction with NaOH was found to depends on the chemical reaction of the two reactants, with a higher than 41.84 kJ/mol of the activation energy. 相似文献

An evaluation of a river health using the index of biological integrity along with relations to chemical and habitat conditions 总被引:13,自引:0,他引:13
We evaluated the health condition of a temperate river during June-November 1999 through applications of the index of biological integrity (IBI) using fish assemblages and qualitative habitat evaluation index (QHEI) as well as chemical analyses. Overall IBI values ranged from 13 to 37 and averaged 23 (n = 25, standard error = 1.16), indicating a "poor" or "very poor" condition according to the criteria of modified Karr [Fisheries 6 (1981) 21]. The values of mean IBI declined at a rate of 0.22 km(-1) (R2 = 0.91, p < 0.05) along the longitudinal distance from the headwaters to the downstream sites. Reduced IBI values at downstream sites reflected low forest cover, high population density and high nutrient enrichments. Ecotoxicity tests using the river water also showed that toxic impacts were evident in the downriver sites. These factors resulted in decreases of riffle benthic species and insectivores and increases of tolerant species, anormalies and exotic species in the river. Spatial pattern in IBI agreed with QHEI values, which showed a linear relation (R2 = 0.998, p < 0.001) with species richness. Field measurements of conductivity and pH, an indicators for variation of conservative ions, showed that the river water was diluted by 40% fold by summer monsoon rain and surface run-off from the watershed, resulting in a physical and chemical instability during the monsoon. For these reasons, average IBI values during the monsoon and postmonsoon decreased >20% compared to the premonsoon, indicating that IBI values were also affected by flow regime. Based on the overall physico-chemical data and IBI values, the river health is rapidly degrading due to the combined effect of chemical contaminations and habitat modifications. 相似文献
