This study examined the effects of brewers yeast, fructooligosaccharide (FOS), and GroBiotic-A, a mixture of partially autolyzed brewers yeast, dairy components and dried fermentation products, on the intestinal microbial community of red drum, Sciaenops ocellatus. Gastrointestinal (GI) tracts were aseptically removed from three sub-adult red drum previously maintained on a commercial diet and placed in an anaerobic chamber. Intestinal contents were removed, diluted and incubated in vitro in one of four liquid media: normal diet alone, diet + 2% (w/w) GroBiotic-A, diet + 2% brewers yeast, and diet + 2% FOS. After 24 and 48 h of incubation at 25 degrees C, supernatants were removed for volatile fatty acid (VFA) analysis and DNA was extracted for denaturing gradient gel electrophoresis (DGGE) analysis. Polymerase chain reaction (PCR) was performed on a highly conserved region of M 16S rDNA and the amplicons were subjected to DGGE. The microbial community (MC) fingerprint was used to distinguish microbial populations. The intestinal contents incubated with GroBiotic-A had significantly (P<0.05) higher acetate and total VFA concentrations at 48 h compared to the other treatments. We investigated the distribution and transport of coal tar-derived polycyclic aromatic hydrocarbons (PAHs) in fine-grained residuum and alluvial floodplain deposits that underlie a former manufactured gas plant. All 16 USEPA priority pollutant PAHs are present at this site and have penetrated the entire 4-5m thickness of clayey sediments, which unconformably overly limestone bedrock. Concentrations of less hydrophobic PAHs (e.g., naphthalene, 0.011-384mg kg(-1)) were about 10 times higher than those of highly hydrophobic PAHs (e.g., benzo[g,h,i]perylene -0.002 to 56.03mgkg(-1)). Microscopic examination of thin-sections of the clay-rich sediments showed that fractures and rootholes, which can act as pathways for flow, occur throughout the profiles. Tarry residue was found coating some fractures and rootholes, indicating that coal tar was, in some cases, able to penetrate as an immiscible phase. However, in the vast majority of samples in which PAHs were detected, there was no detectable tar residue, suggesting that much of the transport occurred in the dissolved phase. Examination of thin-sections with an epifluorescent microscope indicated that PAHs, which fluoresce brightly when exposed to UV light, are distributed throughout the soil matrix, rather than being confined to fractures and rootholes. The widespread distribution of PAHs is most likely due to diffusion-controlled exchange between the fast-flow pathways in the fractures and rootholes and the relatively immobile water in the fine-grained matrix. This implies that fractures and rootholes can play a major role in controlling transport of highly hydrophobic compounds in fine-grained sediments, which would otherwise act as barriers to contaminant migration. 相似文献
Alkylphenols (APs), alkylphenol ethoxylates (APEOs), ethoxycarboxylate metabolites (APECs) and bisphenol A were determined in surface water using solid-phase extraction (SPE) followed by triple-quadrupole LC-MS-MS. APs were separated by LC from APECs using an acetonitrile-water-gradient without the addition of any buffer. Nonylphenol ethoxycarboxylates (NPECs) interfere in the detection of nonylphenols (NPs) when using an acidic mobile phase, because they produce the same MS-MS fragment ions (219>133 and 147). 4n-NP shows the characteristic transition 219>106; it is well suited as internal standard. Nonylphenol ethoxylates NPE(n)Os (n=1-17) were analysed separately in a second run by positive ionization using an ammonium acetate mobile phase. Textile industry discharges, the corresponding wastewater treatment plant (WWTP) effluents and the receiving rivers in Belgium and Italy were analysed. Among the substances investigated, NPE1C and NPE2O exhibited the highest concentrations in the water samples, up to 4.5 microg l(-1) NPE1C in a WWTP effluent and 3.6 microg l(-1) NPE2O in a river. The highest NP levels were found in the receiving rivers (max. 2.5 microg l(-1)). The predicted no-effect concentration (PNEC) for NP of 0.33 microg l(-1) for water species was frequently exceeded in the surface waters investigated, suggesting potential adverse effects to the aquatic environment. 相似文献
Iwao's quadratic regression or Taylor's Power Law (TPL) are commonly used to model the variance as a function of the mean for sample counts of insect populations which exhibit spatial aggregation. The modeled variance and distribution of the mean are typically used in pest management programs to decide if the population is above the action threshold in any management unit (MU) (e.g., orchard, forest compartment). For nested or multi-level sampling the usual two-stage modeling procedure first obtains the sample variance for each MU and sampling level using ANOVA and then fits a regression of variance on the mean for each level using either Iwao or TPL variance models. Here this approach is compared to the single-stage procedure of fitting a generalized linear mixed model (GLMM) directly to the count data with both approaches demonstrated using 2-level sampling. GLMMs and additive GLMMs (AGLMMs) with conditional Poisson variance function as well as the extension to the negative binomial are described. Generalization to more than two sampling levels is outlined. Formulae for calculating optimal relative sample sizes (ORSS) and the operating characteristic curve for the control decision are given for each model. The ORSS are independent of the mean in the case of the AGLMMs. The application described is estimation of the variance of the mean number of leaves per shoot occupied by immature stages of a defoliator of eucalypts, the Tasmanian Eucalyptus leaf beetle, based on a sample of trees within plots from each forest compartment. Chemical treatments are widely employed to improve the fiber-matrix adhesion in composites based on eco-friendly fibers such as flax. To better understand the influence of these treatments on processing behavior, this study characterized the surface chemistry and morphology of woven flax fabrics treated by acetone, alkaline, silane and diluted epoxy. Flax/epoxy composites were then manufactured by resin infusion and the flow front and preform thickness evolution was monitored. The alkaline treatment was shown to result in a 50 % increase in equivalent permeability due to an increase in porosity which led to a decrease in flexural properties. The processing results were found to be in good agreement with predictions of a 1-dimensional model. This study suggests that infusion times are not considerably affected by the observed changes in surface energy. However, other implications of the treatments such as an increase in fibrillation can alter the infusion times significantly.
The fermentation process is an important component in the biodegradation of organic compounds in natural and contaminated systems. Comparing with terminal electron-accepting processes (TEAPs), however, research on fermentation processes has to some extent been ignored in the past decades, particularly on the persistence of fermentation process in the presence of toxic organic pollutants. Both field and laboratory studies, presented here, showed that microbial processes in a groundwater-based system exhibited a differential inhibitory response to toxicity of phenolic compounds from coal tar distillation, thus resulting in the accumulation of volatile fatty acids (VFAs) and hydrogen. This indicated that fermentation processes could be more resistant to phenol toxicity than the subsequent TEAPs such as methanogenesis and sulfate reduction, thus providing us with more options for enhancing bioremediation processes. 相似文献
Field trials were established to compare alum-treated poultry litter (ATPL), normal poultry litter (NPL), and triple superphosphate (TSP) as fertilizer sources for corn (Zea mays L.) when applied at rates based on current litter management strategies in Virginia. Trials were established in the Costal Plain and Piedmont physiographic regions near Painter and Orange, VA, respectively. Nitrogen-based applications of ATPL or NPL applied at rates estimated to supply 173 kg of plant-available nitrogen (PAN) ha(-1) resulted in significantly lower grain yields than treatments receiving commercial fertilizer at the same rate in 2000 and 2001 at Painter. These decreases in grain yield at the N-based application rates were attributed to inadequate N availability, resulting from overestimates of PAN as demonstrated by tissue N concentrations. However, at Orange no treatment effects on grain yield were observed. Applications of ATPL did not affect Al concentrations in corn ear-leaves at either location. Exchangeable soil Al concentrations were most elevated in treatments receiving only NH4NO3 as an N source. At N-based application rates, the ATPL resulted in lower Mehlich 1-extractable P (M1-P) and water-extractable soil phosphorus (H2O-P) concentrations compared to the application of NPL. A portion of this reduction could be attributed to lower rates of P applied in the N-based ATPL treatments. Runoff collected from treatments which received ATPL 2 d before conducting rainfall simulations contained 61 to 71% less dissolved reactive phosphorus (DRP) than treatments receiving NPL. These results show that ATPL may be used as a nutrient source for corn production without significant management alterations. Alum-treated poultry litter can also reduce the environmental impact of litter applications, primarily through minimizing the P status of soils receiving long-term applications of litter and reductions in runoff DRP losses shortly after application. 相似文献
Onsite wastewater treatment systems (OWTS) are becoming increasingly important for the treatment and dispersal of effluent
in new urbanised developments that are not serviced by centralised wastewater collection and treatment systems. However, the
current standards and guidelines adopted by many local authorities for assessing suitable site and soil conditions for OWTS
are increasingly coming under scrutiny due to the public health and environmental impacts caused by poorly performing systems,
in particular septic tank-soil adsorption systems. In order to achieve sustainable onsite wastewater treatment with minimal
impacts on the environment and public health, more appropriate means of assessment are required. This paper highlights an
integrated risk based approach for assessing the inherent hazards associated with OWTS in order to manage and mitigate the
environmental and public health risks inherent with onsite wastewater treatment. In developing a sound and cohesive integrated
risk framework for OWTS, several key issues must be recognised. These include the inclusion of relevant stakeholders throughout
framework development, the integration of scientific knowledge, data and analysis with risk assessment and management ideals,
and identification of the appropriate performance goals for successful management and mitigation of associated risks. These
issues were addressed in the development of the risk framework to provide a generic approach to assessing risk from OWTS.
The utilisation of the developed risk framework for achieving more appropriate assessment and management techniques for OWTS
is presented in a case study for the Gold Coast region, Queensland State, Australia. 相似文献
The regional-scale importance of an aquatic stressor depends both on its regional extent (i.e., how widespread it is) and
on the severity of its effects in ecosystems where it is found. Sample surveys, such as those developed by the U.S. Environmental
Protection Agency’s Environmental Monitoring and Assessment Program (EMAP), are designed to estimate and compare the extents,
throughout a large region, of elevated conditions for various aquatic stressors. In this article, we propose relative risk
as a complementary measure of the severity of each stressor’s effect on a response variable that characterizes aquatic ecological
condition. Specifically, relative risk measures the strength of association between stressor and response variables that can
be classified as either “good” (i.e., reference) or “poor” (i.e., different from reference). We present formulae for estimating
relative risk and its confidence interval, adapted for the unequal sample inclusion probabilities employed in EMAP surveys.
For a recent EMAP survey of streams in five Mid-Atlantic states, we estimated the relative extents of eight stressors as well
as their relative risks to aquatic macroinvertebrate assemblages, with assemblage condition measured by an index of biotic
integrity (IBI). For example, a measure of excess sedimentation had a relative risk of 1.60 for macroinvertebrate IBI, with
the meaning that poor IBI conditions were 1.6 times more likely to be found in streams having poor conditions of sedimentation
than in streams having good sedimentation conditions. We show how stressor extent and relative risk estimates, viewed together,
offer a compact and comprehensive assessment of the relative importances of multiple stressors. 相似文献
