This study was conducted to develop methods for the application of an immobilized bioluminescence strain(KG1206),preserved by deep-freezing(DF),for the monitoring of contaminated environments.The immobilized cells,preserved by DF,required approximately 2 hr for reconstitution of their activity.A large reduction in bioluminescence was observed due to the DF process;0.07-0.58 times that of the non deep-frozen(NDF) immobilized strain.The decreased bioluminescence activity induced by the DF process was enhanced by the stimulants,sodium lactate(SL) and KNO 3.However,regardless of the inducer chemical tested,the immobilized strain modified with KNO 3 consistently produced greater bioluminescence than that treated with SL,in the range of 3.0-10.7(avg.6.7 ± 3.69) and 1.2-4.2(avg.2.4 ± 1.47) times that of control,respectively.All KNO 3 treatments of contaminated groundwater samples also resulted in an increase in bioluminescence activity,but the rate of stimulation varied for each sample.Also,no strong linear correlation was observed between the bioluminescence and the total concentration of an inducer,which may related to the complex characteristics of the environmental samples.Overall,the results demonstrated the ability of immobilized genetically engineered bacteria,preserved by DF,to measure a specific group of environmental contaminants using a stimulating agent(KNO 3),suggesting the potential for its preliminary application in a field-ready bioassay.  相似文献

Alkylphenol polyethoxylates (APEO), alkylphenols combined with ethylene oxide, are a class of nonionic surfactants. APEO have been widely used for industrial, agricultural and household applications, and are biodegraded to more persistent and estrogen-active products, namely, nonylphenol (NP), octylphenol (OP), butylphenol (BP), nonylphenol monoethoxylate (NP(1)EO) and nonylphenol diethoxylate (NP(2)EO). This study determined NP levels in commonly consumed foodstuffs to assess daily intake of NP in a Taiwanese population. This study analyzes 318 of samples from 25 types of commonly consumed foodstuffs in northern, central, southern and eastern regions of Taiwan and estimates daily intake of NP in 466 subjects. Moreover, daily NP intake for 3915 additional subjects was estimated by analyzing data from the Nutrition and health survey in Taiwan (NAHSIT). The foodstuff samples were analyzed for five alkylphenol compounds simultaneously by HPLC with fluorescence detection. Additionally, the average compositions of typical foods consumed in Taiwan were investigated. In combination with alkylphenol levels in these foodstuffs, daily intake of NP in Taiwanese was calculated. The average daily intake of NP for the 466 subjects was 28.04+/-25.32 microg/day. Estimated daily intake of NP, based on NP levels in this study as well as the NAHSIT data, was 31.40 microg/day. Rice was the most commonly consumed source of NP, the proportion was 21.46% among daily intake of NP and the following were aquatic products and livestock, which percentage were 17.97% and 17.38%, respectively. Additionally, oysters had the highest NP levels (235.8+/-90.7 ng/g) in four regions of Taiwan, followed by salmon (123.8+/-116.2 ng/g). This study suggested that the average daily NP intake in Taiwan is 4-fold and 8.5-fold higher than daily intake in Germany and New Zealand, respectively and rice was the major source of NP intake.  相似文献

This study proposes a robust prioritization framework for climate change adaptation strategies under uncertain climate change scenarios, using the VIseKriterijumska Optimizacija I Kompromisno Resenje (VIKOR) method, a multi‐criteria decision‐making approach, together with the Shannon entropy‐based weights. The VIKOR method allows us to find a compromise solution between two decision strategies of maximizing group utility and minimizing individual regret, and the Shannon entropy is used to determine objective weights among multiple climate change scenarios. The proposed methodology was applied to the problem of selecting locations of subwatersheds for reusing treated wastewater (TWW) in a Korean urban watershed. Selected based on the sustainability concept, hydro‐environmental and socioeconomic indicators were used to evaluate the sustainability of TWW reuse under multiple climate change scenarios, using the hydrologic simulation model results and statistical data. Finally, sustainability scores under multiple scenarios were aggregated using the VIKOR together with the Shannon entropy‐based weights for the robust prioritization of adaptation strategies. According to the different levels of regret aversion or affinity, our results for water quality showed different sets of adaptation strategies as the best options, suggesting that our framework would help stakeholders seeking the robust options considering both the utility and regret.  相似文献

The performances and kinetic parameters of Fenton oxidation of 2,4- and 2,6-dinitrotoluene (DNT) in water-acetone mixtures and explosive contaminated soil washing-out solutions were investigated at a laboratory scale. The experimental results show that acetone can be a significant hydroxyl radical scavenger and result in serious inhibition of Fenton oxidation of 2,4- and 2,6-DNT. Although no serious inhibition was found in contaminated soil washing-out solutions, longer reaction time was needed to remove 2,4- and 2,6-DNT completely, mainly due to the competition of hydroxyl radicals. Fenton oxidation of 2,4- and 2,6-DNT fit well with the first-order kinetics and the presence of acetone also reduced DNT’s degradation kinetics. Based on the comparison and matching of retention time and ultraviolet (UV) spectra between high performance liquid chromatography (HPLC) and standards, the following reaction pathway for 2,4-DNT primary degradation was proposed: 2,4-DNT → 2,4-dinitro-benzaldehyde → 2,4-dinitrobenzoic acid → 1,3-dinitrobenzene → 3-nitrophenol.  相似文献

The fungal beta-D-glucan is a biological response modifier (BRM), but a major obstacle to the clinical utilization of beta-glucan BRMs is thei relative lack of solubility in aqueous media. Water insoluble fungal glucans extracted by alkali from the mycelia of Ganoderma lucidum were sulfated to yield their corresponding water-soluble derivatives. Insoluble glucan is dissolved in methyl sulfoxide and urea, and is partially sulfated with sulfuric acid. The sulfated glucan (SGL) yield prepared from insoluble glucan (IGL) was 85%, the sulfation degree of SGL was about 14.9%, and the solubility of SGL was above 95% in water. The monosugar SGL content was 34.9% alpha-glucose and 35.9% beta-glucose. The mean molecular weight (MW) of SGL was shown as a single peak on Sepharose CL-4B column chromatography, and their MW was approximately 9.3 kDa. The 13C NMR spectrum analysis shows that SGL has a high similarity with the beta-(1-->3)-linked triple-helical control.  相似文献

This study aims to investigate the anaerobic degradation kinetics of reactive dye, C.I. Reactive Red 141 (Evercion Red H-E7B) by partially granulated anaerobic mixed culture using three carbon sources, namely modified starch (MS), polyvinyl alcohol (PVA) and acrylic size (AS) during batch incubation. There is a first-order kinetics reaction in the decolorization processes using MS and PVA as carbon sources, while a zero-order kinetics relationship describes the decolorization process for the AS carbon source. The k values and color removal rate of decolorization with MS carbon source was higher than those of PVA and AS carbon sources. This is because the MS carbon source was well degraded in comparison to AS and PVA, respectively This study also found dye reduction could be enhanced through the addition of MS as a carbon source. The decolorization rates increased with decrease in dye concentrations of RR 141. In contrast, the decolorization rates increased with increase in COD concentration.  相似文献

In this study, concentrations of major aromatic VOCs were determined from landfill gas (LFG) at a total of five municipal landfill sites in Korea including Nan Ji (NJ), Woon Jung (WJ), Sam Poong (SP), Hoei Chun (HC), and No Hyung (NH). The concentration levels of those VOC were found to be significantly different, mainly as a function of such a parameter as landfill aging. The VOC concentrations measured from the unclosed landfill sites (e.g., WJ) were characterized by exceedingly high values above a few tens of ppm. However, the results of the abandoned site (e.g., SP) were about three orders of magnitude lower than the others so as to merely exceed the typical ambient concentration levels. It was most striking to find a systematic dominance of toluene over other aromatic VOC under most circumstances. The LFG flux values of all aromatic VOC and the four specific major ones (termed as BTEX: benzene, toluene, ethylbenzene, and xylene) were also computed for each vent pipe from all study sites using their concentrations and the concurrently determined environmental parameters. The results, if calculated in terms of the average BTEX quantity emitted per vent pipe, showed that the magnitude of their emissions can vary substantially, with the values ranging from 0.05 (SP) to 49.2 kg yr−1 (WJ in wintertime). The LFG flux values of aromatic VOC, when compared to the contribution of non-methane hydrocarbons (NMHC), were able to explain a constant, but minor, proportion of the LFG carbon budget.  相似文献

A new type of gaseous tracer utilizing nontoxic aliphatic alcohols for the determination of water content and air-water interfacial area is tested on unsaturated sands of low water content. Alcohol vapors are generated at room temperature and passed through the experimental sand column. Breakthrough curves (BTCs) of these vapors are obtained by monitoring their effluent concentrations using GC-FID. The retardation factor with respect to each vapor transport process is obtained by optimizing BTCs data using the CXTFIT program in the reverse problem mode. The water content and the interfacial area are subsequently calculated from their retardation factors by both equilibrium and nonequilibrium transport models. Experimental results indicate that the pentanol tracer is feasible in the determination of water content at conditions when the degree of water saturation is low. In the determination of air-water interfacial area, decanol is selected due to its interfacial adsorption characteristics. By comparing to interfacial areas from theoretical predictions as well as other conventional tarcer methods, the ones determined from the decanol tracer tests are found to be close to the true interfacial areas when the water content is low.  相似文献

In order to incorporate correctly the large or local scale circulation in the model, a nudging term is introduced into the equation of motion. Nudging effects should be included properly in the model to reduce the uncertainties and improve the air flow field. To improve the meteorological components, the nudging coefficient should perform the adequate influence on complex area for the model initialization technique which related to data reliability and error suppression. Several numerical experiments have been undertaken in order to evaluate the effects on air quality modeling by comparing the performance of the meteorological result with variable nudging coefficient experiment. All experiments are calculated by the upper wind conditions (synoptic or asynoptic condition), respectively. Consequently, it is important to examine the model response to nudging effect of wind and mass information. The MM5–CMAQ model was used to assess the ozone differences in each case, during the episode day in Seoul, Korea and we revealed that there were large differences in the ozone concentration for each run.These results suggest that for the appropriate simulation of large or small-scale circulations, nudging considering the synoptic and asynoptic nudging coefficient does have a clear advantage over dynamic initialization, so appropriate limitation of these nudging coefficient values on its upper wind conditions is necessary before making an assessment. The statistical verifications showed that adequate nudging coefficient for both wind and temperature data throughout the model had a consistently positive impact on the atmospheric and air quality field. On the case dominated by large-scale circulation, a large nudging coefficient shows a minor improvement in the atmospheric and air quality field. However, when small-scale convection is present, the large nudging coefficient produces consistent improvement in the atmospheric and air quality field.  相似文献
