Inadvertent selection is an important genetic process that frequently occurs during laboratory culture. The mass-reared strain
of the sweet potato weevil Cylas formicarius exhibits stronger inbreeding depression than the wild strain does. When inbreeding depression occurs in a population, mating
with a close relative is often considered maladaptive; however, in some contexts, the inclusive fitness benefits of inbreeding
may outweigh the costs, favoring individuals that tolerate a low level of inbreeding depression. Theory predicts that mass-reared
strain weevils will avoid inbreeding while wild strain weevils will tolerate inbreeding. To examine this prediction, we compared
the effect of relatedness on the mating and insemination successes in mass-reared and wild strains of C. formicarius. While close relative pairs of the wild strain copulated less frequently than non-kin pairs, almost all mass-reared strain
pairs copulated irrespective of relatedness. The results showed that the strain with weak inbreeding depression (wild strain)
avoided inbreeding, whereas the strain with strong inbreeding depression (mass-reared strain) tolerated inbreeding. The contradiction
between the theoretical prediction and our results is discussed from the perspective of laboratory adaptation, mating systems,

Toluene dioxygenase (tod) is a multicomponent enzyme system in Pseudomonas putida F1. Tod can mediate the degradation of Trichloroethylene (TCE), a widespread pollutant. In this study, we try to explore the TCE-regulated tod expression by using real-time qRT-PCR. The minimal culture media were supplemented with glucose, toluene, or a mixture of glucose/toluene respectively as carbon and energy sources. The TCE was injected into each medium after a 12-hour incubation period. The TCE injection severely affected bacterial growth when cultured with toluene or toluene/glucose mixtures. The cell density dropped 61 % for bacteria growing in toluene and 36 % for bacteria in the glucose/toluene mixture after TCE injection, but the TCE treatment had little effect on bacteria supplied with glucose alone. The decrease in cell number was caused by the cytotoxicity of the TCE metabolized by tod. The results from the real-time qRT-PCR revealed that TCE was capable of inducing tod expression in a toluene-dependent manner and that the tod expression level increased 50 times in toluene and 3 times in the toluene/glucose mixture after 6 hours of TCE treatment. Furthermore, validation of the rpoD gene as a reference gene for P. putida F1 was performed in this study, providing a valuable foundation for future studies to use real-time qRT-PCR in the analysis of the P. putida F1 strain. 相似文献

Lead can be recovered from funnel glass of waste cathode ray tubes via reduction melting. While low-temperature melting is necessary for reduced energy consumption, previously proposed methods required high melting temperatures (1400 °C) for the reduction melting. In this study, the reduction melting of the funnel glass was performed at 900–1000 °C using a lab-scale reactor with varying concentrations of Na2CO3 at different melting temperatures and melting times. The optimum Na2CO3 dosage and melting temperature for efficient lead recovery was 0.5 g per 1 g of the funnel glass and 1000 °C respectively. By the reduction melting with the mentioned conditions, 92% of the lead in the funnel glass was recovered in 60 min. However, further lead recovery was difficult because the rate of the lead recovery decreased as with the recovery of increasing quantity of the lead from the glass. Thus, the lead remaining in the glass after the reduction melting was extracted with 1 M HCl, and the lead recovery improved to 98%. 相似文献

An analytical method for the determination of gaseous and particulate chlorinated polycyclic aromatic hydrocarbons (ClPAHs) was investigated. By means of this method, concentrations and isomer profiles of the 27 target ClPAHs could be analyzed. To evaluate the usefulness of the method for analyzing ClPAH emissions, laboratory-scale scorching tests were performed on polyvinylidene chloride (PVDC) plastic wrap over a flame of the gas burner. Only seven of the target ClPAHs were detected, and all compounds detected had 2, 3, or 4 rings. The detected ClPAHs were present in both the particulate phase and the gaseous phase, but they were present at higher concentrations in the gaseous phase than in the particulate phase. Relationships between the number of chlorine substituents on the naphthalene/phenanthrene rings and the overall concentration and the percentage in the particulate phase were also investigated. 相似文献

Trace elements and butyltins in a Dall's porpoise (Phocoenoides dalli) from the Sanriku coast of Japan 总被引:1,自引:0,他引:1
Concentrations and body burdens of 14 trace elements (Hg, Cr, Mn, Co, Cu, Zn, Sr, Ag, Cd, V, Se, Pb, Mo, and Fe) and butyltins (BTs) (tributyltin TBT, dibutyltin DBT, and monobutyltin MBT) were determined in various tissues of a mature male Dall's porpoise (Phocoenoides dalli) collected off the Sanriku coast of Japan. Selective accumulation in this porpoise was observed for Hg, Mn, Cu, Ag, Mo, Fe, and total BTs (TBT, DBT, and MBT) in the liver, Cd in the kidney, Zn, Sr, V, Pb, and Co in the bone, and Se in the skin. In contrast, Cr concentrations in all tissues were similar. This distribution pattern in this mature porpoise was in general agreement with the accumulation characteristics of trace elements and butyltins reported for other marine mammals. The whole body of the porpoise contained approximately 62 g Fe, 8.8 g Zn, 4.0 g Sr, 0.6g Se, 0.41 g Cu, 0.19 g Hg, 0.17 g Cd, 0.16 g Mn, 0.05 g Cr, 0.009 g Ag, 0.008 g Mo, 0.005 g Pb, 0.004 g Co, and 0.7 mg of BTs (0.4 mg TBT, 0.2 mg DBT, and 0.1 mg MBT). Metabolism of TBT to its breakdown products of this porpoise seems to be limited, since TBT still accounted for about half of the total burden of BTs. As in the cases of Hg, Mn, Cu, Se, and Fe, the muscle was the most important reservoir (43%) for the whole body burden of total BTs, 80% of which was TBT, and thus muscle played a crucial role in the higher body composition of TBT in this Dall's porpoise. 相似文献

Parametric glass milling is presented to machine periodical circular channels on the glass plates for manufacturing micro testing devices. An end mill traverses in the linear motion during the workpiece rotation, which are synchronized by simultaneous control. The glass milling is controlled by 4 parameters in a mathematical model without NC program. Based on the principle of the parametric machining and the effect of the cutter axis inclination on the cutting process, a milling machine was developed to perform the parametric glass milling with an inclined ball end mill. The cutter axis inclination and the actual feed rate are associated with the critical feed rate, the maximum feed rate at which a crack-free surface is finished. Analysing soil microbial communities is often hampered by DNA adsorption on soil organic compounds such as humic acids. However
the role of humic acids in DNA adsorption and stability in soils remains controversial. To characterize DNA–humic acid interactions,
we studied DNA adsorption on two commercially available humic acids and a soil humic acid extracted from an Andosol. Desorption
of the adsorbed DNA using 4 different solutions—distilled water, 0.1 M NaCl, 0.1 M sodium phosphate buffer (pH 6.0), and 1%
sodium dodecyl sulfate solution—was also studied to understand the mechanism of DNA adsorption on humic acids. Here, we show
that humic acids play an important role in DNA adsorption to soils. DNA molecules were adsorbed on the humic acids, with adsorption
increasing proportionally with the DNA concentrations in the solution. The adsorption on all humic acid samples was fitted
with Freundlich equation, and the parameters obtained from the equation indicated a high affinity between the humic acids
and DNA molecules. The total amount of DNA desorbed by the 4 solutions was less than 2% of the total DNA adsorbed on all the
humic acids. The results demonstrate that DNA molecules are able to bind strongly to humic acids by ligand binding, hydrophobic

An alkaline comet assay and a micronucleus test were carried out on erythrocytes of the European chub, Squalius cephalus L., collected in spring and autumn in 2005 and 2006 at three sampling sites in River Sava, near Zagreb, Croatia. The results of comet assay showed the lowest genotoxic influence at the least polluted site, while higher DNA damage was observed at the polluted sites. Although the basal levels of DNA damage were elevated, a clear gradation of DNA damage was found due to pollution intensity in all sampling periods. The lowest cytogenetic damage as revealed by the micronucleus test (MNT) was observed as well at the least polluted site. High variations in MN frequency were observed between sampling periods, although the number of micronucleated erythrocytes was consistently the highest one at the polluted site. The comet assay as a biomarker of genotoxic effect exhibited higher sensitivity in discriminating the genotoxic capacity of studied polluted sites while the MNT was less sensitive. However, both tests should be used together in biomonitoring studies because they can reveal different aspects of DNA damage; comet assay, the early event of genotoxic exposure, and MNT, its final result as a mutagenic potential. 相似文献
