The inhibition of arylamine N‐acetyltransferase (NAT) activity by ellagic acid was determined in a human colon tumor (adenocarcinoma) cell line. Two assay systems were performed, one with cellular cytosols (9000g supernatant), the other with intact colon tumor cell suspensions. The NAT activity in a human colon tumor cell line was inhibited by ellagic acid in a dose‐dependent manner in both types of examined systems: i.e. the greater the concentration of ellagic acid in the reaction, the greater the inhibition of NAT activities in both systems. The data also indicated that ellagic acid decreased the apparent K m and V max of NAT enzymes from human colon tumor cells in both the systems examined. This report is the first demonstration which showed ellagic acid affect human colon tumor cell NAT activity.  相似文献

The epithelial-to-mesenchymal transition is the first step required for breast cancer to initiate metastasis. The aberrant activation of proto-oncogene tyrosine-protein kinase Src regulates multiple functions during tumor progression. Cardiotoxin III, a basic polypeptide isolated from Naja naja atra venom, has been shown to exhibit anticancer activity; however, the effect of cardiotoxin III on the epithelial-to-mesenchymal transition of cancer cells remains elusive. Exposure of MDA-MB-231 cells to cardiotoxin III resulted in morphological changes and upregulation of E-cadherin with a concomitant decrease in N-cadherin and vimentin protein levels, resulting in the inhibition of cell migration and invasion. Cardiotoxin III induced downregulation of snail and slug expression. Simultaneously, cardiotoxin III suppressed Src phosphorylation and downstream activation of focal adhesion kinase, of the docking protein p130cas, and of paxillin. In addition, cardiotoxin III inhibited the phosphorylation of extracellular signal-regulated kinase and phosphatidylinositol 3-kinase. The Src specific inhibitor 4-amino-5-(4-chlorophenyl)-7-(dimethylethyl)pyrazolo[3,4-d]pyrimidine decreased in the phosphorylation and the expression changes of epithelial-to-mesenchymal transition markers in a similar way. Thus, cardiotoxin III has a novel anticancer effect by suppressing proto-oncogene tyrosine-protein kinase activity, reversing epithelial-to-mesenchymal transition.  相似文献

In Asian countries such as China, Malaysia, Pakistan, India, Taiwan, Korea, Japan and Hong Kong, ambient air total suspended particulates and PM2.5 concentration data were collected and discussed during the years of 1998–2015 in this study. The aim of the present study was to (1) investigate and collect ambient air total suspended particulates (TSP) and PM2.5 concentrations for Asian countries during the past two decades. (2) Discuss, analyze and compare those particulates (TSP and PM2.5) annual concentration distribution trends among those Asian countries during the past two decades. (3) Test the mean concentration differences in TSP and PM2.5 among the Asian countries during the past decades. The results indicated that the mean TSP concentration order was shown as China > Malaysia > Pakistan > India > Taiwan > Korea > Japan. In addition, the mean PM2.5 concentration order was shown as Vietnam > India > China > Hong Kong > Mongolia > Korea > Taiwan > Japan and the average percentages of PM2.5 concentrations for Taiwan, China, Japan, Korea, Hong Kong, Mongolia and Other (India and Vietnam) were 8, 21, 6, 8, 14, 13 and 30%, respectively, during the past two decades. Moreover, t test results revealed that there were significant mean TSP and PM2.5 concentration differences for either China or India to any of the countries such as Taiwan, Korea and Japan in Asia during the past two decades for this study. Noteworthy, China and India are both occupied more than 60% of the TSP and PM2.5 particulates concentrations out of all the Asia countries. As for Taiwan, the average PM2.5 concentration displayed increasing trend in the years of 1998–1999. However, it showed decreasing trend in the years of 2000–2010. As for Korea, the average PM2.5 concentrations showed decreasing trend during the years of 2001–2013. Finally, the average PM2.5 concentrations for Mongolia displayed increasing trend in the years of 2004–2013.  相似文献

Local residents may have different views on disaster‐response modes depending on their cultural and socioeconomic background. The purpose of this study was to examine Taiwan residents' opinions on the Incident Command System (ICS). We performed a structured survey through face‐to‐face interviews in mudslide‐affected communities. Quantitative analysis showed that the residents exhibited a clear preference for the ICS core‐principle attributes of ‘integrated communications’, ‘transfer of command’ and ‘modular organisation’. By contrast, the residents tended towards a non‐ICS approach for ‘incident action plan’ and ‘manageable span of control’. Qualitative analysis revealed an uncertain attitude towards ‘transfer of command’ and ‘incident action plan’. Community acceptance is important in the promotion of the ICS. A better understanding of residents' preferences should be acquired through a broader community survey, allowing us to understand perspectives on the ICS among different societies and facilitate implementation of the ICS at the basic community level.  相似文献

In 2004, airborne particulate matter (PM) was collected for several aerosol episodes occurring in the southern region of Taiwan. The particulate samples were taken using both a MOUDI (Micro-orifice Uniform Deposit Impactor) and a nano-MOUDI sampler. These particulate samples were analyzed for major water-soluble ionic species with an emphasis to characterize the mass concentrations and distributions of these ions in the ambient ultrafine (PM0.1, diameter <0.1 μm) and nano mode (PMnano, diameter <0.056 μm) particles. Particles collected at the sampling site (the Da-Liao station) on the whole exhibited a typical tri-modal size distribution on mass concentration. The mass concentration ratios of PMnano/PM2.5, PM0.1/PM2.5, and PM1/PM2.5 on average were 1.8, 2.9, and 71.0%, respectively. The peak mass concentration appeared in the submicron particle mode (0.1 μm < diameter <1.0 μm). Mass fractions (percentages) of the three major water-soluble ions (nitrate, sulfate, and ammonium) as a group in PMnano, PM0.1, PM1, and PM2.5 were 18.4, 21.7, 50.0, and 50.7%, respectively. Overall, results from this study supported the notion that secondary aerosols played a significant role in the formation of ambient submicron particulates (PM0.1−1). Particles smaller than 0.1 μm were essentially basic, whereas those greater than 2.5 μm were neutral or slightly acidic. The neutralization ratio (NR) was close to unity for airborne particles with diameters ranging from 0.18 to 1 μm. The NRs of these airborne particles were found strongly correlated with their sizes, at least for samples taken during the aerosol episodes under study. Insofar as this study is exploratory in nature, as only a small number of particulate samples were used, there appears to be a need for further research into the chemical composition, source contribution, and formation of the nano and ultrafine mode airborne particulates.  相似文献

Fluctuation exposure of trace metal copper (Cu) is ubiquitous in aquatic environments. The purpose of this study was to investigate the impacts of chronically pulsed exposure on biodynamics and subcellular partitioning of Cu in freshwater tilapia (Oreochromis mossambicus). Long-term 28-day pulsed Cu exposure experiments were performed to explore subcellular partitioning and toxicokinetics/toxicodynamics of Cu in tilapia. Subcellular partitioning linking with a metal influx scheme was used to estimate detoxification and elimination rates. A biotic ligand model-based damage assessment model was used to take into account environmental effects and biological mechanisms of Cu toxicity. We demonstrated that the probability causing 50% of susceptibility risk in response to pulse Cu exposure in generic Taiwan aquaculture ponds was ~33% of Cu in adverse physiologically associated, metabolically active pool, implicating no significant susceptibility risk for tilapia. We suggest that our integrated ecotoxicological models linking chronic exposure measurements with subcellular partitioning can facilitate a risk assessment framework that provides a predictive tool for preventive susceptibility reduction strategies for freshwater fish exposed to pulse metal stressors.  相似文献

This study determines whether environment-protecting pervious pavement can dilute pollutants immediately after emissions from vehicle. The turbulence-driven dry-deposition process is too slow to be considered in this aspect. The pavement used is the JW pavement (according to its inventors name), a high-load-bearing water-permeable pavement with patents in over 100 countries, which has already been used for more than 8 years in Taiwan and is well suited to replacing conventional road pavement, making the potential implementation of the study results feasible. The design of this study included two sets of experiments. Variation of the air pollutant concentrations within a fenced area over the JW pavement with one vehicle discharging emissions into was monitored and compared with results over a non-JW pavement. The ambient wind speed was low during the first experiment, and the results obtained were highly credible. It was found that the JW pavement diluted vehicle pollutant emissions near the ground surface by 40%-87% within 5 min of emission; whereas the data at 2 m height suggested that about 58%-97% of pollutants were trapped underneath the pavement 20 min after emission. Those quantitative estimations may be off by +/- 10%, if errors in emissions and measurements were considered. SO2 and CO2 underwent the most significant reduction. Very likely, pollutants were forced to move underneath due to the special design of the pavement. During the second experiment, ambient wind speeds were high and the results obtained had less credibility, but they did not disprove the pollutant dilution capacity of the JW pavement. In order to track the fate of pollutants, parts of the pavement were removed to reveal a micro version of wetland underneath, which could possibly hold the responsibility of absorbing and decomposing pollutants to forms harmless to the environment and human health.  相似文献

One of the major challenges in assessing the potential metal stress to aquatic organisms is explicitly predicting the internal dose in target organs. We aimed to understand the main sources of copper (Cu) accumulation in target organs of tilapia (Oreochromis mossambicus) and to investigate how the fish alter the process of Cu uptake, depuration, and accumulation (toxicokinetics (TK)) under prolonged conditions. We measured the temporal Cu profiles in selected organs after single and combined exposure to waterborne and dietary Cu for 14 days. Quantitative relations between different sources and levels of Cu, duration of treatment, and organ-specific Cu concentrations were established using TK modeling approaches. We show that water was the main source of Cu in the gills (>94 %), liver (>89 %), and alimentary canal (>86 %); the major source of Cu in the muscle (>51 %) was food. Cu uptake and depuration in tilapia organs were mediated under prolonged exposure conditions. In general, the uptake rate, depuration rate, and net bioaccumulation ability in all selected organs decreased with increasing waterborne Cu levels and duration of exposure. Muscle played a key role in accounting for the rapid Cu accumulation in the first period after exposure. Conversely, the liver acted as a terminal Cu storage site when exposure was extended. The TK processes of Cu in tilapia were highly changed under higher exposure conditions. The commonly used bioaccumulation model might lead to overestimations of the internal metal concentration with the basic assumption of constant TK processes.  相似文献

ABSTRACT

This study investigated the pyrolysis characteristics of sludge from wastewater treatment plants in the petrochemical industry and focused on the pyrolysis kinetics, elemental composition of residue, and volatile organic compounds (VOCs) of exhaust gas. As pyrolysis temperature increased to 773 K, the increasing rate of crude oil production tended to a stable condition. The result indicated that the optimal temperature of crude oil and water mixed production was 773 K. When pyrolysis temperature increased from 673 to 973 K, carbon, oxygen, nitrogen, and hydrogen concentrations of residue decreased and the sulfur concentration of residue increased. The concentrations of benzene, toluene, ethylbenzene, and styrene increased by the increasing pyrolysis temperature. We found that the reaction order of sludge pyrolysis was 2.5 and the activation energy of the reaction was 11.06 kJ/mol. We believe that our pyrolysis system is transitional between devolatilization and combustion.  相似文献

ABSTRACT

Acidic aerosol concentrations measured by an annular denuder system (ADS) and a honeycomb denuder system (HDS) in Hsinchu, Taiwan, were compared. Aerosols were also sampled by a MOUDI (micro-orifice uniform deposit impactor) and analyzed by an ion chromatograph to determine the size distributions of different species. Using the measured aerosol size distribution, theoretical analysis showed that positive HNO3 artifact due to volatiliza-tion of NH4NO3 is generally negligible for both samplers.4 3Comparing two different denuder samplers, the average concentration of HNO3 measured by the ADS was found3to be lower than that measured by the HDS, while the difference between the two samplers for the average concentration of other species was found to be within ±15%. A possible cause of the difference in HNO3 con-3centrations is due to a greater loss of HNO3 in the cyclone3 used by the ADS than in the impactor used by the HDS. The study also showed incomplete absorption of the evaporated HCl and HNO3 from the particles on the Teflon3filter by the first nylon filter in the filter pack of the ADS. Collection efficiency and capacity of HCl and HNO3 by3the nylon filters need further investigation.  相似文献
