The switch from within-hive activities to foraging behavior is a major transition in the life cycle of a honeybee (Apis mellifera) worker. A prominent regulatory role in this switch has long been attributed to juvenile hormone (JH), but recent evidence also points to the yolk precursor protein vitellogenin as a major player in behavioral development. In the present study, we injected vitellogenin double-stranded RNA (dsVg) into newly emerged worker bees of Africanized genetic origin and introduced them together with controls into observation hives to record flight behavior. RNA interference-mediated silencing of vitellogenin gene function shifted the onset of long-duration flights (>10 min) to earlier in life (by 3–4 days) when compared with sham and untreated control bees. In fact, dsVg bees were observed conducting such flights extremely precociously, when only 3 days old. Short-duration flights (<10 min), which bees usually perform for orientation and cleaning, were not affected. Additionally, we found that the JH titer in dsVg bees collected after 7 days was not significantly different from the controls. The finding that depletion of the vitellogenin titer can drive young bees to become extremely precocious foragers could imply that vitellogenin is the primary switch signal. At this young age, downregulation of vitellogenin gene activity apparently had little effect on the JH titer. As this unexpected finding stands in contrast with previous results on the vitellogenin/JH interaction at a later age, when bees normally become foragers, we propose a three-step sequence in the constellation of physiological parameters underlying behavioral development. Several studies have proven that natural or genetically modified bacteria, such as Pseudomonas putida strain, degrade recalcitrant organic sulfur compounds. However, from a practical point of view, the biodesulfurization (BDS) process has to be performed with really high proportions of organic solvents. In this work, the dibenzothiophene (DBT) was selected as recalcitrant model compound, and hexadecane as model organic solvent. It has been observed that P. putida CECT 5279 was able to desulfurize DBT even in the presence of 50% (v/v) of hexadecane. A concentration of 400 ppm of DBT was converted at a specific rate of generation of desulfurized final product, 2-hydroxybiphenyl (HBP), of 2.3 and 1.5 mg HBP L-1 (g DC L-1 h)-1 for 27% and 50% (v/v) of hexadecane, respectively. Finally, the Haldane kinetic model was used to describe the process evolution. The study is relevant as it has been proven that the strain CECT 5279 is a potential biocatalyst for developing an efficient BDS process.  相似文献

Assessment of carcinogenic heavy metal levels in Brazilian cigarettes   总被引:1,自引:0,他引:1  
Total mercury (Hg(T)) and bioavailability Hg (Hg(HCl)) concentrations in soil were determined in five districts in Wuhu urban area. Spatial pattern of soil Hg concentration was generated through kriging technology. Results showed that Hg concentration in soil ranged from 0.024 to 2.844 mg kg(?-1) with an average of 0.207 mg kg(?-1). Hg concentration in soil appeared to have a block distribution and decreased from downtown to surrounding district. And Hg concentrations appeared to have a medium scale spatial auto correlation, strongly affected by human activity. The maximal Hg average concentration (0.332 mg kg(?-1)) in soil appeared in Jinghu district, where the high intensity of human activities is. Second highest Hg average concentration (0.263 mg kg(?-1)) in soil appeared in development district, where the intensive industrial activities are. Bioavailability Hg concentration in soil ranged from 2.6 to 4.9 μg kg(?-1) with an average of 3.8 μg kg(?-1), which had a ratio of 0.28~6.44% to total Hg. The ratios of bioavailability Hg to total Hg in vegetable soil were bigger than those of park soil. Correlation analysis showed that total Hg, organic matter, total phosphorus, and bioavailability Hg concentrations in soil were significantly positively correlated. Hg concentration in vegetable ranged from 2.7 to 15.2 μg kg(?-1) with an average of 6.5 μg kg(?-1). Hg concentration in vegetable was positively correlated with Hg(HCl) concentration in soil. According to the calculation on hazard quotient (HQ) for children, inhalation of Hg vapor from soil is the main exposure pathway, in which HQ is 2.517 × 10(?-2), accounting for 80.3% of the four exposure pathways. Hazard index (HI) of the four exposure pathways is lower than the "safe" level of HI = 1; therefore, exposure of soil Hg exhibited little potential health risk to children in Wuhu urban area.  相似文献

The presence of polycyclic aromatic hydrocarbon (PAH) compounds in sediment and water samples collected in the estuary area of Paranagua, southern Brazil, was investigated. There is a lot of port activity in the region. Recreational fishing is widespread; thus, there is concern about possible contamination by PAHs. The 16 priority PAHs were investigated, and only eight were found. The total concentration of PAHs ranged from 40.8 to 406.8 ng/g. High molecular weight were the most abundant, while PAHs with a low molecular weight were absent. There are suspicions that the main source of PAHs is combustion, but some uncertainties exist, and there may even be the presence of PAHs resulting from accidental spills of crude oil. Although the sediments contain PAHs, the amount is below the maximum concentrations allowed by the Brazilian environmental legislation, as well as the maximum levels at which adverse effects are observed. From the analytical results, a probable bioaccumulation was assessed in the local trophic chain using a mathematical model (Arnot and Gobas, Environ Toxicol Chem 23(10):2343?C2355, 2004). The model showed that there is a possibility of biomagnification along the food chain selected. Three fishes with high local consumption were selected, and the concentration of some PAHs could be found in those fishes.  相似文献

Perfluorinated chemicals (PFCs) are extremely persistent and have been found extensively in the environment and wildlife. Oceans are the final sink for many persistent organic pollutants (POPs) including PFCs. However, to date, there has been a lack of studies that investigated the environmental consequences of PFCs on marine organisms. To fill in this gap, environmental toxicity of two dominant PFCs, perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA), was examined in a sentinel species, green mussel Perna viridis, using a series of biomarkers corresponding to different biological levels (molecular, cellular, and physiological). Correlations among these biomarkers were also investigated. The results showed that the tested compounds can induce a series adverse effect at different biological levels, including oxidative stress, DNA damage, membrane instability, suppressed filtration rate, and reduced body weight. Correlation analysis revealed that excess production of reactive oxygen species could be the major toxic pathway. An indirect mode of toxic action was also explored where adverse impacts could be secondary effects of PFC exposure. The joint analysis of biomarkers from multiple biological levels resulted in a comprehensive understanding of how PFC exposure can influence the health of organisms. The correlations of these biomarkers also provided a new perspective of the ecological consequences of PFCs.  相似文献
