We undertook a detailed analysis of the lipid composition ofSolemya velum (Say), a bivalve containing endosymbiotic chemoautotrophic bacteria, in order to determine the presence of lipid biomarkers of endosymbiont activity. The symbiont-free clamMya arenaria (L.) and the sulfur-oxidizing bacteriumThiomicrospira crunogena (Jannasch et al.) were analyzed for comparative purposes. The 13C ratios of the fatty acids and sterols were also measured to elucidate potential carbon sources for the lipids of each bivalve species. Both fatty acid and sterol composition differed markedly between the two bivalves. The lipids ofS. velum were characterized by large amounts of 18: 17 (cis-vaccenic acid), 16:0, and 16 : 17 fatty acids, and low concentrations of the highly unsaturated plant-derived fatty acids characteristic of most marine bivalves. Cholest-5-en-3-ol (cholesterol) accounted for greater than 95% of the sterols inS. velum. In contrast,M. arenaria had fatty acid and sterol compositions similar to typical marine bivalves and was characterized by large amounts of the highly unsaturated fatty acids 20 : 53 and 22 : 63 and a variety of plant-derived sterols. The fatty acids ofT. crunogena were similar to those ofS. velum and were dominated by 18:17, 16:0 and 16:17 fatty acids. Thecis-vaccenic acid found inS. velum is almost certainly symbiontderived and serves as a potential biomarker for symbiontlipid incorporation by the host. The high concentrations ofcis-vaccenic acid (up to 35% of the total fatty acid content) in both symbiont-containing and symbiont-free tissues ofS. velum demonstrate the importance of the endosymbionts in the lipid metabolism of this bivalve. The presence ofcis-vaccenic acid in all the major lipid classes ofS. velum demonstrates both incorporation and utilization of this compound. Axenic tissue cultures ofRuppia maritima L. were established and propagated clonally in vitro from terminal rhizome segments collected from Tampa Bay, Florida, USA. Cultures were maintained in a base medium consisting of synthetic seawater supplemented with half-strength Murashige and Skoog salts and 1% sucrose at pH 5.6. The effects of five cytokins [6-furfurylaminopurine (kinetin), 6-benzylaminopurine (BAP), 2-isopentyladenine (2iP), 6-(4-hydroxy-3-methyl-but-2-enylamino) purine (zeatin), andn-phenyl-n-1,2,3-thidiazol-5yl urea (thidiazuron)] and one auxin [napthalene acetic acid (NAA)] on explant growth and development were investigated. Cytokinin additions resulted in a 3- to 4-fold increase in nodal production, branching, and biomass ofR. maritima after 12 wk in culture. Cultures responded in a dose-dependent manner to 2iP but exhibited broad dose-response curves to kinetin, BAP, zeatin, and thidiazuron. NAA addition resulted in increased leaf and internodal lengths, but reduced the number of leaves per node and the rhizome biomass. The addition of NAA almost completely suppressed root growth in media without cytokinins and had an antagonistic effect on nodal production and branching in cytokinin-supplemented media.  相似文献

The relative tissue accumulation of Pb, Cd and Zn were compared in two populations each of two species of slugs (Arion subfuscus and Deroceras reticulatum). One population was resident at a contaminated Pb/Zn mine site, and the other population was from an uncontaminated site and was transferred for 20 days to microcosms established at the mine site. It was found that when the experiment was conducted during late spring (May), but not in late winter (February), that the Pb and Zn concentrations in the tissues of the 'transplants', were significantly higher than in the tissues of 'residents'; the Cd concentrations in the transplants, although significantly higher in the May sample than in February, did not exceed those of their 'resident' counterparts. It was postulated that: (a) Pb and Zn tolerance in slugs is phenotypically expressed by a reduction in metal accumulation; (b) Cd tolerance, if present, may be characterized by enhanced storage capacity; and (c) the presence of local metal-tolerant ecotypes is a biotic variable that may confound the relationship between dry tissue and environmental metal concentrations that forms a basis of pollution biomonitoring.  相似文献

The translocation of some heavy metals, such as Hg, Cd, Cu, Co, Ni and Zn, as affected by organic pollutants, i.e. methanol, ethanol, propanol, formaldehyde, acetaldehyde, benzaldehyde, acetone, ethyl methyl ketone and cyclo-hexanone, was studied in an Indian red soil using soil thin layer chromatography. It was observed that an increase in the concentration of organic compounds in developer enhances the heavy metal mobility, except in the case of Cu and Hg which show a decreasing trend. The results are discussed in relation to the physico-chemical characteristics of the soil and adsorption/desorption phenomena.  相似文献
