利用二苯碳酰二肼显色,Triton X-100-正辛醇浊点萃取,建立了一种分光光度法测定水中Cr(Ⅵ)的新方法,并探讨了不同测定条件对测定效果的影响。优化后的测定条件为(总体积50 mL)1 mol/L稀硫酸加入量1.0mL、2 g/L二苯碳酰二肼溶液加入量1.5 mL、10 g/L Triton X-100溶液加入量1.5 mL、3 g/L苯甲酸溶液加入量4.0mL、正辛醇加入量5.0 mL。Cr(Ⅵ)质量浓度在0~20μg/L范围内符合朗伯-比尔定律,线性回归方程的相关系数为0.995 5。该方法应用于水样中痕量Cr(Ⅵ)的测定,相对标准偏差小于2.5%,加标回收率为95.7%。  相似文献

The toxic effects of tributyltin (TBT) have been extensively documented in several types of cells, but the molecular mechanisms related to the genotoxic effects of TBT have still not been fully elucidated. Our study showed that exposure of human hepatoma G2 cells to 1-4 μmol/L TBT for 3 hr caused severe DNA damage in a concentration-dependent manner. Moreover, the expression levels of key DNA damage sensor genes such as the replication factor C, proliferating cell nuclear antigen and poly (ADP-ribose) polymerase-1 were inhabited in a concentration-dependent manner. We further demonstrated that TBT induced cell apoptosis via the p53-mediated pathway, which was most likely activated by the ataxia telangiectasia mutated and rad-3 related (ATR) protein kinase. The results also showed that cytochrome c, caspase-3, caspase-8, caspase-9, and the B-cell lymphoma 2 were involved in this process. Taken together, we demonstrated for the first time that the inhibition of the DNA repair system might be more responsible for TBT-induced genotoxic effects in cells. Then the generated DNA damage induced by TBT initiated ATR-p53-mediated apoptosis.  相似文献

The feasibility of copper recovery by induced crystallization in a long period(174 days) was investigated in a seeded fluidized bed reactor(FBR). The process was divided into 3 periods according to different influent conditions, and the period III was separated into III-a and IIIb due to the adjustment of the molar ratio of CO_3~(2-)concentration to copper concentration([CT]/[Cu~(2+)]). The removal efficiency could exceed 95% and the average effluent copper concentration decreased to 3.0 mg/L. The mean particle size of seed grains with copper crystals coating on, raised to 0.36 mm from initial 0.18 mm. During period III-a, the supersaturation exceeded 2.88 × 104, the removal efficiency decreased to 60%–80% and the particle size dropped to 0.30 mm, due to the generation of fines by homogeneous crystallization and seeds breaking. And the morphology of the crystals on the seed grains changed from rod-like to spherical which lead to the particle size decreasing. In period III-b,the supersaturation was modified by adjusting the molar ratio of [CT]/[Cu2+] to 1.2 from 2.The efficiency was back to 95% and the mean particle size grew to 0.36 mm at the end of IIIb, the crystals coating on the seeds turned back to rod-like products of good stability. This study illustrated that the copper salt crystal could keep on growing on the seed grains for over 150 days, the feasibility and controllability of copper recovery by induced crystallization process in FBR were satisfactory, even under the dramatic changes in influent conditions.  相似文献

由编号为RL-2的短芽孢杆菌(Bacillus brevis)产生的生物高分子,经研究发现是一种对高岭土悬液有较高絮凝活性的阴离子型絮凝剂.对絮凝剂的热处理及酶处理表明,其活性成分为多糖;经茚三酮试验,Molish反应,薄层色谱及红外光谱分析表明,该絮凝剂为一种阴离子型多糖絮凝剂;ζ电位测定及絮凝剂与高岭土颗粒之间结合键的检测表明,絮凝剂与高岭土颗粒间的结合力为氢键,絮凝过程基于"桥联"机理;絮凝剂和高岭土在絮凝剂的活性部位--多糖中的羟基或羧基以氢键相结合,经架桥作用絮凝沉淀.  相似文献

A new method has been developed to determine heptachlor and its metabolites heptachlor-exo-epoxide and heptachlor-endo-epoxide in pork. The pork samples were extracted with acetone–n-hexane (2:8, V:V) and cleaned up by gel permeation chromatography and florisil solid-phase extraction cartridge. The extract was then determined by gas chromatography equipped with electron capture detector (GC-ECD), followed by validation using gas chromatography–mass spectrometry (GC–MS) with negative chemical ionization. Linearity of calibration curves ranged from 0.01 to 0.5 mg L?1, with correlation coefficients of more than 0.9980 for GC-ECD and GC–MS, respectively. At spiked concentrations of 0.01, 0.05, and 0.1 mg kg?1, the average recovery and relative standard deviation values were 87.1–102.2 and 4.0–11.3 %, respectively. The limit of quantification for each analyte was 0.01 mg kg?1, which satisfied the current maximum residue limit permitted in pork. Our results showed that the method developed was successfully used to determine heptachlor and heptachlor epoxide residues in real pork samples.  相似文献

Heavy metal concentration in soil was investigated at three sites with different topography (cut slope, flat and embankment) within the vicinity of Chengdu-Kunming railway in Sichuan, China. Surface soil was sampled at certain distances from the track at each site and was analyzed for Cu, Mn, Pb, Zn, Cd by atomic absorption spectrometry. Cu, Cd and Zn concentrations in some soil exceeded the thresholds for non-polluted soil following the soil quality standard set by the State Environmental Protection Agency of China. Compared to local background values, the highest enrichment factor values of Cu, Mn, Zn and Cd were 2.7, 3.4, 3.7 and 7.7, respectively, indicating a moderate or significant enrichment of these metals in soil closest to the railway. Pb showed little accumulation with the EF values generally nearer 1 at the chosen sites. Topography profile was found to influence metal levels and distribution in soil alongside railway. At the cut slope site, Mn, Zn, Cd showed the highest concentrations and the smallest dispersion distance of 2 m, while Cu showed further dispersion distance of 25 m due to a main Cu emission source, the head-over traction cable, being located higher than any other metal emission source (wheels and tracks). Heavy metal concentrations decreased conversely as compared to distance from the track, peak values occurring at locations closest to the tracks, whilst embankment site soil Cd concentrations peaked at distances of 25 m. Significant correlation was found amongst Mn, Cu, Zn and Cd, which indicates that these metals have the same anthropogenic origin there. Organic matter content had no significant correlation to the elements Mn, Cu and Zn, which implies relatively high mobility to those metals.  相似文献

It is planned that the Dayanghan Wetland in China will be transformed into a national park but little is known about its current water quality and pollution status. Thus, we monitored the physical and chemical characteristics of the Dayanghan Wetland, which showed that the water quality was generally good. However, the chemical oxygen demand was more than double the reference value, which may be attributable to previous tillage for vegetable crops and other farmlands. In addition, nickel and chromium caused low-level pollution in the water bodies of the Dayanghan Wetland. The mean trophic level index and nutrient quality index were 39.1 and 2.69, respectively. Both indices suggest that the water bodies of the Dayanghan Wetland are in a mesotrophic state and that no eutrophication has occurred. The study would provide a precise report on the status of environmental quality of the water bodies of a typical pre-construction wetland for the administration and decision of the local government and the planning agent.  相似文献
