Delta-aminolevulinate dehydratase (ALAD) activity has been used in prokaryotes and eukaryotes as a biomarker for environmental lead (Pb) exposure and toxicity. Microorganisms are sensitive indicators of toxicity at the fundamental level of ecological organization, but bacterial biomarker studies are focused on the Pseudomonas strains in Group I and E coli. The objectives of the present work were to determine if Burkholderia gladioli belonging to group II, due to its 16SrRNA similarity, can be used as biomarker in metal contamination and compare its possible usage with Pseudomonas aeruginosa and Citrobacter freundii (previously known as Esherichia freundii) and Bacterium freundii which are classified in Group I. In this study, ALAD activity in an environmental strains of Burkholderia gladioli, Pseudomonas aeruginosa, Citrobacter freundii were investigated to evaluate potential inhibition by Pb and other toxic metals. When the ALAD activity of Burkholderia gladioli was tested, Co and Pb decreased activity by 27 and 71%, respectively. In addition to these findings, Zn increased the activity up to 26%. These effects were found to be statistically meaningful (p < 0.05). It was determined that the increase of lead concentration inhibites the ALAD activity at each of the three strains. There was a statistically significant dose–response relationship between ALAD activity in cells of Burkholderia gladioli and Pb (Pearson correlation coefficent = −0.665; r 2 = 0.665, and p < 0.001). The strongest ALAD inhibition which was measured was 90% at Burkholderia gladioli when protein extracts were incubated with 750 μM of Pb. The relationship between Pb and ALAD activity was statistically described by [ALAD Activity] = 0.476−0.000597 × [Pb]. According to the obtained results, we suggest that the ALAD of Burkholderia gladioli can be used as a biomarker for lead contamination in the environment.  相似文献

El Niño, an interannual climate event characterized by elevated oceanic temperature, is a prime threat for coral reef ecosystems worldwide, owing to their thermal threshold sensitivity. Phytoplankton plays a crucial role in the sustenance of reef trophodynamics. The cell size of the phytoplankton forms the “master morphological trait” with implications for growth, resource acquisition, and adaptability to nutrients. In the context of a strong El Niño prediction for 2015–2016, the present study was undertaken to evaluate the variations in the size-structured phytoplankton of Kavaratti reef waters, a major coral atoll along the southeast coast of India. The present study witnessed a remarkable change in the physicochemical environment of the reef water and massive coral bleaching with the progression of El Niño 2015–2016 from its peak to waning phase. The fluctuations observed in sea surface temperature, pH, and nutrient concentration of the reef water with the El Niño progression resulted in a remarkable shift in phytoplankton size structure, abundance, and community composition of the reef waters. Though low nutrient concentration of the waning phase resulted in lower phytoplankton biomass and abundance, the diazotroph Trichodesmium erythraeum predominated the reef waters, owing to its capability of the atmospheric nitrogen fixation and dissolved organic phosphate utilization.  相似文献

In this study, the relationship among water quality, soil properties, and plant coverage in the region of the Akarçay stream was examined. Correlation analyses were carried out between soil samples taken from each of four plant communities in the Akarçay basin and water in the Akarçay stream. The four plant communities in the study area are as follows: Limonium lilacinum (Boiss. et Bal.) Wag., Alhagi pseudalhagi (M. Bieb.) Desv. Peganum harmala L., and Hordeum marinum Huds. subsp. marinum. B, Cl, EC, K, Mg, Na, pH, and SO4 data from both soil and water samples were subjected to statistical analysis, and significant correlations were obtained (p < 0.05). These correlations indicated that the chemical features of the soil had a major effect on water quality. The important parameters were B, Cl, EC, K, Mg, Na, pH, and SO4 for Limonium lilacinum communities; Ca, K, and pV for Peganum harmala; and B, Cl, Mg, pH, and pV for Alhagi pseudalhagi. There were also statistically significant relationships (p < 0.05) among the parameters examined. These findings strongly suggested that these plant communities can be used as indicators for soil chemistry and water quality.  相似文献

In this study, the effect of Afyonkarahisar Sugar Factory's discharge water on germination percentage, root growth and mitotic divisions of the root tip cells of Hordeum vulgare L. were investigated. Six concentrations of wastewater and ranging from 10(0), 10(?-1), 10(?-2), 10(?-3), 10(?-4), 10(?-5), were applied for 6, 12, 18 and 24 h, respectively. It was observed that the treatments reduced the germination percentages of H. vulgare grains and inhibited the root growth as well as reduced mitotic index compared to the control group at all concentrations. It was also observed that the increase of the concentrations of wastewater decreased the cell division, and several mitotic anomalies such as c-mitosis, lagging chromosomes, multipolar anaphases and chromosome bridges increased.  相似文献

δ-Aminolevulinic acid dehydratase (ALAD; E.C. 4.2.1.24) is a metalloprotein and plays a crucial role in heme synthesis. Its sensitivity to toxic metals depends on the metallic co-factors. In this study the effects of some heavy metals on ALADs activity of five Pseudomonas isolates from Akar?ay stream (Afyonkarahisar) have been studied in order to determine whether their ALADs could be used as biosensor for lead and other heavy metals contamination. The data obtained from the study were analysed statistically by using SPSS 10.0 software for Windows. According to the results, Ni(II) increased the ALAD activity of Pseudomonas putida, Pseudomonas pseudoalcaligenes and Pseudomonas aureginosa ATTC 27853. Mn(II) also increased the enzymic activity in all strains examined except P. pseudoalcaligenes. These were found not to be statistically meaningful. P. aeruginosa 2’s enzymic activity was inhibited by Mg(II) and Zn(II), significantly (p’lt;’.05). There was a statistically meaningful relation between enzymic activity of both P. pseudoalcaligenes, P. putida and increasing Pb(II) concentration (p’lt;’.05). In addition, a formula was also deviced in order to determine the doses of metals in the environment of the organisms. As a result of the study, we can suggest that Pseudomonas ALADs can be used as a biosensor for lead and some other heavy metal exposure in aquatic environments.  相似文献

This study was carried out to determine some bioclimatic characteristics such as humidity category (Q2), winter variant (m), the length of the dry season (LDS) and the dry season water deficit (DSWD) of naturally growing two endemic oak taxa, Quercus macranthera subsp. syspirensis and Q. petraea subsp. pinnatiloba, living in Turkey. Our findings showed that bioclimatic tolerance range of Q. macranthera subsp. syspirensis possess 7 different types of Mediterranean bioclimate while Q. petraea subsp. pinnatiloba had 8 of them. Although Q. macranthera subsp. syspirensis was ranging among the semiarid, freezing and very cold, Q. petraea subsp. pinnatiloba was among sub-humid, freezing and very cold ambient. It was briefly established that Q. macranthera subsp. syspirensis prefers semi-arid and very cold/freezing conditions and Q. petraea subsp. pinnatiloba prefers sub-humid and cold/very cold climatic conditions.  相似文献

In this study, the levels of some heavy metals (Al, Cd, Co, Cr, Cu, Fe, Li, Mn, Ni, Pb and Zn) in muscle, gill, and liver of Carassius carassius and in the water samples from Eber Lake (Afyonkarahisar, Turkey) have been investigated. Additionally, one of the lipid peroxidation markers, malondialdehide (MDA) and glutathion (GSH) levels were investigated. All the metal analysis was performed by using ICP-AES. According to results obtained, it was observed that heavy metals were accummulated in liver in the highest degree and lowest one in the muscle tissues. MDA and GSH levels varied in the seasons but their winter levels were found to be statistically meaningful when compared with other seasons. The obtained data of metals from the study were compared with the acceptable levels of Turkish governmental regulation and they were found not to be harmful for human health. On the other hand, it was suggested that oxidative stress markers should be checked regularly in order to get important data for continuous life of aquatic organisms.  相似文献

In this study, the potential re-use of waste foundry sand in high-strength concrete production was investigated. The natural fine sand is replaced with waste foundry sand (0%, 5%, 10%, and 15%). The findings from a series of test program has shown reduction in compressive and tensile strengths, and the elasticity modulus which is directly related to waste foundry inclusion in concrete. Nevertheless the concrete with 10% waste foundry sand exhibits almost similar results to that of the control one. The slump and the workability of the fresh concrete decreases with the increase of the waste foundry sand ratio. Although the freezing and thawing significantly reduces the mechanical and physical properties of the concrete. The obtained results satisfies the acceptable limits set by the American Concrete Institute (ACI).  相似文献
