Variations in the gene encoding the low-density lipoprotein receptor (LDLR) can cause familial hypercholesterolemia (FH), one of the most common inherited metabolic disorders in humans. The functional
effects of the p.Gln92Glu and p.Asn564His alterations are predicted as benign, but the c.313 + 1G>C and p.Lys799_Phe801del
changes are believed to cause disease. Although p.Gln92Glu and c.313 + 1G>C have been observed only in Spain, p.Asn564His
and p.Lys799_Phe801del are widespread in Western Europe. In order to estimate the ages (t generations) of these four variants of the gene, to determine their possible origin and to consider the influence of age
and selective pressure on their spread, we analyzed 86 healthy individuals and 126 FH patients in Spain. Most of the FH patients
investigated carried two of these four LDLR variants simultaneously, while only one patient carried three of them simultaneously. Haplotype analyses were based on five
LDLR SNPs: c.81T>C, c.1413G>A, c.1725C>T, c.1959T>C and c.2232G>A. The results suggest that p.Gln92Glu and c.313 + 1G>C arose
at about the same time (99 and 103 generations ago, respectively) in the CACTG haplotype and that p.Asn564His and p.Lys799_Phe801del
appeared in the CGCCG haplotype and might be slightly more recent variations (92 and 95 generations ago, respectively). Low
selective pressures could explain the maintenance of these variants in spite of their ages. The origin of p.Gln92Glu and c.313 + 1G>C
appears to be in Spain whereas p.Asn564His and p.Lys799_Phe801del could have been introduced in Spain by Celtic migrations

Water is scarce in Mediterranean countries: cities are crowded with increasing demand; food is produced with large amounts of water; ecosystems demand more water that is often available; drought affects all. As climate change impacts become more noticeable and costlier, some current water management strategies will not be useful. According to the findings of CIRCE, the areas with limited water resources will increase in the coming decades with major consequences for the way we produce food and we protect ecosystems. Based on these projections this paper discusses water policy priorities for climate change adaptation in the Mediterranean. We first summarise the main challenges to water resources in Mediterranean countries and outline the risks and opportunities for water under climate change based on previous studies. Recognising the difficulty to go from precipitation to water policy, we then present a framework to evaluate water availability in response to natural and management conditions, with an example of application in the Ebro basin that exemplifies other Mediterranean areas. Then we evaluate adaptive capacity to understand the ability of Mediterranean countries to face, respond and recover from climate change impacts on water resources. Social and economic factors are key drivers of inequality in the adaptive capacity across the region. Based on the assessment of impacts and adaptive capacity we suggest thresholds for water policy to respond to climate change and link water scarcity indicators to relevant potential adaptation strategies. Freshwater resources are under stress due to naturally occurring conditions and human impacts. One of the consequences is the proliferation of cyanobacteria, microphytoplankton organisms that are capable to produce toxins called cyanotoxins. Anatoxin-a is one of the main cyanotoxins. It is a very potent neurotoxin that was already responsible for some animal fatalities. In this review we endeavor to divulgate much of the internationally published information about toxicology, occurrence and detection methods of anatoxin-a. Cyanobacteria generalities, anatoxin-a occurrence and production as well as anatoxin-a toxicology and its methods of detection are the aspects focused in this review. Remediation of anatoxin-a occurrence will be addressed with a public health perspective. Final remarks call the attention for some important gaps in the knowledge about this neurotoxin and its implication to public health. Alterations of aquatic ecosystems caused by anatoxin-a is also addressed. Although anatoxin-a is not the more frequent cyanotoxin worldwide, it has to be regarded as a health risk that can be fatal to terrestrial and aquatic organisms because of its high toxicity. 相似文献

Microorganisms are the most ancient cells on this planet and they include key phyla for understanding cell evolution and Earth
history, but, unfortunately, their microbial records are scarce. Here, we present a critical review of fossilized prokaryotic
and eukaryotic microorganisms entrapped in Cretaceous ambers (but not exclusively from this geological period) obtained from
deposits worldwide. Microbiota in ambers are rather diverse and include bacteria, fungi, and protists. We comment on the most
important microbial records from the last 25 years, although it is not an exhaustive bibliographic compilation. The most frequently
reported eukaryotic microfossils are shells of amoebae and protists with a cell wall or a complex cortex. Likewise, diverse
dormant stages (palmeloid forms, resting cysts, spores, etc.) are abundant in ambers. Besides, viral and protist pathogens
have been identified inside insects entrapped in amber. The situation regarding filamentous bacteria and fungi is quite confusing
because in some cases, the same record was identified consecutively as a member of these phylogenetically distant groups.
To avoid these identification errors in the future, we propose to apply a more resolute microscopic and analytical method
in amber studies. Also, we discuss the most recent findings about ancient DNA repair and bacterial survival in remote substrates,
which support the real possibility of ancient DNA amplification and bacterial resuscitation from Cretaceous resins. 相似文献

ABSTRACT: This paper reports on new methods of linking climate change scenarios with hydrologic, agricultural an water planning models to study future water availability for agriculture, an essential element of sustainability. The study is based on the integration of models of water supply and demand, and of crop growth and irrigation management. Consistent modeling assumptions, available databases, and scenario simulations are used to capture a range of possible future conditions. The linked models include WATBAL for water supply; CERES, SOYGRO, and CROPWAT for crop and irrigation modeling; and WEAP for water demand forecasting, planning and evaluation. These models are applied to the U.S. Cornbelt using forecasts of climate change, agricultural production, population and GDP growth. Results suggest that, at least in the near term, the relative abundance of water for agriculture can be maintained under climate change conditions. However, increased water demands from urban growth, increases in reservoir evaporation and increases in crop consumptive use must be accommodated by timely improvements in crop, irrigation and drainage technology, water management, and institutions. These improvements are likely to require substantial resources and expertise. In the highly irrigated basins of the region, irrigation demand greatly exceeds industrial and municipal demands. When improvements in irrigation efficiency are tested, these basins respond by reducing demand and lessening environmental stress with an improvement in system reliability, effects particularly evident under a high technology scenario. Rain-fed lands in the Cornbelt are not forced to invest in irrigation, but there is some concern about increased water-logging during the spring and consequent required increased investment in agricultural drainage. One major water region in the Cornbelt also provides a useful caveat: change will not necessarily be continuous and monotonic. Under one GCM scenario for the 2010s, the region shows a significant decrease in system reliability, while the scenario for the 2020s shows an increase. 相似文献

This paper includes the proposals made in the report “Basis for a National Strategy for Coastal Zone Management” prepared
for the Portuguese Ministry of Environment, Territorial Planning and Regional Development. The final version of that report
was presented in June 2006. This paper describes a theme framework followed by a discussion of concepts. Nine Primary Principles,
eight Principal Objectives, and 37 Strategic Options for an Integrated Coastal Zone Management are then presented. These Strategic
Options are set hierarchically according to their sequential priority, identifying the dominant types of Associated Measures.
The analysis of these arrives at a set of Structural Measures, which interlink and aggregate various actions and propose a
new method of integrated management for the coastal zone, which includes the “Legal Basis of Coastal Zone”; the Organization

The new properties of engineered nanoparticles drive the need for new knowledge on the safety, fate, behavior and biologic effects of these particles on organisms and ecosystems. Titanium dioxide nanoparticles have been used extensively for a wide range of applications, e.g, self-cleaning surface coatings, solar cells, water treatment agents, topical sunscreens. Within this scenario increased environmental exposure can be expected but data on the ecotoxicological evaluation of nanoparticles are still scarce. The main purpose of this work was the evaluation of effects of TiO2 nanoparticles in several organisms, covering different trophic levels, using a battery of aquatic assays. Using fish as a vertebrate model organism tissue histological and ultrastructural observations and the stress enzyme activity were also studied. TiO2 nanoparticles (Aeroxide® P25), two phase composition of anatase (65%) and rutile (35%) with an average particle size value of 27.6±11 nm were used. Results on the EC50 for the tested aquatic organisms showed toxicity for the bacteria, the algae and the crustacean, being the algae the most sensitive tested organism. The aquatic plant Lemna minor showed no effect on growth. The fish Carassius auratus showed no effect on a 21 day survival test, though at a biochemical level the cytosolic Glutathione-S-Transferase total activity, in intestines, showed a general significant decrease (p<0.05) after 14 days of exposure for all tested concentrations. The presence of TiO2 nanoparticles aggregates were observed in the intestine lumen but their internalization by intestine cells could not be confirmed. 相似文献

The bottlenose dolphin (Tursiops truncatus) is the most common cetacean species worldwide and the only marine mammal species resident in the Croatian part of the Adriatic Sea. To gain insight into genetic diversity of bottlenose dolphins at adaptively important loci relevant to conservation, we analysed the polymorphism of major histocompatibility complex (MHC) genes, which play a key role in pathogen confrontation and clearance. Specifically, we examined the diversity of MHC class II DRA, DQA and DQB alleles in 50 bottlenose dolphins from the Adriatic Sea collected between 1997 and 2011 and in 12 animals from other Mediterranean locations. Notable variation in DQA, DQB and three-locus haplotypes was found, with all 10 DQA and 12 DQB alleles encoding unique protein products. Analysis of the ratio of non-synonymous to synonymous substitution rates suggests that positive selection acts at both highly variable loci. Phylogenetic analyses revealed trans-species polymorphism at the DQB locus, strongly indicating the influence of balancing selection in the long term. In fact, the balancing selection observed in bottlenose dolphins is higher than that reported for most other cetaceans and comparable to that seen in terrestrial mammals. 相似文献

Trace metal concentrations were determined in particulate matter (PM10) in ambient air of four purposively selected residential areas in Ibadan, Nigeria namely Bodija market (BM), Ojo Park (OP), Oluyole Estate (OE) and University of Ibadan (UI). PM10 was determined in the morning (7–10 a.m.) and afternoon (2–5 p.m.) for 12 weeks in the dry season months of January–March using a volumetric sampler following standard procedures and levels compared with WHO guideline limits. Glass-fibre filter papers exposed to the particulate matter were digested using appropriate acid mixtures, and the digest analysed for trace metals including Ni, Cr, Mn, Zn, and Pb using ICPMS method and levels compared with WHO limits. Data was analysed using ANOVA and Pearson correlation test at 5 % level of significance. The highest mean PM10 concentrations 502.3 ± 39.9 μg/m3 were recorded in the afternoon period at BM, while the lowest concentration 220.6 ± 69.9 μg/m3 was observed in the morning hours at UI. There was a significant difference between the PM10 levels across the various locations (p < 0.05), and all the levels were higher than WHO limit of 50 μg/m3. The highest levels of Ni, Zn and Pb were recorded at BM, which also had the highest PM10 burden. The trend in Pb levels across the locations was BM > UI > OP > OE with the highest level 5.70 μg/m3 in BM nearly fourfolds WHO limits of 1.5 μg/m3. There was a significant correlation between PM10 and Ni (p < 0.05).Urban communities with increased human activities especially motor traffic recorded both higher levels of PM10 and toxic trace metals. There is need to carry out source apportionment to establish the origin of these trace metals in future studies. 相似文献
