Nickel-resistant bacteria isolated from underneath Ni-hyperaccumulators growing on serpentine soils were screened for production
of polyhydroxyalkanoates. These rhizobacteria accumulated poly-3-hydroxybutyric acid [P(3HB)] accounting 3.9–67.7% of cell
dry weight during growth in gluconate and/or glucose. Cupriavidus pauculus KPS 201 utilized only gluconate and accumulated about 67.7% P(3HB) while, Bacillus firmus AND 408 utilized both carbon sources for polymer synthesis. The isolates being resistant to Ni also accumulated substantial
amount of P(3HB) when grown in presence of the heavy metal and this was revealed by transmission electron microscopic studies.
Although B. firmus AND 408 produced only P(3HB) at higher concentrations of gluconate, C. pauculus KPS 201 synthesized copolymer of 3-hydroxybutyric acid (3HB) and 3-hydroxyvaleric acid (3HV) [P(3HB-co-3HV)]. In presence
of 0.8% gluconate and 4 mM Ni, KPS 201 cells produced PHA amounting 81% CDW, which contained 76 and 24 mol% 3HB and 3HV monomers,

The present study describes the assimilation of di-n-octyl phthalate by an aerobic bacterium, isolated from municipal waste-contaminated soil sample utilizing di-n-octyl phthalate as the sole source of carbon and energy. The isolate was identified as Gordonia sp. based on the morphological, nutritional and biochemical characteristics as well as 16S rRNA gene sequence analysis. A combination of chromatographic and spectrometric analyses revealed a complete di-n-octyl assimilation pathway. In the degradation process, mono-n-octyl phthalate, phthalic acid, protocatechuic acid and 1-octanol were identified as the degradation products of di-n-octyl phthalate. Furthermore, phthalic acid was metabolized via protocatechuic acid involving protocatechuate 3,4-dioxygenase while 1-octanol was metabolized by NAD+-dependent dehydrogenases to 1-octanoic acid, which was subsequently degraded via β-oxidation, ultimately, leading to tricarboxylic acid cycle intermediates. Apart from phthalic acid and 1-octanol metabolizing pathway enzymes, two esterases, di-n-octyl phthalate hydrolase and mono-n-octyl phthalate hydrolase involved in di-n-octyl phthalate degradation were found to be inducible in nature. This is the first report on the metabolic pathway involved in the complete degradation of di-n-octyl phthalate by a single bacterial isolate, which is also capable of efficiently degrading other phthalate esters of environmental concern having either shorter or longer alkyl chains. 相似文献

Mating behaviour and mating patterns are affected to a large extent by body size in both hermaphrodites and gonochorists.
Detailed research on mating patterns, mate choice, pre- and post-copulatory sexual selection in marine pulmonates is wanting,
thus warranting more attention as a study system in the future. The simultaneous hermaphroditic limpet Siphonaria capensis (Pulmonata: Basommatophora) shows size-dependent fecundity, and acts as a suitable organism to test the effects of body size
on mating patterns, mating success, gender expression and reciprocity in hermaphroditic marine gastropods. We mainly used
bootstrap resampling techniques to estimate the effects of different factors on mating patterns. In the populations studied,
a strong size-assortative mating pattern was observed, where small-scale spatial distribution of potential mating partners
(the mate availability hypothesis) could explain 65% of this pattern, while mate choice and mating constraints explaining
the rest. No significant difference in mating success between limpets with different body size was found, even though in one
population, the sperm recipients were larger than the non-copulating limpets. Interestingly, we found that intromission was
non-reciprocal during copulations. This may mean that this species does not conform to the common rule of reciprocity predicted
for hermaphrodites, unless there is sex-role alternation between individuals in a mating pair. The mating partners consisted
of similar sized, acting males and females, thus without any indication of the body size determining the gender expression.
The matings took place in early mornings only at spring tides and the animals were observed laying egg masses only during
neap tides. Since siphonariid limpets possess both a spermatheca and a seminal vesicle, which may have either a sperm storing
and/or digestive function, post-copulatory sexual selection (e.g., sperm competition) cannot be dismissed. 相似文献

Fission track technique has been used for uranium estimation in drinking water samples collected from some areas of Amritsar District, Punjab, India. The uranium concentration in water samples is found to vary from 3.19 to 45.59 microg l(-1). Some of the physico-chemical properties such as pH, conductance and hardness and the content of calcium, magnesium, total dissolved solids (TDS), sodium, potassium, chloride, nitrate and heavy metals viz. zinc, cadmium, lead and copper have been determined in water samples. An attempt has been made to correlate uranium concentration with these water quality parameters. A positive correlation of conductance, nitrate, chloride, sodium, potassium, magnesium, TDS, calcium and hardness with uranium concentration has been observed. This study aimed to evaluate musculoskeletal disorders (MSDs) and postural stress among female craftworkers. The study was carried out on 75 adult female craftworkers in different districts of West Bengal. The prevalence of MSDs, body part discomfort (BPD) rating and body joint angles of the workers were evaluated with standard methods. Electromyography (EMG) of the shoulder and back muscles was recorded with the BIOPAC system. The prevalence of MSDs, BPD rating and deviation of joint angle were comparatively lower in the case of sitting on the floor with folded legs than squatting and sitting on the floor with stretched legs postures. The EMG and rms values of the shoulder and back muscles were comparatively lower in this posture. Therefore, it was concluded that sitting on the floor with folded legs was less hazardous and it imposed less postural stress in comparison to other sitting postures. 相似文献

Carbon content in soils changes depending on the land use system, type of management practice and time. There is an increasing concern about the soil quality vis-à-vis organic carbon content in soils due to global warming and enhanced CO2 concentration in the atmosphere. This has led to estimate carbon stock in soils at global and regional levels. The objective of the present study was to evaluate RothC model to estimate total organic carbon (TOC) changes under four long term fertilizer experimental sites representing sub-humid moist (Sarol and Nabibagh), sub-humid dry (Panjri) and semi-arid (Teligi) climate in India. The plant carbon input rate was calibrated using organic carbon and other soil parameters using RothC. The results showed that RothC could simulate changes in TOC in two contrasting eco-sites for surface soil layers. The root mean square error (RMSE) considered as modelling error ranged from 11.50 to 15.01, 4.70 to 11.60, 2.14 to 6.52 and 1.45 to 13.74 in the surface layers of Sarol, Nabibagh, Panjri, and Teligi sites, respectively. The simulation biases expressed by M (relative error) by Student‘t’ value for all the treatments at these sites were non-significant with two exceptions. Observed trends in TOC consist of an increase for all the four treatments in the sub-humid site of Sarol and Nabibagh; while manures alone or in combination increase TOC appreciably in Teligi and Panjri. TOC remained, however, almost similar over years for the control (no fertilizer or manure) and NPK treatments in all the four sites. Analysis of RothC output data showed that this model could be used as a tool to arrive at different threshold values of rainfall to influence decomposition rate modifier and thus to find out rate of organic carbon sequestration in various bioclimatic systems. 相似文献

Adverse effects of diesel oil on microanatomical structure of the alimentary canal of O. nilotica were studied using SEM observations. The study revealed irregular arrangement of the stratified epithelial cells along with fragmentation of the normal concentric pattem of microridges of the same cells in buccopharynx and oesophagus. The excessive secretion of mucus of buccopharynx and oesophagus were the salient changes caused by diesel oil pollution. The destruction and degeneration of the mucosal folds of stomach and intestine along with their epithelial cells exhibited a concrete hyperactivity resulting in abundant' secretion of mucus over the microridges of the epithelial cells. In the intestinal region the columnar epithelial cells showed tumefaction and microvilli of the plasma membrane of epithelial cells get heavily damaged. Disarray of the microridges of epithelial cells, excessive secretions of mucus formation of even cell sheet were the most conspicuous changes in rectum. It was concluded that chronic exposure of diesel oil may hamper the absorption of the nutrients through alimentary canal resulting into ill-growth and production of the fish. 相似文献

Glyphosate is used widely to control weeds. Glyphosate is a broad spectrum, non-selective, systemic and post-emergent herbicide. Glyphosate excessive use and impact on the environment is promoting the analysis of glyphosate in water, soil and food materials. Methods to analyse glyphosate at low levels are needed because glyphosate has a short half-life due to easy microbial degradation. Glyphosate has a high polarity and solubility in water, has high binding affinity with soil and is non-volatile. The absence of chromophoric groups in the molecular structure makes the detection difficult. Therefore, detection can be achieved by derivatisation, which makes glyphosate more volatile and stable for spectroscopic analysis. Derivatisation is commonly done by alkyl chloroformates, acylating agents, 9-fluoroenylmethylchloroformate, 4-methoxybenzenesulfonylfluoride and o-phthalaldehyde. Immunosensors allow detection at microlevels. Nanocrystals and nanotechnology allow detection at nanolevels. Here, we review methods to derivatise and analyse glyphosate. 相似文献

Subgrid-scale (SGS) modeling is a long-standing problem and a critical component in the large-eddy simulation (LES) of atmospheric boundary layer. A variety of SGS models with different levels of sophistication have been proposed for different needs, such as Smagorinsky's (1963) eddy viscosity model, Mason and Thomson's (1992) stochastic backscattering model, and Sullivan et al.'s (1994) near surface model. A modified Smagorinsky SGS model has been used in the LES version of Terminal Area Simulation System (TASS-LES). It has successfully simulated the buoyancy-dominated, convective atmospheric boundary layer flows, while simulations of the shear-dominated, slightly unstable, neutral, and stably stratified boundary layer flows are not so good. For the later, we used a simpler version of Sullivan et al.'s subgrid-scale model in which turbulent kinetic energy equation is not included and the model is still the first-order closure. A momentum profile matching approach is adopted in the proposed model. A series of simulations for shear-dominated, slightly unstable and neutral boundary layers are performed using different subgrid-scale models and different grid resolutions. The results are compared with those of Sullivan et al. (1994) and with empirical similarity relations for the surface layer. The simulations with the new SGS model appear to be far more satisfactory than those with the modified Smagorinsky model. 相似文献
