A novel approach to predict aquatic toxicity from molecular structure   总被引:1,自引:0,他引:1  
The main aim of the study was to develop quantitative structure-activity relationship (QSAR) models for the prediction of aquatic toxicity using atom-based non-stochastic and stochastic linear indices. The used dataset consist of 392 benzene derivatives, separated into training and test sets, for which toxicity data to the ciliate Tetrahymena pyriformis were available. Using multiple linear regression, two statistically significant QSAR models were obtained with non-stochastic (R2=0.791 and s=0.344) and stochastic (R2=0.799 and s=0.343) linear indices. A leave-one-out (LOO) cross-validation procedure was carried out achieving values of q2=0.781 (scv=0.348) and q2=0.786 (scv=0.350), respectively. In addition, a validation through an external test set was performed, which yields significant values of Rpred2 of 0.762 and 0.797. A brief study of the influence of the statistical outliers in QSAR's model development was also carried out. Finally, our method was compared with other approaches implemented in the Dragon software achieving better results. The non-stochastic and stochastic linear indices appear to provide an interesting alternative to costly and time-consuming experiments for determining toxicity.  相似文献

This study presents an analysis of the ability of a two-parameter response surface, a multiple linear regression and a neural network model to produce global quantitative structure-activity relationships (QSARs) to predict the toxic potency of phenols to Tetrahymena pyriformis. The phenolic toxicity data set analysed is characterised by multiple mechanisms of toxic action. The study aimed to evaluate the confidence that can be applied to the modelling of the differing mechanisms of action. Assessment of confidence was decided in terms of whether the statistics for the global models reflect the ability of the QSARs to model the individual mechanisms of toxic action present in the data set. The results showed that the global statistics only reflected the ability of models to predict the two non-covalent mechanisms (polar narcosis and respiratory uncoupling), with the metabolically transformed and electrophilic mechanism (pre-electrophiles and soft electrophiles) being modelled poorly by all three model building methods. The results confirm the difficulty in modelling electrophilic mechanisms of toxic action. The results also highlight the fact that this poor predictivity is often 'hidden' in good statistical fit of some global models. In particular these results emphasise that for practical predictive purposes the mechanistic applicability domain is required to give confidence to estimated toxicity values.  相似文献

Composite sampling techniques for identifying the largest individual sample value seem to be cost effective when the composite samples are internally homogeneous. However, since it is not always possible to form homogeneous composite samples, these methods can lead to higher costs than expected. In April-May 1998, mass coral bleaching was observed in the lagoon of Rangiroa Atoll, Tuamotu Archipelago, French Polynesia. Six months later, the extent of bleaching-induced coral mortality was assessed at three sites. Corals in the fast-growing genus Pocillopora had experienced >99% mortality. Many large colonies of the slow-growing genus Porites (mean horizontal cross-sectional area 5.8 m2) had also died - a phenomenon not previously observed in French Polynesia and virtually unprecedented world-wide. At one site, 25% of colonies, or 44% of the pre-bleaching cover of living Porites, experienced whole-colony mortality. At the two other sites, recently dead Porites accounted for 41% and 82% of the pre-bleaching live cover. Mortality in Porites was negatively correlated with depth between 1.5 and 5 m. Using a 50-year dataset of mean monthly sea surface temperature (SST), derived from ship- and satellite-borne instruments, we show that bleaching occurred during a period of exceptionally high summer SST. 1998 was the first year in which mean monthly SSTs exceeded the 1961-1990 upper 95% confidence limit (29.4°C) for a period of three consecutive months. We suggest that the sustained 3-month anomaly in local summer SST was a major cause of coral mortality, but do not discount the synergistic effect of solar radiation. Recovery of the size-frequency distribution of Porites colonies to pre-bleaching levels may take at least 100 years.  相似文献

Acetylene reduction rates were measured in lagoonal sediments, cyanobacterial mats and limestone surfaces between 1991 and 1995 at many sites, depths and seasons; all the studied substrata contained cyanobacteria. The acetylene reduction/15N2 fixation ratio was measured for the different communities and varied between 1.8 and 4.8, depending on substratum. Fixation rates were 1.7 to 7 times higher during daylight compared to night-time rates. N2 fixation rates ranged from 0.4 to 3.9 mg N m-2 day-1 for the lagoonal sediment/mat communities, and the rate was about 2 mg N m-2 day-1 for the lagoonal limestone substrata. Total lagoonal benthic N2 fixation contributed 24.4% of the total nitrogen requirement for the benthic primary production of benthic communities of the lagoon. The input of N2 fixation by the microbial planktonic communities (including cyanobacteria) of the lagoon, which are highly productive, is unquantified but is likely to be large.  相似文献

Lignoid chemical defenses in the freshwater macrophyte Saururus cernuus   总被引:3,自引:0,他引:3  
Summary. Chemical defense against herbivores has rarely been investigated for freshwater plants, possibly due to the common misconception that herbivory on aquatic macrophytes is low and would not select for chemical defenses. In previous work, the freshwater angiosperm Saururus cernuus was shown to be a low preference food for omnivorous crayfish despite its high nutrient value and relatively soft texture. We used feeding by the crayfish Procambarus clarkii to guide fractionation of the deterrent lipid-soluble extract of this plant, leading to the identification of seven deterrent lignoid metabolites, (–)-licarin A, (+)-saucernetin, (–)-dihydroguaiaretic acid, (–)-sauriols A and B, (–)-saucerneol, and (–)-saucerneol methyl ether. Lignans have been implicated in terrestrial plant chemical defenses as insect growth inhibitors, insect toxins, nematocides, antibacterial, and antifungal agents. However, these activities have rarely been demonstrated using ecologically relevant methodologies in terrestrial systems, and never before in freshwater systems. The widespread nature of lignans amongst very distantly related plants, along with their rich diversity of molecular structure, suggests that they could play a large role in mediating plant-herbivore interactions. In addition to the lignoid compounds we identified, there were other compounds present in low concentration or unstable compounds that were deterrent, that did not appear to be lignans, but that we were unable to identify. A stow net (90 m2 opening) was used, at regular intervals between July 1989 and June 1993, to collect fish over ca. 4 h on both the flood and ebb tides at five sites distributed at regularly spaced distances throughout the Elbe Estuary. Overall, 53.4% of the species were marine, and some of these were abundant, e.g.Clupea harengus, Pleuronectes flesus, Pomatoschistus minutus, Merlangius merlangus and Sprattus sprattus. However, in terms of number of individuals, such species contributed only 9.1%, compared with 90.0% by anadromous species, which was predominantly due to exceptionally high catches of Osmerus eperlanus. The ichthyofaunal compositions on the flood and ebb tides were similar, even downstream where salinities change markedly during each tidal cycle. Although this is probably due in part to groups of fish being swept first upstream on the flood tide and then back downstream on the ebb tide, several species clearly tended to occupy particular regions of the estuary. Each year the species composition changed sequentially from the most downstream site (max. salinity=ca. 31.4‰) to the most upstream site (max. salinity=ca. 1.5‰), mainly as a result of a sequential decline in the number and abundance of marine species and a progressive increase in the contributions of both anadromous and freshwater species. However, the marine species diagnostic of differences in the compositions among regions varied between years. Thus, for example, the diagnostic species for the downstream sites were C. harengus and M. merlangus in 1990/1991 and S. sprattus in 1992/1993, reflecting differences in the recruitment strengths of these marine species in the two years. The ichthyofaunal composition exhibited pronounced annual cyclical changes in each region and thus occurred irrespective of whether or not the salinity in a region underwent pronounced changes during the year. This cyclicity was attributable to intra-annual variations in the times of recruitment of certain marine species, particularly in the downstream region, and to seasonal migrations of diadromous species into each region, as well as to the movements of freshwater species into the upstream region in winter and early spring. Three isopod species (Crustacea: Isopoda), commonly found in the intertidal and supratidal zones of the North American Pacific coast, were studied with respect to symbiotic microbiota in their midgut glands (hepatopancreas). Ligia pallasii (Oniscidea: Ligiidae) contained high numbers of microbial symbionts in its hepatopancreatic caeca. Numbers of endosymbionts were strongly reduced by ingestion of antibiotics. By contrast, the hepatopancreas of Idotea wosnesenskii (Valvifera: Idoteidae) and Gnorimosphaeroma oregonense (Sphaeromatidea: Sphaeromatidae) did not contain any microbiota. Results of feeding experiments suggest that microbial endosymbionts contribute to digestive processes in L. pallasii, the most terrestrial of the three isopods that we studied. The acquisition of digestion-enhancing endosymbionts may have been an important evolutionary step allowing isopods to colonize terrestrial habitats where relatively indigestible leaf litter is the primary food source. By contrast, the ability to digest phenolic compounds was most developed in one of the more marine species, suggesting that this trait may have evolved independently in isopod species that consume a phenolic-rich diet, whether in marine habitats or on land. We document the distribution and abundance of seagrasses, as well as the intra-annual temporal patterns in the abundance of seagrasses and the productivity of the nearshore dominant seagrass (Thalassia testudinum) in the south Florida region. At least one species of seagrass was present at 80.8% of 874 randomly chosen mapping sites, delimiting 12,800 km2 of seagrass beds in the 17,000-km2 survey area. Halophila decipiens had the greatest range in the study area; it was found to occur over 7,500 km2. The range of T. testudinum was almost as extensive (6,400 km2), followed by Syringodium filiforme (4,400 km2), Halodule wrightii (3,000 km2) and Halophila engelmanni (50 km2 ). The seasonal maxima of standing crop was about 32% higher than the yearly mean. The productivity of T. testudinum was both temporally and spatially variable. Yearly mean areal productivity averaged 0.70 g m−2day−1, with a range of 0.05–3.29 g m−2 day−1. Specific productivity ranged between 3.2 and 34.2 mg g−1 day−1, with a mean of 18.3 mg g−1 day−1. Annual peaks in specific productivity occurred in August, and minima in February. Integrating the standing crop for the study area gives an estimate of 1.4 × 1011 g T. testudinum and 3.6 × 1010 g S. filiforme, which translate to a yearly production of 9.4 × 1011 g T. testudinum leaves and 2.4 × 1011 g S. filiforme leaves. We assessed the efficacy of rapid visual surveys for estimating abundance of seagrasses in south Florida by comparing these results to measures of leaf biomass for T. testudinum and S. filiforme. Our rapid visual surveys proved useful for quantifying seagrass abundance, and the data presented in this paper serve as a benchmark against which future change in the system can be quantified. This study continues a comparative cytogenetic analysis of the fish family Mugilidae, reporting the karyotype characterization of the leaping mullet, Liza saliens, by C-banding, Ag- and fluorochrome-staining, and completing the fluorescence in situ hybridization (FISH)-mapping of the 18S and 5S rRNA genes (rDNA) to the chromosomes of the six Mediterranean mullets, namely L. saliens, L. ramada, L. aurata, Mugil cephalus, Chelon labrosus and Oedalechilus labeo. In all species, except M. cephalus, the 5S rDNA sites were localized on a medium-sized acrocentric chromosome pair, which was considered homeologous in all of them. In L. saliens, an additional 5S rDNA site was detected in a location close to the one shown by major ribosomal genes in M. cephalus, i.e. the subtelomeric region of chromosome pair 1. The 5S rDNA site in M. cephalus is located on the smallest chromosome pair of the complement, which, on the other hand, though on a different position, bears 18S rDNA in all the species of Liza and Chelon examined. The heterochromatin composition and the major and minor ribosomal gene locations suggest that the karyotype of L. saliens (subgenus Protomugil) can be considered intermediate between the karyotype of the more primitive M. cephalus and those of the other Liza (subgenus Liza) species and of the representatives of the more derived genera Chelon and Oedalechilus.  相似文献
