The present study evaluated the effect of culture conditions and phytopathogenic strain co-culture on the production of antimicrobial metabolites and antifungal activity of Penicillium chrysogenum R1, which PCR identified. Antimicrobial activity was determined using the Hunter-Hunter experimental design with three factors (pH, incubation temperature, and inoculum, at two levels each). The antifungal metabolites, β 1-3 glucanase and chitinase, produced in the presence of live and inactivated Fusarium oxysporum Fsox C11 biomass, were evaluated using HPLC-MS and GC-MS. Results showed that P. chrysogenum inhibited the growth of five phytopathogenic fungal strains, and the most significant inhibition was observed for F. oxysporum Fsox C11. The best conditions to achieve the highest antifungal activity of the cell-free extract were pH 7, 28°C, 1 × 106 spores/mL, and 144 h of fermentation, observing 86% inhibition of F. oxysporum Fsox C11 growth. Production of antifungal metabolites such as 1,4-benzoquinone imine, viridicatic acid, phenol-5-methyl-2-(1-methyl ethyl), and hydrolytic enzymes β 1-3 glucanase and chitinase was detected. The results define the perspective in designing new processes and products for biocontrol phytopathogens.  相似文献

The Scanning Imaging Absorption Spectrometer for Atmospheric Chartography (SCIAMACHY) onboard the European Envisat spacecraft performs continuous spectral observations of reflected, scattered and transmitted sunlight in various observation geometries. A unique feature of SCIAMACHY is the capability of probing the atmosphere in three different observation geometries:The nadir, limb, and occultation measurement modes. In nadir mode, column densities of trace gases are retrieved with a spatial resolution of typically 30× 60 km using the Differential Optical Absorption Spectroscopy (DOAS) technique (Platt and Perner, 1983). Alternating with the nadir measurement, vertical profiles of absorber concentration in the stratosphere are derived in limb and occultation. In this paper we present an overview over some applications of SCIAMACHY data in space-based monitoring of atmospheric pollution. The DOAS algorithms for the retrieval of total column amounts from nadir spectra are briefly described and case studies of pollution events are presented. We also illustrate the technique used to derive stratospheric concentration profiles from limb observations and show comparisons with other remote sensing systems. Special emphasis will be given to techniques, which take advantage of SCIAMACHY's different viewing geometries. In particular, we will discuss the potential and limits of strategies to infer tropospheric abundances of O3and NO2.  相似文献

Titanium dioxide is the most important photocatalysts used for purifying applications. If a TiO2- containing material is left outdoors as a form of flat panels, it is activated by sunlight to remove harmful NOx gases during the day. The photocatalytic efficiency of a TiO2-treated mortar for removal of NOx was investigated in the frame of this work. For this purpose a fully equipped monitoring system was designed at a pilot site. This system allows the in situ evaluation of the de-polluting properties of a photocatalytic material by taking into account the climatologic phenomena in street canyons, accurate measurements of pollution level and full registration of meteorological data The pilot site involved three artificial canyon streets, a pollution source, continuous NOx measurements inside the canyons and the source as well as background and meteorological measurements. Significant differences on the NOx concentration level were observed between the TiO2 treated and the reference canyon. NOx values in TiO2 canyon were 36.7 to 82.0% lower than the ones observed in the reference one. Data arising from this study could be used to assess the impact of the photocatalytic material on the purification of the urban environment.  相似文献

Man-made and natural radionuclides in Lake Wallersee were determined in the pre-Alpine environment at the northern slope of the Alps, which was heavily affected by the Chernobyl fallout in May 1986. The objective of this study was to get knowledge of location and quantity of man-made radionuclide input (especially (137)Cs) generated in the Chernobyl accident to lake sediments. Eleven sediment cores were sampled and activity depth profiles of (137)Cs and (210)Pb were determined with 5mm depth-resolution. The Chernobyl fallout produced an extreme (137)Cs peak in the sediment cores providing an excellent time marker. The chronological interpretation of deeper sediment layers was done by radiochemical analysis of (90)Sr and (239+240)Pu, which were released during atmospheric weapons' tests in the 1950s and 1960s. This allowed a complete chronological analysis of the sediment cores with a very compact set of data.  相似文献

Modeling concentration-response function became extremely popular in ecotoxicology during the last decade. Indeed, modeling allows determining the total response pattern of a given substance. However, reliable modeling is consuming in term of data, which is in contradiction with the current trend in ecotoxicology, which aims to reduce, for cost and ethical reasons, the number of data produced during an experiment. It is therefore crucial to determine experimental design in a cost-effective manner. In this paper, we propose to use the theory of locally D-optimal designs to determine the set of concentrations to be tested so that the parameters of the concentration-response function can be estimated with high precision. We illustrated this approach by determining the locally D-optimal designs to estimate the toxicity of the herbicide dinoseb on daphnids and algae. The results show that the number of concentrations to be tested is often equal to the number of parameters and often related to the their meaning, i.e. they are located close to the parameters. Furthermore, the results show that the locally D-optimal design often has the minimal number of support points and is not much sensitive to small changes in nominal values of the parameters. In order to reduce the experimental cost and the use of test organisms, especially in case of long-term studies, reliable nominal values may therefore be fixed based on prior knowledge and literature research instead of on preliminary experiments.  相似文献
