Most of the research on the epigenetic phenomenon of DNA methylation has been performed with vertebrates and plants. Knowledge on DNA methylation in Daphnia magna, a key test organism in aquatic toxicology, is completely lacking. Through epigenetic inheritance, effects of transient chemical exposure could be transferred to non-exposed generations, which could have a major impact on ecological risk assessment procedures. In this study, we determined if CpG methylation occurs in D. magna and if this can be influenced by exposure to toxic substances. Homologs of human DNA methyltransferases DNMT1, DNMT2 and DNMT3A were found in the partially available D. magna genome. Using an optimized “Amplification of Intermethylated Sites (AIMS)” technique, two methylated fragments were discovered in D. magna DNA. No homology was found for these sequences. The methylation and the D. magna origin of the fragments were confirmed with Southern analysis. This optimized AIMS technique was then applied to DNA of D. magna which were exposed to 180 µg/L Cd for two generations. Exposure resulted in a significant decrease in reproduction. The same methylated fragments with the same band intensity were observed in DNA of both non-exposed and exposed daphnids. As such, it could not be demonstrated that Cd exposure altered DNA methylation. However, the presence of DNA methylation in D. magna shows that potentially epigenetic effects may occur in this species.  相似文献

A sensitivity study was performed to investigate the responses of potential natural vegetation distribution in China to the separate and combined effects of temperature, precipitation and [CO2], using the process-based equilibrium terrestrial biosphere model BIOME4. The model shows a generally good agreement with a map of the potential natural vegetation distribution based on a numerical comparison using the ΔV statistic (ΔV = 0.25). Mean temperature of each month was increased uniformly by 0–5 K, in 0.5- or 1-K intervals. Mean precipitation of each month was increased and decreased uniformly by 0–30%, in 10% intervals. The analyses were run at fixed CO2 concentrations of 360 and 720 ppm. Temperature increases shifted most forest boundaries northward and westward, expanded the distribution of xeric biomes, and confined the tundra to progressively higher elevations. Precipitation increases led to a greater area occupied by mesic biomes at the expense of xeric biomes. Most vegetation types in the temperate regions, and on the Tibetan Plateau, expanded westward into the dry continental interior with increasing precipitation. Precipitation decreases had opposite effects. The modelled effect of CO2 doubling was to partially compensate for the negative effect of drought on the mesic biomes and to increase potential ecosystem carbon storage by about 40%. Warming tended to counteract this effect, by reducing soil carbon storage. Forest biomes showed substantial resilience to climate change, especially when the effects of increasing [CO2] were taken into account. Savannas, dry woodland and tundra biomes proved sensitive to temperature increases. The transition region of grassland and forest, and the Tibetan plateau, was the most vulnerable region.  相似文献

The brominated flame retardant decabromodiphenylether (DBDE) was analysed in wild birds to identify the most suitable species for monitoring time trends in DBDE contamination. This information was later used for the design of a 10-year trend study on DBDE in the European Union. DBDE was measured in muscle tissue, liver, and eggs from 10 terrestrial and four aquatic bird species. DBDE was detected in 47% of the terrestrial bird samples (nine species) and in 9% of the aquatic bird samples (six species). Peregrine falcon and sparrowhawk specimens were selected as most suitable species to determine temporal trends of DBDE. For sparrowhawks, no significant change in DBDE concentrations between 1973 and 2001 was found, although in later years more DBDE concentrations were above the detection limit. Peak DBDE levels measured in peregrines in 1995, were followed by a decline in concentrations until 2001. The same species were used for a trend study on hexabromocylcododecane (HBCD). Twenty-four percent of peregrine falcon eggs and 12% of sparrowhawk muscle samples demonstrated measurable HBCD residues. Three diastereomers of HBCD were analysed and the α-diastereomer was the predominant one in most samples. No clear time trends were observed for HBCD in either species. This study demonstrated that these DBDE and HBCD are bioavailable to birds of Northern Europe, although bioaccumulation seems to occur to a limited extent.  相似文献

Halogenated persistent organic pollutants [polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs)] along with total lipid, were measured in the liver and muscle of three species of deep water fish (black scabbard, black dogfish (liver only) and roundnose grenadier) collected from the Rockall fishing area, to the west of Scotland, between 2006 and 2008. Both contaminant groups were detected in the muscle and liver, with concentrations of PCBs being higher than PBDEs. There were no significant differences in the PCB or PBDE concentrations between the three species, or different sampling locations in the Rockall fishing area. PCB concentrations (ΣICES (International Council for the Exploration of the Sea)7 PCBs) greater than 500 μg kg−1 lipid weight were found in 26 of the 106 liver samples. PCB concentrations were compared to OSPAR assessment criteria, concentrations were above background but below Environmental Assessment Criteria. Estimated Toxic Equivalent (TEQ) concentrations, calculated using published models, in the fish muscle and liver indicated that consumption of deep water fish is unlikely to represent a risk to human health. The high squalene content in some of the black dogfish liver necessitated an additional clean-up step, involving gel permeation chromatography, when analyzing for PBDEs. Concentrations of PBDEs were low with many congeners being below detection limits, particularly in the muscle. There are currently no assessment criteria available for PBDEs. Furthermore, there is only very limited data on PBDEs in deep water fish. However, the concentrations observed in this study were similar to the concentrations recently reported in Mediterranean deep water fish.  相似文献

The high degree of heterogeneity within sediments can make interpreting one-dimensional measurements difficult. The recent development and use of in situ techniques that measure two-dimensional distributions of porewater solutes have facilitated investigation of the role of spatial heterogeneity in sediment biogeochemistry. A colourimetric diffusive equilibration in thin films method has been developed that allows two-dimensional, high-resolution measurement of reactive phosphate in sediment porewaters. A method detection limit of 0.22 μM, an effective upper limit of ∼1000 μM and relative standard deviations typically below 5% were achieved. This method was evaluated by deployment in seagrass (Zostera capricorni) colonised sediments, as part of combined probes with similar colourimetric methods for sulfide and iron(II). The two-dimensional, high resolution distributions obtained provide a highly representative measurement of the co-distributions of porewater solutes, allowing heterogeneous features and biogeochemical processes to be observed and interpreted. Microniches of high phosphate concentration >100 μM were observed throughout the distributions and were interpreted to be due to localised zones of rapid organic matter mineralisation, possibly using electron acceptors other than iron(III) oxyhydroxides (e.g. aerobic respiration) as often they did not correspond with microniches of higher Fe(II) concentration.  相似文献

The (1)H NMR spectrum and the crystal structure of HCDBCO [(1R,2R,5R,6R,9S,10S)-5,6-dibromo-1,10,11,12,13,13-hexachlorotricyclo[8,2,1,0(2,9)]-tridec-11-ene)] are reported. The measured dihedral angles from the X-ray structure correlate very well with those calculated from the proton-proton coupling constants indicating that the conformations in solution and in the solid state are probably very similar. Attempts at calculating the 3D model structure of HCDBCO only produced a very poor match between the measured dihedral angles between vicinal protons and the observed proton-proton coupling constants from the (1)H NMR spectrum. GC/MS analysis with an injector temperature of 250 degrees C produced minor amounts of debrominated HCDBCO. Reducing the temperature to 200 degrees C eliminated this problem.  相似文献
